Abstract

The invention relates to a method for providing an aseptic chromatography column, said method comprising the steps of: pre-sterilize an empty chromatography column; pre-sterilize a chromatography medium; introducing the pre-sterilized chromatography medium into the pre-sterilized chromatography column using aseptic equipment, thereby providing an aseptic chromatography column comprising chromatography medium.

Claims (27)

1. An aseptic chromatography column comprising a presterilized empty chromatography column (the column) wet packed with a pre-sterilized particulate chromatography medium (the medium), comprising,

a housing comprising an elongated tubular side wall;

opposed, axially spaced first and second end units separated by said side wall, wherein at least one said end unit is axially movable by drive means relative to the other said end unit;

tie rods for securing said first end unit against said second end unit;

a first filter adjacent to said first unit and a second filter adjacent to said second unit which together with the side wall defining an enclosed bed space, containing the medium, wherein at least one of the first and second filters are movable together with the first and the second end units respectively to alter a height of the enclosed bed space;

said first end unit comprising a first valve means for filling the bed space with said particulate medium and a first two way port for adding a liquid to or removing a liquid from the bed space;

said second end unit defining a single passageway having a first end in fluid communication with the enclosed bed space and a second end in fluid communication with a single second two way port for adding a liquid to or removing a liquid from the bed space, wherein the first two way port and the second two way port are at substantially the same level or elevation.

2. The column of claim 1 , wherein the particulate chromatography medium is selected from the group consisting of ion exchange medium, reverse phase medium, size exclusion medium and affinity medium.

3. The column of claim 1 , wherein the first valve means is a one way valve.

4. The column of claim 1 , wherein the first valve means comprises a first valve that is extendable into the bed space in an open configuration and retractable into the first end unit in a closed configuration.

5. The column of claim 1 , wherein the column is disposable.

6. The column of claim 1 , wherein the particulate chromatography medium is introduced in the form of a slurry.

7. The column of claim 6 , wherein the slurry comprises a pre-sterilized dry chromatography medium suspended in a mobile phase.

The column of claim 1 , wherein the first end of the single passageway is in fluid communication and contact with the enclosed bed space and the second end of the single passageway is in fluid communication and contact with the single second 2 way port for adding a liquid to or removing a liquid from the bed space.

9. An aseptic chromatography column comprising a pre-sterilized empty chromatography column (the column) wet packed with a pre-sterilized particulate chromatography medium (the medium), comprising,

a housing comprising an elongated tubular side wall;

opposed, axially spaced first and second end units separated by said side wall, wherein the second end unit is fixed and the first end unit is axially movable by drive means relative to the second unit;

tie rods for securing said first end unit against said second end unit;

a first filter adjacent to said first unit and a second filter adjacent to said second unit which together with the side wall defining an enclosed bed space, containing the medium, wherein the first filter is movable together with the first end unit to alter a height of the enclosed bed space;

said first end unit comprising a first valve means for filling the bed space with said particulate medium and a first port for adding a liquid to or removing a liquid from the bed space;

said second end unit defining a single passageway having a first end in fluid communication with the enclosed bed space and a second end in fluid communication with a single second port for adding a liquid to or removing a liquid from the bed space, wherein the first port and the single second port are at substantially the same level or elevation.

10. The column of claim 9 , wherein the particulate chromatography medium is selected from the group consisting of ion exchange medium, reverse phase medium, size exclusion medium and affinity medium.

11. The column of claim 9 , wherein the first valve means is a one way valve.

12. The column of claim 9 , wherein the first valve means comprises a first valve that is extendable into the bed space in an open configuration and retractable into the first end unit in a closed configuration.

13. The column of claim 9 , wherein the column is disposable.

14. The column of claim 9 , wherein the particulate chromatography medium is introduced in the form of a slurry.

15. The column of claim 14 , wherein the slurry comprises a pre-sterilized dry chromatography medium suspended in a mobile phase.
