Abstract

Transdermal therapeutic system for the transdermal administration of a systemically active agent comprising a self-adhesive layer structure, comprising A) a backing layer, and B) a dried biphasic layer, the dried biphasic layer having a) an outer phase having a composition comprising 75% to 100% of a polymer or polymer mixture, and b) an inner phase having a composition comprising at least one active agent, wherein the inner phase forms dispersed deposits in the outer phase, and c) 0.1% to 3.5% of an interface mediator with a kinematic viscosity of from 10 cSt to 100 000 cSt at 25° C., and C) optionally an additional skin contact layer.

Claims (25)

1. A transdermal therapeutic system for transdermal administration of rotigotine containing 2.0 mg/cm 2 to 4.0 mg/cm 2 rotigotine base in a self-adhesive layer structure, comprising:

A) a backing layer; and

B) a dried biphasic layer having:

a) an outer phase having a composition comprising 75% to 100% of one or more pressure-sensitive adhesive polysiloxane(s) having tack to adhere to skin for up to 7 days;

b) an inner phase comprising rotigotine base present in the dried biphasic layer in an amount ranging from 16 to 26%, said rotigotine base in a solid solution with polyvinylpyrrolidone, wherein the inner phase forms dispersed deposits in the form of droplets in the outer phase; and

c) 0.1% to 3.0% of a silicone oil with a kinematic viscosity of from 100 cSt to 100 000 cSt at 25° C.; and

C) optionally an additional skin contact layer,

and said dispersed deposits formed with 1% silicone oil have a maximum droplet diameter that is approximately 29 to approximately 54% smaller than that of dispersed deposit droplets within a comparable transdermal therapeutic system that does not contain silicone oil.

The transdermal therapeutic system according to claim 1 ; wherein the rotigotine base contained in the self-adhesive layer structure, is present in an amount of 2.0 mg/cm 2 to 3.0 mg/cm 2 ; and wherein the transdermal therapeutic system comprises a therapeutically effective amount of rotigotine base for an administration period on the skin of a human patient of 7 days.

A method comprising: utilizing the transdermal therapeutic system according to claim 2 to treat a patient suffering from Parkinson's disease, Parkinson's plus syndrome, depression, anxiety, ARDS, fibromyalgia, the restless-legs syndrome, or to treat or prevent dopaminergic neuron loss, a cognitive disorder, dementia, or lewy body disease, by providing therapeutically effective amounts of rotigotine base for 7 days by said transdermal therapeutic system during an administration period on the skin of a human patient of 7 days.

4. A transdermal therapeutic system in accordance with claim 1 , wherein the interface mediator has a kinematic viscosity of from 10 000 cSt to 100 000 cSt at 25° C.

5. A transdermal therapeutic system in accordance with claim 1 , wherein said dried biphasic layer does not comprise a tackifier or plasticizer.

6. The transdermal therapeutic system in accordance with claim 1 , wherein a maximum droplet size of the dispersed deposits of 5 μm to 25 μm is provided.

7. A transdermal therapeutic system for transdermal administration of rotigotine containing 2.0 mg/cm 2 to 4.0 mg/cm 2 rotigotine base in a self-adhesive layer structure, comprising:

A) a backing layer; and

B) a dried biphasic layer having:

a) an outer phase having a composition comprising 75% to 100% of one or more pressure-sensitive adhesive polysiloxane(s) having tack to adhere to skin for up to 7 days;

b) an inner phase comprising rotigotine base in a solid solution with a hydrophilic agent, wherein the inner phase forms dispersed deposits in the form of droplets in the outer phase; and

c) 0.1% to 3.5% of a silicone oil with a kinematic viscosity of from 100 cSt to 12 500 cSt at 25° C.; and

C) an additional skin contact layer,

wherein the dispersed deposits have a maximum droplet size of 5 μm to 25 μm;

said interface mediator fills a cavity between the inner and outer phase;

said dried biphasic layer does not comprise a tackifier or plasticizer,

the silicone oil in an amount of 1% results in dispersed deposits whose maximum droplet diameter is approximately 29 to approximately 54% smaller than that of dispersed deposit droplets within a comparable transdermal therapeutic system that does not contain silicone oil; and

the rotigotine is present in the dried biphasic layer in an amount ranging from 16 to 26%.
