Abstract

The invention relates to biologically functional scaffolds having a porous structure, methods of preparing, and methods of use thereof. The invention also relates to methods of repairing a defect, methods of culturing cells and promoting differentiation of stem cells using the same.

Claims (15)

1. A method of preparing a porous soft tissue, comprising loosening a network of extracellular materials in a native soft tissue in the presence of a solid at a temperature of 0-50 ° C.; dissolving the solid during and/or after the loosening; and freezing and drying the loosened porous soft tissue after the dissolving, whereby a dry porous soft tissue is produced, wherein the dry porous soft tissue has more space among the extracellular materials and an increased void voume as compared with the native soft tissue and does not comprise a non-naturally occurring crosslinker.

2. The method of claim 1 , wherein the dry porous soft tissue does not comprise a non-naturally occurring carrier.

3. The method of claim 1 , wherein the method does not include crosslinking the native soft tissue or the porous soft tissue by a non-naturally occurring bond.

4. The method of claim 1 , further comprising sterilizing the porous soft tissue.

5. The method of claim 1 , wherein the loosening is carried out at a temperature below 24° C.

6. The method of claim 1 , wherein the solid comprises ice granulate, salt granulate, sugar granulate, or a combination thereof.

7. The method of claim 1 , wherein a weight percentage of the onc or morc native soft tissucs tissue in the disperscd porous soft tissue is from 2% to 80% in a dry state.

8. The method of claim 1 , wherein the native soft tissue is decellularized and devitalized.

9. The method of claim 1 , wherein the native soft tissue is selected from the group consisting of muscle, fat, blood vessel, nerve, tendon, ligament, lining of joints, skin, dermis, pericardium, fascia, cartilage, dura mata, endocardium, mucosal tissue placental membrane, periosteum, bladder, small or large intestine, urethra, and placenta.

10. A method of preparing a biologically functional scaffold, comprising:

a. loosening a network of extracellular materials in a native soft tissue in the presence of a solid at a temperature of 0-50 ° C., whereby a dispersed porous soft tissue is produced, wherein the porous soft tissue has more space among the extracellular materials and an increased void volume as compared with the native soft tissue and comprises randomly interwoven collagen fibers or collagen fiber bundles;

b. dissolving the solid during and/or after the loosening;

c. shaping the dispersed porous soft tissue to a scaffold to generate a shaped dispersed porous soft tissue; and

d. dehydrating the shaped dispersed porous soft tissue, whereby a biologically functional scaffold is prepared.

11. The method of claim 10 , wherein the solid comprises ice granulate, salt granulate, sugar granulate, or a combination thereof.
