Abstract

This invention relates to QTL responsible for significantly increased firmness in tomato fruit in the cultivated plant producing said tomato fruit, compared to fruit from a control tomato plant which does not have said genetic elements. A cultivated tomato plant producing tomato fruit with significantly increased fruit firmness and a method for detecting QTLs linked to significantly increased fruit firmness are also provided.

Claims (36)

1. A method for selecting a cultivated Solanum lycopersicum tomato plant with fruit having increased firmness as compared with fruit from a control Solanum lycopersicum tomato plant, the method comprising:

a. detecting in the cultivated Solanum lycopersicum tomato plant the presence of QTL1, wherein detecting the presence of QTL1 comprises:

i. providing a sample of genomic DNA from the cultivated Solanum lycopersicum tomato plant; and

ii. detecting in the sample of genomic DNA at least one DNA marker linked to QTL1 selected from DNA markers TG599, NT3374, TG246, and NT5880, wherein:

DNA marker TG599 can be detected in a nucleic acid amplification reaction by a forward primer of SEQ ID NO: 19 and a reverse primer of SEQ ID NO: 20,

DNA marker NT3374 can be detected in a nucleic acid amplification reaction by a forward primer of SEQ ID NO: 7 and a reverse primer of SEQ ID NO: 8,

DNA marker TG246 can be detected in a nucleic acid amplification reaction by a forward primer of SEQ ID NO: 17 and a reverse primer of SEQ ID NO: 18, and

DNA marker NT5880 can be detected in a nucleic acid amplification reaction by a forward primer of SEQ ID NO: 9 and a reverse primer of SEQ ID NO: 10; and

b. selecting a cultivated Solanum lycopersicum tomato plant comprising QTL1, thereby selecting a cultivated Solanum lycopersicum tomato plant with fruit having increased firmness as compared with a control Solanum lycopersicum tomato plant that does not comprise QTL1.

2. The method according to claim 1 , wherein the fruit from the cultivated Solanum lycopersicum tomato plant has increased fruit firmness at the harvesting stage as compared with the fruit from the control Solanum lycopersicum tomato plant.

3. A method of introgressing a trait for increased fruit firmness from a donor tomato plant, which produces fruit with increased firmness as compared with fruit from a control Solanum lycopersicum tomato plant, into a recipient Solanum lycopersicum tomato plant that does not comprise the trait, wherein the donor tomato plant comprises a genetic element that is identical to the genetic element of Solanum lycopersicum Q7774 deposited with NCIMB under accession number 41949, the method comprising:

a. crossing the donor tomato plant with the recipient Solanum lycopersicum tomato plant to provide offspring Solanum lycopersicum tomato plants;

b. detecting in offspring Solanum lycopersicum tomato plants the presence of QTL1, wherein detecting the presence of QTL1 comprises:

i. isolating genomic DNA from the offspring Solanum lycopersicum tomato plant; and

ii. detecting in the genomic DNA at least one DNA marker linked to QTL1 selected from DNA markers TG599, NT3374, TG246, and NT5880, wherein:

DNA marker TG599 can be detected in a nucleic acid amplification reaction by a forward primer of SEQ ID NO: 19 and a reverse primer of SEQ ID NO: 20,

DNA marker NT3374 can be detected in a nucleic acid amplification reaction by a forward primer of SEQ ID NO: 7 and a reverse primer of SEQ ID NO: 8,

DNA marker TG246 can be detected in a nucleic acid amplification reaction by a forward primer of SEQ ID NO:17 and a reverse primer of SEQ ID NO:18, and

DNA marker NT5880 can be detected in a nucleic acid amplification reaction by a forward primer of SEQ ID NO: 9 and a reverse primer of SEQ ID NO: 10; and

c. selecting an offspring Solanum lycopersicum tomato plant comprising QTL1, thereby introgressing the trait for increased fruit firmness from the donor tomato plant into the recipient Solanum lycopersicum tomato plant.

4. The method according to claim 3 , wherein the fruit from the donor tomato plant has increased fruit firmness at the harvesting stage as compared with the fruit from the control Solanum lycopersicum tomato plant.

5. A method of introducing a trait for increased fruit firmness into a hybrid Solanum lycopersicum tomato variety, the method comprising:

a. crossing a donor tomato plant that provides fruit with increased firmness compared with fruit from a control Solanum lycopersicum tomato plant with a recipient inbred Solanum lycopersicum tomato plant to provide F1 progeny plants, wherein the donor tomato plant comprises a genetic element that is identical to the genetic element of Solanum lycopersicum Q7774 deposited with NCIMB under accession number 41949;

b. selecting F1 progeny plants that have increased fruit firmness;

c. backcrossing the selected progeny plant(s) with the recipient inbred Solanum lycopersicum tomato plant to produce backcross progeny plants;

d. selecting for backcross progeny plants that have increased fruit firmness and morphological and physiological characteristics of the recipient inbred Solanum lycopersicum tomato plant, wherein the selection for increased fruit firmness comprises:

i. isolating genomic DNA of the backcross progeny; and

ii. testing the isolated genomic DNA for the presence of at least one DNA marker linked to QTL1 selected from DNA markers TG599, NT3374, TG246, and NT5880, wherein:

DNA marker TG599 can be detected in a nucleic acid amplification reaction by a forward primer of SEQ ID NO: 19 and a reverse primer of SEQ ID NO: 20,

DNA marker NT3374 can be detected in a nucleic acid amplification reaction by a forward primer of SEQ ID NO: 7 and a reverse primer of SEQ ID NO: 8,

DNA marker TG246 can be detected in a nucleic acid amplification reaction by a forward primer of SEQ ID NO: 17 and a reverse primer of SEQ ID NO: 18, and

DNA marker NT5880 can be detected in a nucleic acid amplification reaction by a forward primer of SEQ ID NO: 9 and a reverse primer of SEQ ID NO: 10; and

e. repeating steps (c) and (d) two or more times in succession to produce selected third or higher backcross progeny,

f. optionally, selfing selected backcross progeny in order to identify homozygous plants; and

g. crossing at least one of the backcross progeny or selfed plants with another inbred Solanum lycopersicum tomato plant to generate a hybrid Solanum lycopersicum tomato variety with increased fruit firmness and all of the morphological and physiological characteristics of a control hybrid Solanum lycopersicum tomato variety from the same two parents in the absence of the trait for increased fruit firmness when grown under the same environmental conditions.

6. The method according to claim 5 , wherein the fruit from the donor tomato plant has increased fruit firmness at the harvesting stage as compared with the fruit from the control Solanum lycopersicum tomato plant.
