Methods of preventing inflammation and treating pain using anti-NGF compositions
This invention pertains to monovalent agents, including Fab fragments and monovalent monoclonal antibodies analogous to MetMab, having binding specificity to human Nerve Growth Factor (“NGF”), and methods of treating pain in an individual wherein there is no substantial increase in the inflammatory response of the individual following administration of the monovalent agents.
1. A rabbit anti-human nerve growth factor (NGF) antibody or antigen binding fragment which comprises the light chain CDR polypeptide sequences of SEQ ID NO: 95; SEQ ID NO: 96; and SEQ ID NO: 97 and the heavy chain CDR polypeptide sequences of SEQ ID NO: 98; SEQ ID NO: 99; and SEQ ID NO: 100.
2. A humanized antibody or antibody fragment which specifically binds to human nerve growth factor comprising the light chain CDR polypeptide sequences of SEQ ID NO: 95; SEQ ID NO: 96; and SEQ ID NO: 97 and the heavy chain CDR polypeptide sequences of SEQ ID NO: 98; SEQ ID NO: 99; and SEQ ID NO: 100.
3. The antibody or antibody fragment of claim 2 , which comprises the variable light chain sequence of SEQ ID NO: 91 and the variable heavy chain sequence of SEQ ID NO: 93.
4. The antibody or antibody fragment of claim 2 , which comprises the light chain sequence of SEQ ID NO: 92 and the heavy chain sequence of SEQ ID NO: 94.
5. The antibody or antibody fragment of claim 2 , which comprises human constant regions.
6. The antibody or antibody fragment of claim 2 , which comprises human IgG1, IgG2, IgG3 or IgG4 constant regions.
7. The antibody or antibody fragment of claim 2 , which comprises human IgG1 constant regions.
8. The antibody or antibody fragment of claim 2 , wherein said antibody fragment is selected from Fab, Fab′, Fv, and scFv.
