Abstract

This disclosure is directed to a device and a system for picking a target analyte of a suspension. A picker introduces at least one force, such as by a magnetic gradient and/or by a pressure gradient, to extract the target analyte from a specimen. The magnetic gradient may be introduced by a magnet, such as a permanent magnet or an electromagnet, and the pressure gradient may be introduced by a pump which moves within a fluid-primed cannula to create the pressure gradient, thereby drawing the target analyte into the cannula. The picker may also expel the target analyte onto or into a substrate, such as a well plate, after the target analyte has been drawn into the picker by reversing the pressure gradient or removing the magnetic gradient.

Claims (65)

1. A system comprising:

a picker comprising

a pump block comprising a first end and a second end comprising a seal,

a piston at least partially housed within the pump block and partially extending through the seal, wherein the piston translates relative to the pump block, and

a cannula comprising an adapter end and a tube end, wherein the adapter end is connected to the second end of thepump block and the tube end extends away from the pump block;

a driver;

a coupling mating the driver to the piston; and

a mount connecting the picker to an imaging or detection device.

2. The system of claim 1 , further comprising a light source connected to the picker.

3. The system of claim 1 , wherein a full rotation of the piston includes 1-10,000 steps.

4. The system of claim 3 , wherein each step includes 1-10,000 micro-steps.

5. The system of claim 4 , wherein each step or micro-step equates to a volume equal to or less than 1 picoliter, 10 picoliters, 100 picoliters, 1 nanoliter, 1 microliter, or 1 milliliter.

6. The system of claim 1 , further comprising a housing to encase at least a portion of the driver, the coupling, a first end of the piston, and at least a portion of the pump block.

7. The system of claim 6 , wherein at least one side of the pump block is biased against at least one side of the housing to inhibit rotational motion between the pump block and the housing to reduce or eliminate backlash.

8. The system of claim 1 , further comprising at least one weight on top of the driver or a housing to the base to bias and preload threads of the piston to reduce or eliminate backlash.

9. The system of claim 1 , wherein the coupling provides zero backlash and is axially and torsionally stiff.

10. The system of claim 9 , wherein the coupling is a non-expanding bellows or a split-beam drive assembly.

11. A method comprising:

providing a system comprising

an imaging or detection device,

a picker comprising

a pump block comprising a first end and a second end comprising a seal,

a piston at least partially housed within the pump block and partially extending through the seal, wherein the piston translates relative to the pump block, and

a cannula comprising an adapter end and a tube end, wherein the adapter end is connected to the second end of the pump block and the tube end extends away from the pump block,

a driver,

a coupling mating the driver to the piston, and

a mount connecting the picker to the imaging or detection system;

placing the picker above a target material located on or within a substrate which is located on or within the imaging or detection device; and

exerting a force on the target material by the picker to draw the target material into the cannula of the picker.

The method of claim 11 , wherein the target material is a cell, an ova, a circulating tumor cell, a nucleated red blood cell, a fetal cell, a circulating endothelial cell, a vesicle, a liposome, a protein, a nucleic acid, a biological molecule, a naturally occurring microscopic unit having an enclosed membrane, an artificially prepared microscopic unit having an enclosed membrane, a parasite, a microorganism, or an inflammatory cell.

13. The method of claim 11 , wherein the force is a pressure gradient provided by the picker.

14. The method of claim 11 , further comprising the step of attaching a magnetic particle to the target material,

wherein the attaching step is perfoimed before the placing step, and

wherein the force is magnetic or electromagnetic.

15. A method comprising:

providing a system comprising

an imaging or detection device,

a picker comprising

a pump block comprising a first end and a second end comprising a seal

a piston at least partially housed within the pump block and partially extending through the seal, wherein the piston translates relative to the pump block, and

a cannula comprising an adapter end and a tube end, wherein the adapter end is connected to the pump block and the tube end extends away from the pump block,

a driver,

a coupling mating the driver to the piston, and

a mount connecting the picker to the imaging or detection system;

placing the picker above a target material located on or within a substrate which is located on or within the imaging or detection device;

touching the cannula to the substrate;

moving the picker horizontally or orthogonally to detach the target material from the substrate; and

exerting a force on the target material by the picker to draw the target material into the cannula of the picker,

wherein the force is a pressure gradient provided by the picker.

16. A method comprising:

providing a system comprising

an imaging or detection device,

a picker comprising

a pump block comprising a first end and a second end comprising a seal,

a piston at least partially housed within the pump block and partially extending through the seal, wherein the piston translates relative to the pump block, and

a cannula comprising an adapter end and a tube end, wherein the adapter end is connected to the pump block and the tube end extends away from the pump block,

a driver,

a coupling mating the driver to the piston, and

a mount connecting the picker to the imaging or detection system;

placing the picker above a target material located on or within a substrate which is located on or within the imaging or detection device:

touching the cannula to the substrate;

oscillating the picker up and down to detach the target analyte from the substrate; and

exerting a force on the target material by the picker to draw the target material into the cannula of the picker,

wherein the force is a pressure gradient provided by the picker.

17. The method of claim 16 , wherein the oscillation occurs at a frequency less than or equal to 10 kHz.
