Abstract

This invention relates methods of using a non-fucosylated anti-CD40 antibody for treatment of cancer and chronic infectious diseases.

Claims (180)

1. A method of treating cancer, the method comprising administering a composition comprising an anti-CD40 antibody to a patient in need thereof, wherein the anti-CD40 antibody comprises a heavy chain variable region comprising amino acids 1-113 of SEQ ID NO:1 and a light chain variable region comprising amino acids 1-113 of SEQ ID NO:2, and a human constant region;

wherein the constant region has an N-glycoside-linked sugar chain at residue N297 according to the EU index; and wherein less than 5% of N-glycoside-linked sugar chains in the composition comprise a fucose residue; and

wherein the anti-CD40 antibody is administered at a dose level between 2-50 μg/kg (μg per kilogram of patient body weight).

2. The method of claim 1 , wherein the patient has a CD40 positive cancer.

3. The method of claim 1 , wherein the patient has a CD40 negative cancer.

4. A method of treating cancer, the method comprising administering an anti-PD-1 antibody and a composition comprising an anti-CD40 antibody to a patient in need thereof, wherein the anti-PD-1 antibody is nivolumab or pembrolizumab,

wherein the anti-CD40 antibody comprises a heavy chain variable region comprising amino acids 1-113 of SEQ ID NO:1 and a light chain variable region comprising amino acids 1-113 of SEQ ID NO:2, and a human constant region;

wherein the constant region has an N-glycoside-linked sugar chain at residue N297 according to the EU index; wherein less than 5% of N-glycoside-linked sugar chains in the composition comprise a fucose residue; and wherein the anti-CD40 antibody is administered at a dose level between 2-50 μg/kg (μg per kilogram of patient body weight).

5. The method of claim 1 , wherein the cancer is a hematologic cancer.

6. The method of claim 1 , wherein the cancer is a solid tumor.

7. A method of treating cancer, the method comprising administering a composition comprising an anti-CD40 antibody to a patient in need thereof,

wherein the anti-CD40 antibody comprises a heavy chain variable region comprising amino acids 1-113 of SEQ ID NO:1 and a light chain variable region comprising amino acids 1-113 comprising amino acids 1-113 of SEQ ID NO:2, and a human constant region;

wherein the constant region has an N-glycoside-linked sugar chain at residue N297 according to the EU index; and wherein less than 5% of N-glycoside-linked sugar chains in the composition comprise a fucose residue; and

wherein the anti-CD40 antibody is administered at a dose level of 30 μg/kg (μg per kilogram of patient body weight).

8. The method of claim 7 , wherein the anti-CD40 antibody is administered at least once every three weeks.

9. The method of claim 8 , wherein the anti-CD40 antibody is administered on day one of a three week cycle.

10. The method of claim 8 , wherein the anti-CD40 antibody is administered on days one and eight of a three week cycle.

11. The method of claim 7 , wherein the patient has a CD40 positive cancer.

12. The method of claim 7 , wherein the patient has a CD40 negative cancer.

13. The method of claim 1 , wherein the cancer is selected from the group consisting of melanoma, breast cancer, metastatic breast cancer, lung cancer, non-small cell lung cancer, bladder cancer, pancreatic cancer, B-cell lymphoma, Hodgkin lymphoma, diffuse large cell B-cell lymphoma, follicular lymphoma, chronic lymphocytic leukemia, small cell lung cancer, ovarian cancer, kidney cancer, cervical cancer, head and neck cancer, prostate cancer, glioblastoma, non-Hodgkin lymphoma, hepatocellular carcinoma, and multiple myeloma.

14. The method of claim 13 , wherein the cancer is pancreatic cancer.

15. The method of claim 4 , wherein the cancer is selected from the group consisting of melanoma, breast cancer, metastatic breast cancer, lung cancer, non-small cell lung cancer, bladder cancer, pancreatic cancer, B-cell lymphoma, Hodgkin lymphoma, diffuse large cell B-cell lymphoma, follicular lymphoma, chronic lymphocytic leukemia, small cell lung cancer, ovarian cancer, kidney cancer, cervical cancer, head and neck cancer, prostate cancer, glioblastoma, non-Hodgkin lymphoma, hepatocellular carcinoma, and multiple myeloma.

