Abstract

The present invention relates to compositions comprising factor IX coagulation factors linked to extended recombinant polypeptide (XTEN), isolated nucleic acids encoding the compositions and vectors and host cells containing the same, and methods of making and using such compositions in treatment of coagulation factor-related diseases, disorders, and conditions.

Claims (21)

. A factor IX fusion protein comprising a factor IX polypeptide and an extended recombinant polypeptide (XTEN), wherein the XTEN comprises an amino acid sequence having at least 90% identity to the amino acid sequence set forth in SEQ ID NO: 36, 37, 38, or any combination thereof.

. The factor IX fusion protein of claim 1 , wherein the XTEN comprises an amino acid sequence having at least 90% sequence identity to the amino acid sequence set forth in SEQ ID NO: 36.

- 18 . (canceled)

. The factor IX fusion protein of claim 1 , wherein the XTEN comprises the amino acid sequence set forth in SEQ ID NO: 36.

. The factor IX fusion protein of claim 1 , wherein the XTEN comprises an amino acid sequence having at least 90% sequence identity to the amino acid sequence set forth in SEQ ID NO: 37.

. The factor IX fusion protein of claim 1 , wherein the XTEN comprises the amino acid sequence set forth in SEQ ID NO: 37.

. The factor IX fusion protein of claim 1 , wherein the XTEN comprises an amino acid sequence having at least 90% sequence identity to the amino acid sequence set forth in SEQ ID NO: 38.

. The factor IX fusion protein of claim 1 , wherein the XTEN comprises the amino acid sequence set forth in SEQ ID NO: 38.

. The factor IX fusion protein of claim 1 , wherein the factor IX polypeptide is linked at its C-terminus to the XTEN.

. The factor IX fusion protein of claim 1 , wherein the factor IX polypeptide is linked at its N-terminus to the XTEN.

. The factor IX fusion protein of claim 1 , wherein the FIX polypeptide comprises an activation peptide sequence, and wherein the XTEN is incorporated within the activation peptide sequence.

. The factor IX fusion protein of claim 1 , which comprises more than one XTEN.

. The factor IX fusion protein of claim 1 , wherein the XTEN comprises an amino acid sequence having at least 90% sequence identity to the amino acid sequence set forth in SEQ ID NO: 243, 265, 271, 297, or any combination thereof.

. The factor IX fusion protein of claim 1 , wherein the XTEN comprises an amino acid sequence having at least 95% sequence identity to the amino acid sequence set forth in SEQ ID NO: 243, 265, 271, 297, or any combination thereof.

. The factor IX fusion protein of claim 1 , wherein the XTEN comprises an amino acid sequence set forth in SEQ ID NO: 243, 265, 271, 297, or any combination thereof.

. The factor IX fusion protein of claim 30 , wherein the XTEN comprises an amino acid sequence comprising SEQ ID NO: 243.

. The factor IX fusion protein of claim 30 , wherein the XTEN comprises an amino acid sequence comprising SEQ ID NO: 271.

. The factor IX fusion protein of claim 30 , wherein the XTEN comprises an amino acid sequence comprising SEQ ID NO: 297.

. The factor IX fusion protein of claim 1 , wherein the XTEN is about 72 amino acids in length.

. The factor IX fusion protein of claim 1 , wherein the fusion protein comprises an amino acid sequence having at least 90% sequence identity to from the amino acid sequence of SEQ ID NO: 602, 604, 606, 608, 612, 614, 616, or 618.

. The factor IX fusion protein of claim 35 , wherein the fusion protein comprises an amino acid sequence set forth in SEQ ID NO: 602, 604, 606, 608, 612, 614, 616, or 618.
