Abstract

The invention relates to substituted piperazine compounds and methods of use and uses thereof, and further to the pharmaceutical compositions comprising the compounds and uses thereof, wherein the compound has Formula (I) or a stereoisomer, a tautomer, an N-oxide, a solvate, a metabolite, a pharmaceutically acceptable salt or a prodrug thereof. The substituted piperazine compounds and pharmaceutical compositions comprising the compounds disclosed herein can be used for inhibiting 5-hydroxytryptamine reuptake and/or stimulating 5-HT 1A receptors. The invention also relates to processes for preparing these compounds and pharmaceutical compositions, and their uses in the treatment of a central nervous system dysfunction.

Claims (34)

1. A compound having Formula (I) or Formula (II) or a stereoisomer, a tautomer, an N-oxide, a solvate, or a pharmaceutically acceptable salt thereof,

or

wherein:

X is CH or N;

r is 0, 1, 2 or 3;

W is one of the following heteroarylene rings:

wherein each of Formula (W-1) to (W-3) and (W5) to (W-14) is optionally and independently substituted with one, two, three or four R x groups;

each R x is independently H, D, F, Cl, Br, I, —NO 2 , —CN, —NR a R b , —OR c , —SR c , —C(═O)R d , —C(═O)OR c , —C(═O)NR a R b , —OC(═O)R d , —N(R a )C(═O)R d , —S(═O)R d , —S(═O) 2 R d , —S(═O) 2 OR c , —S(═O) 2 NR a R b , —N(R a )S(═O) 2 R d , oxo (═O), C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl or C 1 -C 6 haloalkyl;

each R 1 is independently H, D, F, Cl, Br, I, —NO 2 , —CN, —NH 2 , —OH, —SH, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 1 -C 6 haloalkyl, C 1 -C 6 alkoxyl, C 1 -C 6 haloalkoxy, (C 1 -C 6 alkoxyl)-(C 1 -C 4 alkylene)-, C 1 -C 6 alkylamino, (C 1 -C 6 alkylamino)-(C 1 -C 4 alkylene)-, C 1 -C 6 alkylthio or (C 1 -C 6 alkylthio)-(C 1 -C 4 alkylene)-;

each R 2 is independently H, D, F, Cl, C 1 -C 6 alkyl, C 1 -C 6 haloalkyl, —NR a R b , —OR c , —C(═O)R d , —C(═O)OR c , —C(═O)NR a R b or (C 6 -C 10 aryl)-(C 1 -C 6 alkylene)-, or two adjacent R 2 , together with the carbon atoms to which they are attached, form a C 3 -C 6 carbocyclic ring, a benzene ring, a 3- to 7-membered heterocyclic ring or a 5- to 6-membered heteroaromatic ring, or two R 2 attached to one carbon atom, together with the carbon atom form a C 3 -C 6 carbocyclic ring or a 3- to 7-membered heterocyclic ring;

each R 3 is independently H, D, F, Cl, Br, I, —NO 2 , —CN, —NR a R b , —OR c , —SR c , —C(═O)R d , —C(═O)OR c , —C(═O)NR a R b , —OC(═O)R d , —N(R a )C(═O)R d , —S(═O)R d , —S(═O) 2 R d , —S(═O) 2 OR c , —S(═O) 2 NR a R b , —N(R a )S(═O) 2 R d , —N(R a )C(═O)OR c , —N(R a )C(═O)NR a R b , alkyl, alkenyl, alkynyl, haloalkyl, cycloalkyl, cycloalkylalkylene, heterocyclyl, heterocyclylalkylene, aryl, arylalkylene, heteroaryl or heteroarylalkylene, or two adjacent R 3 , together with the carbon atoms to which they are attached, form a methylenedioxy or ethylenedioxy group, wherein each —NR a R b , —OR c , —SR c , —C(═O)R d , —C(═O)OR c , —C(═O)NR a R b , —OC(═O)R d , —N(R a )C(═O)R d , —S(═O)R d , —S(═O) 2 R d , —S(═O) 2 OR c , —S(═O) 2 NR a R b , —N(R a )S(═O) 2 R d , —N(R a )C(═O)OR c , —N(R a )C(═O)NR a R b , alkyl, alkenyl, alkynyl, haloalkyl, cycloalkyl, cycloalkylalkylene, heterocyclyl, heterocyclylalkylene, aryl, arylalkylene, heteroaryl, heteroarylalkylene, methylenedioxy and ethylenedioxy is optionally and independently substituted with one or more R 4 groups;