16. The method of claim 4 , wherein the anti-PD-1 antibody is pembrolizumab.

17. The method of claim 4 , wherein the cancer is pancreatic cancer, and the anti-PD-1 antibody is pembrolizumab.

The method of claim 7 , wherein the cancer is selected from the group consisting of melanoma, breast cancer, metastatic breast cancer, lung cancer, non-small cell lung cancer, bladder cancer, pancreatic cancer, B-cell lymphoma, Hodgkin lymphoma, diffuse large cell B-cell lymphoma, follicular lymphoma, chronic lymphocytic leukemia, small cell lung cancer, ovarian cancer, kidney cancer, cervical cancer, head and neck cancer, prostate cancer, glioblastoma, non-Hodgkin lymphoma, hepatocellular carcinoma, and multiple myeloma.

19. The method of claim 18 , wherein the cancer is pancreatic cancer.

20. The method of claim 7 , further comprising administering an anti-PD-1 antibody to the patient, wherein the anti-PD-1 antibody is nivolumab or pembrolizumab.

21. The method of claim 20 , wherein the cancer is pancreatic cancer.

22. The method of claim 20 , wherein the cancer is pancreatic cancer, and the anti-PD-1 antibody is pembrolizumab.

23. The method of claim 1 , wherein the anti-CD40 antibody is administered at a dose level of 10 μg/kg or 30 μg/kg.

24. The method of claim 1 , further comprising administering one or more chemotherapeutic agents to the patient.

25. The method of claim 4 , further comprising administering one or more chemotherapeutic agents to the patient.

26. The method of claim 7 , further comprising administering one or more chemotherapeutic agents to the patient.

27. The method of claim 4 , wherein the cancer is a hematologic cancer.

28. The method of claim 4 , wherein the cancer is a solid tumor.

29. The method of claim 7 , wherein the cancer is a hematologic cancer.

30. The method of claim 7 , wherein the cancer is a solid tumor.

31. The method of claim 22 , further comprising administering one or more chemotherapeutic agents to the patient.

32. The method of claim 31 , wherein the one or more chemotherapeutic agents comprise gemcitabine, paclitaxel, or both.

33. A method of treating cancer, the method comprising administering a composition comprising an anti-CD40 antibody to a patient in need thereof,

wherein the anti-CD40 antibody comprises a heavy chain variable region comprising amino acids 1-113 of SEQ ID NO:1 and a light chain variable region comprising amino acids 1-113 of SEQ ID NO:2, and a human constant region;

wherein the constant region has an N-glycoside-linked sugar chain at residue N297 according to the EU index; and wherein less than 5% of N-glycoside-linked sugar chains in the composition comprise a fucose residue; and

wherein the anti-CD40 antibody is administered at a dose level of 10 μg/kg (μg per kilogram of patient body weight).

34. The method of claim 33 , wherein the anti-CD40 is administered at least once every three weeks.

35. The method of claim 34 , wherein the anti-CD40 antibody is administered on day one of a three week cycle.

36. The method of claim 33 , wherein the patient has a CD40 positive cancer.

37. The method of claim 33 , wherein the patient has a CD40 negative cancer.

38. The method of claim 33 , wherein the cancer is selected from the group consisting of melanoma, breast cancer, metastatic breast cancer, lung cancer, non-small cell lung cancer, bladder cancer, pancreatic cancer, B-cell lymphoma, Hodgkin lymphoma, diffuse large cell B-cell lymphoma, follicular lymphoma, chronic lymphocytic leukemia, small cell lung cancer, ovarian cancer, kidney cancer, cervical cancer, head and neck cancer, prostate cancer, glioblastoma, non-Hodgkin lymphoma, hepatocellular carcinoma, and multiple myeloma.

39. The method of claim 38 , wherein the cancer is pancreatic cancer.

40. The method of claim 33 , further comprising administering an anti-PD-1 antibody to the patient, wherein the anti-PD-1 antibody is nivolumab or pembrolizumab.

