Methods for treating aging and skin disorders using nucleic acids targeting Tyr or MMP1
The present invention relates to RNAi constructs with improved tissue and cellular uptake characteristics and methods of use of these compounds in dermal applications.
1. A chemically modified double stranded nucleic acid molecule, wherein the chemically modified double stranded nucleic acid molecule comprises an antisense strand and a sense strand, wherein the region of the molecule that is double stranded is from 8-15 nucleotides long, wherein the antisense strand contains a single stranded region that is 4-12 nucleotides long, wherein the single stranded region of the antisense strand contains 3, 4, 5, 6, 7, 8, 9, 10, 11 or 12 phosphorothioate modifications, wherein at least 40% of the nucleotides of the double stranded nucleic acid are modified, and wherein the sense strand comprises at least 12 contiguous nucleotides of a sequence selected from SEQ ID NOs 258, 260, 262 and 263 and/or the antisense strand comprises at least 12 contiguous nucleotides of a sequence selected from SEQ ID NOs 297, 298, 299, 300 and 301.
2. The chemically modified double stranded nucleic acid molecule of claim 1 , wherein:
the sense strand sequence is selected from the group consisting of SEQ ID NOs 258, 260, 262 and 263, including the modification indicated in Table 7, and the antisense strand sequence is selected from the group consisting of SEQ ID NOs 297, 298, 299, 300 and 301, including the modifications indicated in Table 8.
3. A method for treating retinitis pigmentosa, Addison's disease, neuroblastoma, glioblastoma or Parkinson's disease comprising administering to a subject in need thereof a therapeutically effective amount of a chemically modified double stranded nucleic acid molecule of claim 1 .
4. A composition comprising a chemically modified double stranded nucleic acid molecule of claim 1 .
