HUMAN SKIN EQUIVALENTS EXPRESSING EXOGENOUS POLYPEPTIDES
The present invention relates generally to compositions for wound closure. More specifically, the present invention provides human skin equivalents engineered to express exogenous polypeptides (e.g., antimicrobial polypeptides and keratinocyte growth factor 2) and compositions and methods for making human skin equivalents engineered to express exogenous polypeptides. In addition, the present invention provides methods for treatment of wounds with human skin equivalents engineered to express exogenous polypeptides.
1 . An organotypic human skin equivalent comprising stratified squamous epithelia produced from Near-diploid Immortalized Keratinocytes (NIKS) cells stably transfected with a vector comprising a nucleotide sequence encoding cathelicidin polypeptide operably linked to an exogenous promoter, wherein said skin equivalent exhibits antimicrobial activity.
2 . The organotypic human skin equivalent of claim 1 , wherein said human skin equivalent further comprises keratinocytes from a patient.
3 . The organotypic human skin equivalent of claim 1 , further comprising a dermal equivalent.
4 . The organotypic human skin equivalent of claim 1 , further comprising Near-diploid Immortalized Keratinocytes (NIKS) cells that do not express exogenous cathelicidin.
5 . The organotypic human skin equivalent of claim 1 , further comprising keratinocytes expressing at least one additional antimicrobial polypeptide.
6 . The organotypic human skin equivalent of claim 1 , wherein said exogenous promoter is a constitutive promoter.