41. The method of claim 40 , wherein the anti-PD-1 antibody is pembrolizumab.

42. The method of claim 33 , further comprising administering one or more chemotherapeutic agents to the patient.

43. The method of claim 33 , wherein the cancer is a hematologic cancer.

44. The method of claim 33 , wherein the cancer is a solid tumor.

45. The method of claim 1 , wherein the anti-CD40 antibody is administered at least once every three weeks.

46. The method of claim 13 , wherein the cancer is melanoma.

47. The method of claim 13 , wherein the cancer is non-small cell lung cancer.

48. The method of claim 13 , wherein the cancer is head and neck cancer.

49. The method of claim 24 , wherein the cancer is selected from the group consisting of melanoma, breast cancer, metastatic breast cancer, lung cancer, non-small cell lung cancer, bladder cancer, pancreatic cancer, B-cell lymphoma, Hodgkin lymphoma, diffuse large cell B-cell lymphoma, follicular lymphoma, chronic lymphocytic leukemia, small cell lung cancer, ovarian cancer, kidney cancer, cervical cancer, head and neck cancer, prostate cancer, glioblastoma, non-Hodgkin lymphoma, hepatocellular carcinoma, and multiple myeloma.

50. The method of claim 49 , wherein the cancer is pancreatic cancer.

51. The method of claim 49 , wherein the cancer is melanoma.

52. The method of claim 49 , wherein the cancer is non-small cell lung cancer.

53. The method of claim 49 , wherein the cancer is head and neck cancer.

54. The method of claim 45 , wherein the anti-CD40 antibody is administered on day one of a three week cycle.

55. The method of claim 54 , further comprising administering one or more chemotherapeutic agents to the patient.

56. The method of claim 55 , wherein the one or more chemotherapeutic agents comprise gemcitabine, paclitaxel, or both.

57. The method of claim 1 , wherein the anti-CD40 antibody is administered intravenously or subcutaneously to the patient.

58. The method of claim 4 , wherein the patient has a CD40 positive cancer.

59. The method of claim 4 , wherein the patient has a CD40 negative cancer.

60. The method of claim 4 , wherein the dose level is 10 μg/kg or 30 μg/kg.

61. The method of claim 15 , wherein the anti-CD40 antibody is administered intravenously or subcutaneously to the patient.

62. The method of claim 61 , wherein the cancer is melanoma.

63. The method of claim 61 , wherein the cancer is non-small cell lung cancer.

64. The method of claim 61 , wherein the cancer is head and neck cancer.

65. The method of claim 16 , wherein the cancer is selected from the group consisting of melanoma, breast cancer, metastatic breast cancer, lung cancer, non-small cell lung cancer, bladder cancer, pancreatic cancer, B-cell lymphoma, Hodgkin lymphoma, diffuse large cell B-cell lymphoma, follicular lymphoma, chronic lymphocytic leukemia, small cell lung cancer, ovarian cancer, kidney cancer, cervical cancer, head and neck cancer, prostate cancer, glioblastoma, non-Hodgkin lymphoma, hepatocellular carcinoma, and multiple myeloma.

71. The method of claim 70 , wherein the cancer is pancreatic cancer.

75. The method of claim 70 , wherein the anti-PD-1 antibody is pembrolizumab.

The method of claim 75 , wherein the cancer is selected from the group consisting of melanoma, breast cancer, metastatic breast cancer, lung cancer, non-small cell lung cancer, bladder cancer, pancreatic cancer, B-cell lymphoma, Hodgkin lymphoma, diffuse large cell B-cell lymphoma, follicular lymphoma, chronic lymphocytic leukemia, small cell lung cancer, ovarian cancer, kidney cancer, cervical cancer, head and neck cancer, prostate cancer, glioblastoma, non-Hodgkin lymphoma, hepatocellular carcinoma, and multiple myeloma.

77. The method of claim 76 , wherein the cancer is pancreatic cancer.

81. The method of claim 25 , wherein the anti-PD-1 antibody is pembrolizumab.

82. The method of claim 81 , wherein the anti-CD40 antibody is administered intravenously or subcutaneously to the patient.

The method of claim 82 , wherein the cancer is selected from the group consisting of melanoma, breast cancer, metastatic breast cancer, lung cancer, non-small cell lung cancer, bladder cancer, pancreatic cancer, B-cell lymphoma, Hodgkin lymphoma, diffuse large cell B-cell lymphoma, follicular lymphoma, chronic lymphocytic leukemia, small cell lung cancer, ovarian cancer, kidney cancer, cervical cancer, head and neck cancer, prostate cancer, glioblastoma, non-Hodgkin lymphoma, hepatocellular carcinoma, and multiple myeloma.

