Abstract

The present invention relates to novel compounds of formula (I), process for preparation of the same and composition comprising these compounds.

Claims (306)

1. A compound of the formula (I):

wherein,

X 1 , X 2 , X 3 , X 4 and X 5 are each independently N, O, S or CH; and

X 4 and X 5 combine to form a five membered ring comprising one or more of heteroatoms each independently selected from N, O and S and the additional five membered ring may be further optionally substituted with one or more of the group selected from F, Cl, Br, I, CF 3 and C 1-6 alkyl;

wherein “ ” is a single or a double bond

R 1 and R 2 is independently selected from the group comprising —H, —O, C 1-6 alkyl, C 1-6 alkoxy, —(CH 2 )n, amino, —CO, —CONH, —NH(Alkyl), —N(Alkyl) 2 , —NH-aralkyl, —CH 2 O, —OCH(CH 3 ), halogen, COOR 3 , —CONR 3 R 4 , and NR 3 COR 4 ;

R 3 and R 4 is independently selected from the group comprising hydrogen and C 1-6 straight chain or branched chain alkyl which may be further substituted with halogen or C 1-6 alkyl;

n is 0, 1, 2 or 3;

8. The compound of claim 2 showing significant dose dependent glucose reduction at both 3mpk and 10mpk in oral glucose tolerance test in mice.

9. The compound of claim 2 showing active GLP-1 secretion greater than ˜1 fold with respect to the vehicle.

18. The compound of claim 1 , which is

2-(1-(1-(3-isopropyl-1,2,4-oxadiazol-5-yl)piperidin-4-yl)ethoxy)-6-(thiazol-5-yl)imidazo[2,1-b][1,3,4]thiadiazole;

2-((S)-1-(1-(3-isopropyl-1,2,4-oxadiazol-5-yl)piperidin-4-yl)ethoxy)-6-(2-methyl-4-(methylsulfonyl)phenyl)imidazo[2,1-b][1,3,4]thiadiazole; or

ethyl 4-((S)-1-(6-(2-fluoro-4-(methylsulfonyl)phenyl)imidazo[2,1-b][1,3,4]thiadiazol-2-yloxy)ethyl)piperidine-1-carboxylate.

19. A method of treating diseases and conditions mediated through GPR119 comprising administering the compound of claim 1 to a patient in need thereof.

20. A method of treatment diseases and conditions mediated through GPR119 comprising administering the compound of claim 2 to a patient in need thereof.

21. The method of claim 19 , wherein said disease is metabolic disorder.

22. The method of claim 20 , wherein said disease is metabolic disorder.

23. The method of claim 21 , wherein said disease is diabetes.

24. The method of claim 22 , wherein said disease is diabetes.

25. The method of claim 23 , wherein said disease is Type II diabetes.

26. The method of claim 24 , wherein said disease is Type II diabetes.