each R 4 is independently F, Cl, Br, I, —NO 2 , —CN, —N 3 , —NH 2 , —OH, —SH, C 1 -C 4 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 1 -C 4 haloalkyl, C 1 -C 4 alkoxyl, C 1 -C 4 haloalkoxy, C 1 -C 4 alkylamino, C 1 -C 4 alkylthio, NH 2 —(C 1 -C 4 alkylene)-, HO—(C 1 -C 4 alkylene)-, HS—(C 1 -C 4 alkylene)-, (C 1 -C 4 alkylamino)-(C 1 -C 4 alkylene)-, (C 1 -C 4 alkoxyl)-(C 1 -C 4 alkylene)-, (C 1- C 4 alkylthio)-(C 1 -C 4 alkylene)-, C 3 -C 6 cycloalkyl, (C 3 -C 6 cycloalkyl)-(C 1 -C 4 alkylene)-, 3- to 7-membered heterocyclyl, (3- to 7-membered heterocyclyl)-(C 1 -C 4 alkylene)-, phenyl, phenyl-(C 1 -C 4 alkylene)-, 5- to 6-membered heteroaryl or (5- to 6-membered heteroaryl)-(C 1 -C 4 alkylene)-;

each R a and R b is independently H, C 1 -C 6 alkyl, C 3 -C 6 alkenyl, C 3 -C 6 alkynyl, C 1 -C 6 haloalkyl, C 3 -C 6 cycloalkyl, (C 3 -C 6 cycloalkyl)-(C 1 -C 4 alkylene)-, 3- to 7-membered heterocyclyl, (3- to 7-membered heterocyclyl)-(C 1 -C 4 alkylene)-, phenyl, phenyl-(C 1 -C 4 alkylene)-, 5- to 6-membered heteroaryl or (5- to 6-membered heteroaryl)-(C 1 -C 4 alkylene)-, or R a and R b together with the nitrogen atom to which they are attached, form a 3- to 7-membered heterocyclic ring;

each R c and R d is independently H, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 1 -C 6 haloalkyl, C 3 -C 6 cycloalkyl, (C 3 -C 6 cycloalkyl)-(C 1 -C 4 alkylene)-, 3- to 7-membered heterocyclyl, (3- to 7-membered heterocyclyl)-(C 1 -C 4 alkylene)-, phenyl, phenyl-(C 1 -C 4 alkylene)-, 5- to 6-membered heteroaryl or (5- to 6-membered heteroaryl)-(C 1 -C 4 alkylene)-;

each m and n is independently 0, 1, 2, 3, or 4;

k is 0, 1, 2, 3, 4, 5 or 6; and

p is 0, 1, 2, 3, 4, or 5.

2. The compound according to claim 1 having Formula (III), or a stereoisomer, a tautomer, an N-oxide, a solvate, or a pharmaceutically acceptable salt thereof,

wherein Y is O, S or NH.

3. The compound according to claim 1 having Formula (V) or (VI) or a stereoisomer, a tautomer, an N-oxide, a solvate, or a pharmaceutically acceptable salt thereof,

wherein X is CH or N;

Y is O, S or NH; and

r is 0, 1, 2 or 3.

