Abstract

Oral dosage forms of osteoclast inhibitors, such as nitrogen-containing bisphosphonates, such as zoledronic acid in an acid or a salt form, or in a molecular complex can be used to treat or alleviate pain or related conditions, such as treating pain associated with Paget's disease of bone.

Claims (32)

1. A method of treating pain associated with Paget's disease of bone, comprising orally administering zoledronic acid in a disodium salt form to a human being in need thereof, wherein the zoledronic acid in the disodium salt form is orally administered in a dosage form that does not contain a bioavailability enhancer, wherein the human being receives a total dose of the zoledronic acid in the disodium salt form in a month that is about 100 mg to about 500 mg; and wherein the zoledronic acid in the disodium salt form enhances bioavailability of zoledronic acid as determined by the following test:

comparing the area under the curve of the plasma concentration (AUC) of zoledronic acid resulting from orally administering the dosage form to a beagle dog to the area under the curve of the plasma concentration of zoledronic acid resulting from orally administering a reference dosage form to a beagle dog,

wherein the reference dosage form contains same molar amount of zoledronic acid in the diacid form instead of zoledronic acid in the disodium salt form.

2. The method of claim 1 , wherein each orally administered dose contains about 50 mg to about 60 mg of the zoledronic acid in the disodium salt form.

3. The method of claim 1 , wherein the zoledronic acid in the disodium salt form is orally administered at an interval of about 5 to 10 days.

4. The method of claim 2 , wherein the zoledronic acid in the disodium salt form is orally administered about once a week.

5. The method of claim 1 , wherein each orally administered dose of the zoledronic acid in the disodium salt form is orally administered in a manner that provides an AUC of zoledronic acid of about 140 ng·h/mL to about 150 ng·h/mL, measured over a 24 hour period.

6. The method of claim 4 , wherein each orally administered dose of the zoledronic acid in the disodium salt form is orally administered in a manner that provides an AUC of zoledronic acid of about 130 ng·h/mL to about 150 ng·h/mL, measured over a 24 hour period.

7. The method of claim 1 , wherein the human being experiences significant pain relief for a period of at least about 24 hours.

8. The method of claim 1 , wherein the human being experiences significant pain relief for a period of at least about 1 week.

9. The method of claim 1 , wherein each orally administered dose contains about 55 mg to about 60 mg of the zoledronic acid in the disodium salt form.

10. The method of claim 1 , wherein the human being experiences significant pain relief for a period of at least 1 month.

11. The method of claim 1 , wherein the zoledronic acid in the disodium salt form is orally administered about once a week for 6 weeks.

12. The method of claim 1 , wherein the zoledronic acid in the disodium salt form is orally administered about once a week for 3 weeks.

13. The method of claim 1 , wherein the human being experiences significant pain relief at 3 months after the first dose of the zoledronic acid in the disodium salt form is administered.

14. The method of claim 1 , wherein the human being experiences significant pain relief at 6 months after the first dose of the zoledronic acid in the disodium salt form is administered.

15. The method of claim 13 , wherein the zoledronic acid in the disodium salt form is orally administered in a dosage form that contains at least about 20% of the zoledronic acid in the disodium salt form by weight.

16. The method of claim 1 , wherein the human being experiences significant pain relief that lasts for a period of at least 1 year.

17. The method of claim 13 , wherein the zoledronic acid in the disodium salt form is orally administered in a dosage form that contains at least about 50% of the zoledronic acid in the disodium salt form by weight.

18. The method of claim 1 , wherein the human being experiences significant pain relief that lasts for a period of at least 5 years.

19. The method of claim 15 , wherein the zoledronic acid in the disodium salt form is orally administered in a manner that results in a bioavailability of zoledronic acid in the human being of about 1.4% to about 1.8%.

20. The method of claim 15 , wherein the zoledronic acid in the disodium salt form is orally administered in a manner that results in a bioavailability of zoledronic acid in the human being of about 1.8% to about 2%.

21. The method of claim 15 , wherein the zoledronic acid in the disodium salt form is orally administered in a manner that results in a bioavailability of zoledronic acid in the human being of about 2% to about 2.2%.

22. The method of claim 15 , wherein the zoledronic acid in the disodium salt form is orally administered in a manner that results in a bioavailability of zoledronic acid in the human being of about 2.2% to about 2.4%.

23. The method of claim 15 , wherein the zoledronic acid in the disodium salt form is orally administered in a manner that results in a bioavailability of zoledronic acid in the human being of about 2.4% to about 2.8%.

24. The method of claim 15 , wherein the zoledronic acid in the disodium salt form is orally administered in a manner that results in a bioavailability of zoledronic acid in the human being of about 2.8% to about 3.2%.

25. The method of claim 5 , wherein the zoledronic acid in the disodium salt form is administered at least 2 times in 3 months.

26. The method of claim 1 , wherein administering the zoledronic acid in the disodium salt form to the human being reduces the use of a nonsteroidal anti-inflammatory drug or an opioid by at least about 25% as compared to the use of the nonsteroidal anti-inflammatory drug or the opioid without administration of the zoledronic acid in the disodium salt form.

27. The method of claim 9 , wherein the zoledronic acid in the disodium salt form is orally administered once a week for 6 weeks.

28. The method of claim 1 , wherein the zoledronic acid in the disodium salt form is co-administered with a steroid.

29. The method of claim 1 , wherein the zoledronic acid in the disodium salt form is orally administered in a manner that results in a reduction of serum alkaline phosphatase (ALP) level of at least about 50% from baseline.

30. The method of claim 1 , wherein the Paget's disease of bone does not return within 18 months from the time of the last oral administration of zoledroniac acid.
