Abstract

A method for nucleic acid isolation comprising: receiving a binding moiety solution within a process chamber; mixing the binding moiety solution with a biological sample, within the process chamber, in order to produce a moiety-sample mixture; incubating the moiety-sample mixture during a time window, thereby producing a solution comprising a set of moiety-bound nucleic acid particles and a waste volume; separating the set of moiety-bound nucleic acid particles from the waste volume; washing the set of moiety-bound nucleic acid particles; and releasing a nucleic acid sample from the set of moiety-bound nucleic acid particles. The method preferably utilizes a binding moiety comprising at least one of poly(allylamine) and polypropylenimine tetramine dendrimer, both of which reversibly bind and unbind to nucleic acids based upon environmental pH.

Claims (15)

1. A method for nucleic acid isolation, comprising:

receiving a biological sample;

receiving affinity moiety-coated microparticles comprising magnetic microparticles bonded to a moiety comprising at least one of Poly(allylamine) and Polypropylenimine tetramine dendrimer;

incubating the affinity moiety-coated microparticles with the biological sample for reversibly binding the moiety to nucleic acid material of the biological sample, thereby producing moiety-bound nucleic acid particles within a moiety-sample mixture;

occluding a fluidic pathway at a first set of occlusion positions to define a first truncated pathway upon actuating a cam card relative to a first subset of a set of pins corresponding to the first set of occlusion positions, wherein occluding the fluidic pathway at the first set of occlusion positions defines a fluidic path through a magnetic field and a heating region, and wherein separating the moiety-bound nucleic acid particles comprises delivering the moiety-bound nucleic acid particles into the first truncated pathway and capturing the moiety-bound nucleic acid particles within the magnetic field;

separating the moiety-bound nucleic acid particles from the moiety-sample mixture; and

releasing the nucleic acid material from the moiety of the moiety-bound nucleic acid particles.

The method of claim 1 , wherein the biological sample comprises at least one of cerebrospinal fluid and mucous membrane, wherein the nucleic acid material comprises target Enterovirus RNA, wherein incubating comprises reversibly binding the moiety to the target Enterovirus RNA in the biological sample, and wherein releasing the nucleic acid material comprises releasing the target Enterovirus RNA from the moiety.

3. The method of claim 2 , wherein the moiety comprises the Poly(allylamine), and wherein releasing the target Enterovirus RNA comprises releasing the target Enterovirus RNA from the Poly(allylamine) at an extraction efficiency of at least 90%.

4. The method of claim 2 , further comprising spiking a nucleic acid sample comprising the nucleic acid material with a process control, the process control comprising a set of primers and probes selected based on the target Enterovirus RNA.

The method of claim 1 , wherein the biological sample comprises at least one of blood and urine, wherein the nucleic acid material comprises target Streptococcus DNA, wherein incubating comprises reversibly binding the moiety to the target Streptococcus DNA in the biological sample, and wherein releasing the nucleic acid material comprises releasing the target Streptococcus DNA from the moiety.

The method of claim 5 , wherein the biological sample comprises the blood, wherein the Poly(allylamine) comprises Poly(allylamine) of molecular weight (MW)<40,000 Da, wherein the moiety comprises the Poly(allylamine) of MW<40,000 Da, and wherein incubating comprises reversibly binding the Poly(allylamine) of MW<40,000 Da to the target Streptococcus DNA in the blood for at least two minutes at a binding temperature between room temperature and 45° C., and wherein releasing the nucleic acid material comprises releasing the target Streptococcus DNA from the Poly(allylamine) of MW<40,000 Da.

The method of claim 5 , wherein the Polypropylenimine tetramine dendrimer comprises Polypropylenimine tetramine dendrimer Generation 1, wherein the moiety comprises the Polypropylenimine tetramine dendrimer Generation 1, wherein incubating comprises reversibly binding the Polypropylenimine tetramine dendrimer Generation 1 to the target Streptococcus DNA in the biological sample at a binding temperature between room temperature and 60° C., and wherein releasing the nucleic acid material comprises releasing the target Streptococcus DNA from the Polypropylenimine tetramine dendrimer Generation 1 at an extraction efficiency of at least 90%.

The method of claim 1 , wherein the moiety comprises the Poly(allylamine) and the Polypropylenimine tetramine dendrimer, wherein the nucleic acid material comprises target DNA and target RNA, wherein incubating comprises reversibly binding the Poly(allylamine) and the Polypropylenimine tetramine dendrimer to the target DNA and the target RNA, and wherein releasing the nucleic acid material comprises releasing the target DNA and the target RNA from the Poly(allylamine) and the Polypropylenimine tetramine dendrimer.

9. The method of claim 1 , further comprising delivering a waste volume of the sample mixture to a waste chamber coupled to the fluidic pathway.
