Abstract

A hyaluronic acid product including a cross-linked hyaluronic acid and one or more cyclodextrin molecules. The hyaluronic acid is cross-linked by ether bonds, and the one or more cyclodextrin molecules are grafted onto the cross-linked hyaluronic acid by amide bonds, preferably using a triazine-based coupling reagent. A process of preparing a hyaluronic acid product including a cross-linked hyaluronic acid and one or more cyclodextrin molecules.

Claims (27)

1. A hyaluronic acid product comprising a cross-linked hyaluronic acid and one or more aminocyclodextrin molecules, wherein the hyaluronic acid is cross-linked by ether bonds, and wherein the one or more aminocyclodextrin molecules are grafted onto the cross-linked hyaluronic acid by direct amide bonds between an amino group of the aminocyclodextrin molecule(s) and a carboxyl group of the cross-linked hyaluronic acid.

2. The hyaluronic acid product according to claim 1 , wherein the aminocyclodextrin molecule is constituted by 6-8 glucopyranoside units.

3. The hyaluronic acid product according to claim 2 , wherein the aminocyclodextrin molecule is selected from the group consisting of 2 aminocyclodextrin, 3-aminocyclodextrin and 6-aminocyclodextrin.

4. The hyaluronic acid product according to claim 1 , wherein the aminocyclodextrin molecules and disaccharide units of the cross-linked hyaluronic acid have a molar ratio of more than 2%.

5. The hyaluronic acid product according to claim 1 , wherein the hyaluronic acid product is in the form of gel particles having an average size in the range of 0.01-5 mm.

The hyaluronic acid product according to claim 1 , further comprising a guest molecule capable of forming a guest-host complex with the aminocyclodextrin molecule acting as a host, wherein the guest molecule is selected from drugs and/or biologically active substances used in the treatment of disorders in the field of dermatology, aesthetics, ophthalmology, gynaecology, oncology, angiology, neurology, orthopaedics, rheumatology or aesthetic dermatology, such as anti-infective agents, antimicrobials, anti-inflammatory agents, cytostatic, cytotoxic, antiviral, anaesthetic, hemostatic, vasoconstrictor agents or growth factors.

A process of preparing a hyaluronic acid product comprising a cross-linked hyaluronic acid and one or more cyclodextrin molecules, wherein the hyaluronic acid is cross-linked by ether bonds, and wherein the one or more aminocyclodextrin molecules are grafted onto the cross-linked hyaluronic acid by direct amide bonds between an amino group of the aminocyclodextrin molecule(s) and a carboxyl group of the cross-linked hyaluronic acid, the process comprising:

(a) providing (i) hyaluronic acid and (ii) one or more cyclodextrin molecules;

(b) cross-linking the hyaluronic acid by ether bonds using a bi- or polyfunctional cross-linking agent; and

(c) grafting the one or more cyclodextrin molecules onto the cross-linked hyaluronic acid by amide bonds.

8. The process according to claim 7 , wherein the cyclodextrin molecule is an aminocyclodextrin molecule, and wherein an amino group of the aminocyclodextrin molecule forms said amide bond with a carboxyl group of the cross-linked hyaluronic acid.

9. The process according to claim 7 , wherein the grafting of step (c) comprises:

(i) activating the carboxyl groups on the cross-linked hyaluronic acid with a triazine-based coupling reagent to form an activated, cross-linked hyaluronic acid; and

(ii) coupling the one or more cyclodextrin molecules to the carboxyl groups of the activated cross-linked hyaluronic acid by amide bonds.

10. The process according to claim 9 , wherein the triazine-based coupling reagent is 4-(4,6-dimethoxy-1,3,5-triazin-2-yl)-4-methylmorpholinium chloride (DMTMM).

The process according to claim 7 , wherein said bi- or polyfunctional cross-linking agent is selected from the group consisting of 1,4-butanediol diglycidyl ether (BODE), 1,2-bis(2,3-epoxypropoxy)ethylene (EGDGE) and ethylene glycol diglycidyl ether (EGDE), 1,2-ethanediol diglycidyl ether (EDDE) and diepoxy octane.

12. The process according to claim 11 , wherein said bi- or polyfunctional cross-linking agent is 1,4-butanediol diglycidyl ether (BODE).

13. The process according to claim 7 , wherein the particles of the hyaluronic acid product have an average size in the range of 0.01-5 mm.

14. A process of preparing a formulation comprising contacting:

(i) a solution of a biologically active molecule with

(ii) a hyaluronic acid product comprising a cross-linked hyaluronic acid and one or more aminocyclodextrin molecules, wherein the hyaluronic acid is cross-linked by ether bonds, and wherein the one or more aminocyclodextrin molecules are grafted onto the cross-linked hyaluronic acid by direct amide bonds between an amino group of the aminocyclodextrin molecule(s) and a carboxyl group of the cross-linked hyaluronic acid, to form a complex and optionally

(d) recovering the complex bound to the cross-linked hyaluronic acid.

15. The process according to claim 14 , wherein the cyclodextrin molecule is an aminocyclodextrin molecule, and wherein an amino group of the aminocyclodextrin molecule forms said amide bond with a carboxyl group of the cross-linked hyaluronic acid.

16. The process according to claim 14 , wherein the grafting of step (c) comprises:

(i) activating the carboxyl groups on the cross-linked hyaluronic acid with a triazine-based coupling reagent to form an activated, cross-linked hyaluronic acid; and

(ii) coupling the one or more cyclodextrin molecules to the carboxyl groups of the activated cross-linked hyaluronic acid by amide bonds.

17. The process according to claim 16 , wherein the triazine-based coupling reagent is 4-(4,6-dimethoxy-1,3,5-triazin-2-yl)-4-methylmorpholinium chloride (DMTMM).
