Abstract

The present invention is directed to antibodies and fragments thereof having binding specificity for NGF. Another embodiment of this invention relates to the antibodies described herein, and binding fragments thereof, comprising the sequences of the V H , V L and CDR polypeptides described herein, and the polynucleotides encoding them. The invention also contemplates conjugates of anti-NGF antibodies and binding fragments thereof conjugated to one or more functional or detectable moieties. The invention also contemplates methods of making said anti-NGF antibodies and binding fragments thereof. Embodiments of the invention also pertain to the use of anti-NGF antibodies, and binding fragments thereof, for the diagnosis, assessment and treatment of diseases and disorders associated with NGF.

Claims (16)

1. A rabbit or humanized anti-human NGF antibody or human NGF binding antibody fragment thereof which comprises a variable light chain containing the CDR polypeptide sequences of SEQ ID NO: 65; SEQ ID NO: 66; and SEQ ID NO: 67 and a variable heavy chain containing the CDR polypeptides of SEQ ID NO: 68; SEQ ID NO: 69; and SEQ ID NO: 70.

2. A composition comprising an anti-human NGF antibody or NGF binding antibody fragment thereof according to claim 1 and a pharmaceutically acceptable carrier.

3. A diagnostic composition comprising an anti-human NGF antibody or NGF binding antibody fragment thereof according to claim 1 and a diagnostically acceptable carrier.

4. The antibody of claim 1 , wherein said antibody fragment is selected from an Fab fragment, an Fab′ fragment, scFv, or an F(ab′) 2 fragment.

5. The antibody of claim 1 , wherein:

said antibody or fragment comprises aV H polypeptide comprising the sequence of SEQ ID NO.:63 and aV L polypeptide comprising the sequence of SEQ ID NO.:61.

6. The antibody of claim 1 , wherein said antibody or fragment comprises a human IgG1, IgG2, IgG3, or IgG4 Fc region.

7. The antibody of claim 1 , wherein said antibody or fragment is aglyclosylated and/or comprises an Fc region that has been modified to alter effector function, half-life, proteolysis, and/or glycosylation.

8. The antibody of claim 1 , wherein:

said antibody or fragment comprises a heavy chain polypeptide comprising the sequence of SEQ ID NO.:64 and a light chain polypeptide comprising the sequence of SEQ ID NO.:62.

9. A composition comprising an anti-human NGF antibody or NGF binding antibody fragment thereof according to claim 5 and a pharmaceutically acceptable carrier.

10. A diagnostic composition comprising an anti-human NGF antibody or NGF binding antibody fragment thereof according to claim 5 and a diagnostically acceptable carrier.

11. The antibody of claim 5 , wherein said antibody fragment is selected from an Fab fragment, an Fab′ fragment, scFv, or an F(ab′) 2 fragment.

12. A composition comprising an anti-human NGF antibody according to claim 8 and a pharmaceutically acceptable carrier.

13. A diagnostic composition comprising an anti-human NGF antibody according to claim 8 and a diagnostically acceptable carrier.

14. An antibody according to claim 1 which is humanized.
