Abstract

The invention generally relates to a system for isolating or separating a target from a sample. In certain aspects, processes performed by the target capture system include introducing a plurality of magnetic particles, in which a plurality of the particles include at least one binding moiety specific to a target, into a sample to form at least one target/particle complex and applying a magnetic field to isolate the magnetic particle/target complexes from the sample. The process starts at inputting a sample into the system and ends at delivering a capture target or nucleic acids of the target into a container for further analysis.

Claims (13)

1. A device for capturing a pathogen, the device comprising:

a fluidic cartridge comprising at least one chamber for holding and releasing magnetic particles into a sample;

a cartridge/vessel interface that removably couples a sample collection tube containing a sample to the cartridge; and

at least a first magnetic trap for receiving a sample/magnetic particle mixture from the sample collection tube via the cartridge/vessel interface, wherein the cartridge/vessel interface includes

an output member operable to introduce a fluid from the cartridge into the sample collection tube, and

an input member operable to transfer the sample and the fluid and substances from the sample collection tube into the cartridge.

2. The device of claim 1 , wherein the output member comprises a lumen and a penetrating tip designed to penetrate and extend into the vessel.

The device of claim 1 , wherein the magnetic trap engages with a magnetic assembly to isolate the magnetic particles from the sample, the cartridge further comprising a second magnetic trap, in which both magnetic traps engage with the magnetic assembly to isolate magnetic particles from a sample.

4. The device of claim 3 , wherein the cartridge comprises a lysing mechanism operably associated with the second magnetic trap and operable to lyse at least one target cell bound to at least one of the magnetic particles within the second magnetic trap.

5. The device of claim 1 , wherein the cartridge further comprises a matrix for receiving lysate from the first magnetic trap and retaining nucleic acid from the lysate.

6. The device of claim 5 , wherein the cartridge further comprises a reaction chamber in communication with the matrix for pre-treating the lysate.

7. The device of claim 1 , wherein the chamber contains a plurality of magnetic particles to release into the sample collection tube, further wherein the magnetic particles each comprises one or more binding moieties specific to a pathogen within the sample.

8. The device of claim 1 , wherein the cartridge includes both macrofluidic and microfluidic components so that the cartridge can process both macrofluidic and microfluidic volumes of fluid.
