Abstract

A nerve guidance conduit includes one or more guidance channels formed as porous polymeric structures. The guidance channels are within an outer tubular structure that includes randomly-oriented nanofibers. The guidance channels may have electrospun nanofibers on their inner and outer surfaces in a parallel alignment with the guidance channels. Such aligned nanofibers may also be present on the inner surface of the outer tubular structure. The outer surfaces of the guidance channels and the inner surface of the tubular structure define additional guidance channels. Such a nerve guidance conduit provides augmented surface areas for providing directional guidance and enhancing peripheral nerve regeneration. The structure also has the mechanical and nutrient transport requirements required over long regeneration periods.

Claims (19)

1. A nerve guidance conduit for promoting regeneration of a severed nerve, comprising:

a fibrous tube, said fibrous tube defining a lumen within said fibrous tube, and said fibrous tube having a longitudinal axis, an inner surface, an outer surface, a first end, a second end opposite said first end along said longitudinal axis, and a first length from said first end of said fibrous tube to said second end of said fibrous tube;

a plurality of individual tubular members of a spiral cross-sectional shape positioned entirely within said lumen of said fibrous tube, each tubular member of said plurality of tubular members being porous and having a respective outer wall, a respective inner wall, a respective internal guidance channel defined by said respective inner wall, a respective longitudinal axis in substantially parallel alignment with said longitudinal axis of said fibrous tube, a respective first end, a respective second end opposite said respective first end along said respective longitudinal axis, and a respective second length from said respective first end to said respective second end, said respective second length being less than said first length of said fibrous tube, a first one of said plurality of tubular members bordering a first portion of said inner surface of said fibrous tube and a second one of said plurality of tubular members bordering a second portion of said inner surface of said fibrous tube in close proximity to said first tubular member such that said first and second tubular members cooperate with each other and with said inner surface of said fibrous tube to define at least one peripheral guidance channel external of said first and second tubular members but internal of said fibrous tube;

a first plurality of nanofibers randomly distributed on said outer surface of said fibrous tube;

a second plurality of nanofibers arranged on said inner surface of said fibrous tube substantially parallel to said longitudinal axis of said fibrous tube;

a third plurality of nanofibers arranged on said respective inner wall of at least one of said plurality of tubular members substantially parallel to said longitudinal axis of said fibrous tube; and

a fourth plurality of nanofibers arranged on said respective outer wall of at least one of said plurality of tubular members substantially parallel to said longitudinal axis of said fibrous tube, wherein at least some of said fourth plurality of nanofibers are situated in said at least one peripheral guidance channel.

2. The nerve guidance conduit of claim 1 , wherein at least one of said plurality of tubular members includes guiding means for providing guidance for cell migration and directional axonal regeneration.

The nerve guidance conduit of claim 2 , wherein said guiding means includes a plurality of surface channels on said respective outer wall of each of said at least one of said plurality of tubular members, said plurality of surface channels being substantially parallel to each other and to said longitudinal axis of said fibrous tube.

4. The nerve guidance conduit of claim 3 , further comprising a fifth plurality of nanofibers arranged along at least one of said plurality of surface channels substantially parallel to said longitudinal axis of said fibrous tube.

The nerve guidance conduit of claim 2 , wherein said guiding means includes a plurality of surface channels on said respective inner wall of each of said at least one of said plurality of tubular members, said plurality of surface channels being substantially parallel to each other and to said longitudinal axis of said fibrous tube.

6. The nerve guidance conduit of claim 5 , further comprising a fifth plurality of nanofibers arranged along at least one of said plurality of surface channels substantially parallel to said longitudinal axis of said fibrous tube.

The nerve guidance conduit of claim 2 , wherein said guiding means includes a first plurality of surface channels on said respective outer wall of each of said at least one of said plurality of tubular members, said first plurality of surface channels being substantially parallel to each other and to said longitudinal axis of said fibrous tube, and a second plurality of surface channels on said respective inner wall of each of said at least one of said plurality of tubular members, said second plurality of surface channels being substantially parallel to each other and to said longitudinal axis of said fibrous tube.

The nerve guidance conduit of claim 7 , further comprising a fifth plurality of nanofibers arranged along at least one of said first plurality of surface channels substantially parallel to said longitudinal axis of said fibrous tube, and a sixth plurality of nanofibers arranged along at least one of said second plurality of surface channels substantially parallel to said longitudinal axis of said fibrous tube.

9. The nerve guidance conduit of claim 1 , wherein said plurality of tubular members includes at least three tubular members which cooperate to form a central guidance channel bounded solely by said respective outer wall of each of said at least three tubular members.

10. The nerve guidance conduit of claim 9 , wherein said central guidance channel is substantially parallel to said longitudinal axis of said fibrous tube.

11. The nerve guidance conduit of claim 10 , wherein at least some of said fourth plurality of nanofibers are situated in said central guidance channel.

The nerve guidance conduit of claim 1 , further comprising first attachment means for receiving, housing, and gripping a first stump of a severed nerve within said lumen proximate said first end of said fibrous tube, and second attachment means for receiving, housing, and gripping a second stump of a severed nerve within said lumen proximate said second end of said fibrous tube.

The nerve guidance conduit of claim 12 , wherein said first attachment means includes a first chamber within said lumen of said fibrous tube, and wherein said second attachment means includes a second chamber within said lumen of said fibrous tube, whereby said nerve guidance conduit is adapted to bridge a gap between stumps of a severed nerve without stressing them when they are sutured to said nerve guidance conduit.