The compound according to claim 1 , wherein each R x is independently H, D, F, Cl, —NO 2 , —CN, —NH 2 , —OH, -Me, -Et, -(n-Pr), -(i-Pr), -(t-Bu), —CHF 2 , —CF 3 , —CH 2 CF 3 , —CF 2 CHF 2 , —CHFCF 3 , —CF 2 CF 3 , —CF 2 CH 2 CH 3 , —CF 2 CH 2 CF 3 , —CF 2 CH 2 CHF 2 , —CH 2 CHFCH 3 , —CH 2 CF 2 CH 3 , —CH 2 CF 2 CF 3 , —CH 2 CF 2 CHF 2 , —CH 2 CH 2 Cl, —OMe, —OEt, —O(i-Pr), —O(t-Bu), —NMe 2 , —C(═O)H, —C(═O)OH, —C(═O)OMe, —C(═O)OEt, —C(═O)NH 2 , —C(═O)NMe 2 or oxo (═O).

5. The compound according to claim 1 , wherein each R 1 is independently H, D, F, Cl, Br, —CN, —NO 2 , —NH 2 , —NMe 2 , —OH, —OMe, —OEt, —O(i-Pr), —O(t-Bu), -Me, -Et, -(n-Pr), -(i-Pr), -(t-Bu) or —CF 3 .

6. The compound according to claim 1 , wherein each R 2 is independently H, D, F, Cl, —NH 2 , —OH, -Me, -Et, -(n-Pr), -(i-Pr), -(t-Bu), —CF 3 , —OMe, —O(i-Pr) or —O(t-Bu).

The compound according to claim 1 , wherein each R 3 is independently H, D, F, Cl, Br, I, —NO 2 , —CN, —NR a R b , —OR c , —SR c , —C(═O)R d , —C(═O)OR c , —C(═O)NR a R b , —OC(═O)R d , —N(R a )C(═O)R d , —S(═O)R d , —S(═O) 2 R d , —S(═O) 2 OR c , —S(═O) 2 NR a R b , —N(R a )S(═O) 2 R d , —N(R a )C(═O)OR c , —N(R a )C(═O)NR a R b , C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 1 -C 6 haloalkyl, C 3 -C 10 cycloalkyl, (C 3 -C 10 cycloalkyl)-(C 1 -C 6 alkylene)-, 3- to 10-membered heterocyclyl, (3- to 10-membered heterocyclyl)-(C 1 -C 6 alkylene)-, C 6 -C 10 aryl, (C 6 -C 10 aryl)-(C 1 -C 6 alkylene)-, 5- to 10-membered heteroaryl or (5- to 10-membered heteroaryl)-(C 1 -C 6 alkylene)-, or two adjacent R 3 , together with the carbon atoms to which they are attached, form a methylenedioxy or ethylenedioxy group, wherein the —NR a R b , —OR c , —SR c , —C(═O)R d , —C(═O)OR c , —C(═O)NR a R b , —OC(═O)R d , —N(R a )C(═O)R d , —S(═O)R d , —S(═O) 2 R d , —S(═O) 2 OR c , —S(═O) 2 NR a R b , —N(R a )S(═O) 2 R d , —N(R a )C(═O)OR c , —N(R a )C(═O)NR a R b , C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 1 -C 6 haloalkyl, C 3 -C 10 cycloalkyl, (C 3 -C 10 cycloalkyl)-(C 1 -C 6 alkylene)-, 3- to 10-membered heterocyclyl, (3- to 10-membered heterocyclyl)-(C 1 -C 6 alkylene)-, C 6 -C 10 aryl, (C 6 -C 10 aryl)-(C 1 -C 6 alkylene)-, 5- to 10-membered heteroaryl, (5- to 10-membered heteroaryl)-(C 1 -C 6 alkylene)-, methylenedioxy and ethylenedioxy are each optionally and independently substituted with one or more R 4 groups.

