Abstract

There is disclosed compositions and methods relating to or derived from anti-PD-1 antibodies. More specifically, there is disclosed fully human antibodies that bind PD-1, PD-1-binding fragments and derivatives of such antibodies, and PD-1-binding polypeptides comprising such fragments. Further still, there is disclosed nucleic acids encoding such antibodies, antibody fragments and derivatives and polypeptides, cells comprising such polynucleotides, methods of making such antibodies, antibody fragments and derivatives and polypeptides, and methods of using such antibodies, antibody fragments and derivatives and polypeptides, including methods of treating or diagnosing subjects having PD-1 related disorders or conditions, including various inflammatory disorders and various cancers.

Claims (7)

1. A recombinant fully human anti-PD-1 antibody of an IgG class comprising a heavy chain variable domain comprising the amino acid sequence of SEQ ID NO. 21, and a light chain variable domain comprising the amino acid sequence of SEQ ID NO. 22.

2. An anti-PD-1 recombinant fully human antibody Fab fragment comprising a heavy chain variable domain comprising the amino acid sequence of SEQ ID NO. 21, and a light chain variable domain comprising the amino acid sequence of SEQ ID NO. 22.

An anti-PD-1 recombinant single chain human antibody comprising a heavy chain variable domain and a light chain variable domain which are connected by a peptide linker, wherein the heavy chain variable domain comprises the amino acid sequence of SEQ ID NO. 21, and the light chain variable domain comprises the amino acid sequence of SEQ ID NO. 22.

A method for treating a subject having cancer, the method comprising administering to the subject an effective amount of an anti-PD-1 polypeptide, wherein the anti-PD-1 polypeptide is selected from the group consisting of a recombinant fully human anti-PD-1 antibody of an IgG class, an anti-PD-1 recombinant fully human antibody Fab fragment, and an anti-PD-1 recombinant single chain human antibody; wherein the anti-PD-1 polypeptide comprises a heavy chain variable domain comprising the amino acid sequence of SEQ ID NO. 21, and a light chain variable domain comprising the amino acid sequence of SEQ ID NO. 22; and wherein the cancer is selected from the group consisting of ovarian cancer, colon cancer, breast cancer, lung cancer, myeloma, a neuroblastic-derived CNS tumor, monocytic leukemia, B-cell derived leukemia, T-cell derived leukemia, B-cell derived lymphoma, T-cell derived lymphoma, and a mast cell derived tumor.

5. The anti-PD-1 antibody of claim 1 , wherein the antibody is an IgG1 or an IgG4.

A pharmaceutical composition comprising the recombinant fully human anti-PD-1 antibody of an IgG class or an antigen-binding fragment thereof, the anti-PD-1 recombinant fully human antibody Fab fragment, or the anti-PD-1 recombinant single chain human antibody, of claim 3 , and a pharmaceutically acceptable excipient.

7. The method of claim 4 , wherein the administering to the subject comprises administering to the subject a pharmaceutical composition comprising the anti-PD-1 polypeptide and a pharmaceutically acceptable excipient.
