Abstract

The invention features methods, kits, and compositions for mitochondrial replacement in the treatment of disorders arising from mitochondrial dysfunction. The invention also features methods of diagnosing neuropsychiatric (e.g., bipolar disorder) and neurodegenerative disorders based on mitochondrial structural abnormalities.

Claims (20)

1. A method for enhancing cardiac or cardiovascular function in a human subject in need thereof, said method comprising administering to said subject a pharmaceutical composition comprising isolated and substantially pure mitochondria in an amount sufficient to enhance said cardiac or cardiovascular function, wherein said mitochondria are syngeneic mitochondria or allogeneic mitochondria.

2. The method of claim 1 , wherein said mitochondria are syngeneic mitochondria.

3. The method of claim 1 , wherein said mitochondria are allogeneic mitochondria.

4. The method of claim 1 , wherein said mitochondria are administered systemically.

5. A method for enhancing cardiac or cardiovascular function in a human subject in need thereof, said method comprising administering to said subject a pharmaceutical composition comprising isolated and substantially pure mitochondria in an amount sufficient to enhance said cardiac or cardiovascular function, wherein said mitochondria are administered locally into cardiac muscle.

6. The method of claim 1 , wherein the isolated and substantially pure mitochondria are administered in an amount sufficient to increase the respiratory ability of a heart cell in the subject.

7. The method of claim 4 , wherein the isolated and substantially pure mitochondria are administered in an amount sufficient to increase the respiratory ability of a heart cell in the subject.

8. The method of claim 5 , wherein the isolated and substantially pure mitochondria are administered in an amount sufficient to increase the respiratory ability of a heart cell in the subject.

The method of claim 1 , further comprising administering to said subject an agent selected from vitamin A, vitamin C, vitamin D, vitamin E, vitamin K, folic acid, choline, vitamin B1, vitamin B2, vitamin B5, vitamin B6, vitamin B12, biotin, nicotinamide, betacarotene, coenzyme Q, selenium, superoxide dismutase, glutathione peroxide, uridine, creatine succinate, pyruvate, dihydroxyacetone, acetyl-L-carnitine, alpha-lipoic acid, cardiolipin, omega fatty acid, lithium carbonate, lithium citrate, calcium, and mixtures thereof.

10. The method of claim 1 , wherein the subject has a heart that is ischemic.

11. The method of claim 4 , wherein the subject has a heart that is ischemic.

12. The method of claim 5 , wherein the subject has a heart that is ischemic.

13. The method of claim 1 , wherein the mitochondria are isolated from marrow-derived adult progenitor cells, stem cells, precursor cells, or skeletal muscle cells.

14. The method of claim 4 , wherein the mitochondria are isolated from marrow-derived adult progenitor cells, stem cells, precursor cells, or skeletal muscle cells.

15. The method of claim 5 , wherein the mitochondria are isolated from marrow-derived adult progenitor cells, stem cells, precursor cells, or skeletal muscle cells.

A method for enhancing cardiac or cardiovascular function in a human subject in need thereof, said method comprising administering to said subject a pharmaceutical composition comprising isolated and substantially pure mitochondria in an amount sufficient to enhance said cardiac or cardiovascular function, wherein the human subject has an ischemic heart tissue, and wherein said method comprises locally administering to said heart tissue a pharmaceutical composition comprising isolated and substantially pure syngeneic mitochondria in an amount sufficient to enhance cardiac or cardiovascular function of said subject.

17. The method of claim 16 , wherein the isolated and substantially pure syngeneic mitochondria are administered in an amount sufficient to increase the respiratory ability of the heart cell or tissue.

18. The method of claim 16 , wherein the mitochondria are isolated from skeletal muscle.

19. The method of claim 16 , further comprising administering to said subject an agent selected from vitamin A, vitamin C, vitamin D, vitamin E, vitamin K, folic acid, choline, vitamin B1, vitamin B2, vitamin B5, vitamin B6, vitamin B12, biotin, nicotinamide, betacarotene, coenzyme Q, selenium, superoxide dismutase, glutathione peroxide, uridine, creatine succinate, pyruvate, dihydroxyacetone, acetyl-L-carnitine, alpha-lipoic acid, cardiolipin, omega fatty acid, lithium carbonate, lithium citrate, calcium, and mixtures thereof.

20. The method of claim 16 , wherein the pharmaceutical composition is administered by local injection into cardiac muscle.