The compound according to claim 1 , wherein each R 3 is independently H, D, F, Cl, Br, —CN, —NO 2 , —NH 2 , —NMe 2 , —NHEt, —NEt 2 , —OH, —OMe, —OEt, —O(n-Pr), —O(i-Pr), —O(t-Bu), -Me, -Et, -(n-Pr), -(i-Pr), -(t-Bu), —CF 3 , —CH 2 CH 2 Cl, —OCHF 2 , —OCF 3 , —OCH 2 CF 3 , —OCF 2 CHF 2 , —OCHFCF 3 , —OCF 2 CF 3 , —OCF 2 CH 2 CH 3 , —OCF 2 CH 2 CF 3 , —OCF 2 CH 2 CHF 2 , —OCH 2 CHFCH 3 , —OCH 2 CF 2 CH 3 , —OCH 2 CF 2 CF 3 , —OCH 2 CF 2 CHF 2 , —CONH 2 , —CONHMe, —CONMe 2 , —C(═O)OMe, —C(═O)OEt, —NHC(═O)H, —NHC(═O)CH 3 , —NHC(═O)OMe or phenyl.

The compound according to claim 1 , wherein each R a and R b is independently H, C 1 -C 4 alkyl, C 3 -C 4 alkenyl, C 3 -C 4 alkynyl, C 1 -C 4 haloalkyl, C 5 -C 6 cycloalkyl, (C 5 -C 6 cycloalkyl)-(C 1 -C 4 alkylene)-, 5- to 7-membered heterocyclyl, (5- to 7-membered heterocyclyl)-(C 1 -C 4 alkylene)-, phenyl, phenyl-(C 1 -C 4 alkylene)-, 5- to 6-membered heteroaryl or (5- to 6-membered heteroaryl)-(C 1 -C 4 alkylene)-, or R a and R b together with the nitrogen atom to which they are attached, form a 5- to 7-membered heterocyclic ring; and

each R c and R d is independently H, C 1 -C 4 alkyl, C 2 -C 4 alkenyl, C 2 -C 4 alkynyl, C 1 -C 4 haloalkyl, C 5 -C 6 cycloalkyl, (C 5 -C 6 cycloalkyl)-(C 1 -C 4 alkylene)-, 5- to 7-membered heterocyclyl, (5- to 7-membered heterocyclyl)-(C 1 -C 4 alkylene)-, phenyl, phenyl-(C 1 -C 4 alkylene)-, 5- to 6-membered heteroaryl or (5- to 6-membered heteroaryl)-(C 1 -C 4 alkylene)-.

The compound according to claim 1 , wherein each R a and R b is independently H, -Me, -Et, -(n-Pr), -(i-Pr), -(t-Bu), —CF 3 , —CH 2 CH 2 Cl, cyclopentyl, pyrrolidinyl, piperidinyl, morpholinyl or piperazinyl, or R a and R b together with the nitrogen atom to which they are attached, form a 5- to 6-membered heterocyclic ring; and

each R c and R d is independently H, -Me, -Et, -(n-Pr), -(i-Pr), -(t-Bu), —CHF 2 , —CF 3 , —CH 2 CF 3 , —CF 2 CHF 2 , —CHFCF 3 , —CF 2 CF 3 , —CF 2 CH 2 CH 3 , —CF 2 CH 2 CF 3 , —CF 2 CH 2 CHF 2 , —CH 2 CHFCH 3 , —CH 2 CF 2 CH 3 , —CH 2 CF 2 CF 3 , —CH 2 CF 2 CHF 2 , —CH 2 CH 2 Cl, cyclopentyl, pyrrolidinyl, piperidinyl, morpholinyl or piperazinyl.

11. The compound according to claim 1 having one of the following structures or a stereoisomer, a tautomer, an N-oxide, a solvate, or a pharmaceutically acceptable salt thereof:

12. A pharmaceutical composition comprising the compound of claim 1 ; and a pharmaceutically acceptable excipient, carrier, adjuvant or a combination thereof.