127. The method of claim 42 , wherein the anti-CD40 antibody is administered intravenously or subcutaneously to the patient.

128. The method of claim 42 , further comprising administering an anti-PD-1 antibody to the patient, wherein the anti-PD-1 antibody is nivolumab or pembrolizumab.

129. The method of claim 128 , wherein the anti-PD-1 antibody is pembrolizumab.

130. The method of claim 129 , wherein the cancer is selected from the group consisting of melanoma, breast cancer, metastatic breast cancer, lung cancer, non-small cell lung cancer, bladder cancer, pancreatic cancer, B-cell lymphoma, Hodgkin lymphoma, diffuse large cell B-cell lymphoma, follicular lymphoma, chronic lymphocytic leukemia, small cell lung cancer, ovarian cancer, kidney cancer, cervical cancer, head and neck cancer, prostate cancer, glioblastoma, non-Hodgkin lymphoma, hepatocellular carcinoma, and multiple myeloma.

131. The method of claim 130 , wherein the cancer is pancreatic cancer.

133. The method of claim 130 , wherein the cancer is non-small cell lung cancer.

134. The method of claim 130 , wherein the cancer is head and neck cancer.

The method of claim 42 , wherein the cancer is selected from the group consisting of melanoma, breast cancer, metastatic breast cancer, lung cancer, non-small cell lung cancer, bladder cancer, pancreatic cancer, B-cell lymphoma, Hodgkin lymphoma, diffuse large cell B-cell lymphoma, follicular lymphoma, chronic lymphocytic leukemia, small cell lung cancer, ovarian cancer, kidney cancer, cervical cancer, head and neck cancer, prostate cancer, glioblastoma, non-Hodgkin lymphoma, hepatocellular carcinoma, and multiple myeloma.

136. The method of claim 135 , wherein the cancer is pancreatic cancer.

137. The method of claim 136 , wherein the one or more chemotherapeutic agents comprise gemcitabine, paclitaxel, or both.

138. The method of claim 135 , wherein the cancer is melanoma.

139. The method of claim 135 , wherein the cancer is non-small cell lung cancer.

140. The method of claim 135 , wherein the cancer is head and neck cancer.

141. The method of claim 135 , further comprising administering an anti-PD-1 antibody to the patient, wherein the anti-PD-1 antibody is nivolumab or pembrolizumab.

142. The method of claim 141 , wherein the anti-PD-1 antibody is pembrolizumab.

143. The method of claim 142 , wherein the cancer is selected from the group consisting of melanoma, breast cancer, metastatic breast cancer, lung cancer, non-small cell lung cancer, bladder cancer, pancreatic cancer, B-cell lymphoma, Hodgkin lymphoma, diffuse large cell B-cell lymphoma, follicular lymphoma, chronic lymphocytic leukemia, small cell lung cancer, ovarian cancer, kidney cancer, cervical cancer, head and neck cancer, prostate cancer, glioblastoma, non-Hodgkin lymphoma, hepatocellular carcinoma, and multiple myeloma.

144. The method of claim 143 , wherein the cancer is pancreatic cancer.

145. The method of claim 143 , wherein the cancer is melanoma.

146. The method of claim 143 , wherein the cancer is non-small cell lung cancer.

147. The method of claim 143 , wherein the cancer is head and neck cancer.

148. The method of claim 142 , wherein the one or more chemotherapeutic agents comprise both gemcitabine and paclitaxel.

149. The method of claim 148 , wherein the cancer is pancreatic cancer.

150. The method of claim 142 , wherein the one or more chemotherapeutic agents consist of cisplatin and carboplatin.

151. The method of claim 150 , wherein the cancer is pancreatic cancer.

152. The method of claim 33 , wherein the anti-CD40 antibody is administered intravenously or subcutaneously to the patient.

153. The method of claim 52 , wherein the one or more chemotherapeutic agents comprise cisplatin or carboplatin.

154. The method of claim 153 , wherein the one or more chemotherapeutic agents further comprises gemcitabine.
