Abstract

The present disclosure relates to polymeric materials that may be labeled with a radioisotope, to processes for producing the labeled polymeric material, and to methods of using the materials in analytical and therapeutic applications. Specifically, the disclosure relates to injectable and implantable microparticles, such as microspheres, which are associated with radioisotopes such that the microparticles are both therapeutic and detectable. The radioisotope-containing microparticles are useful for embolization and other therapeutic medical applications.

Claims (32)

1. A method of preparing an embolic microsphere, comprising:

fabricating a polymeric microsphere comprising a polymerized monomer and a chelating agent, wherein fabricating comprises:

obtaining a monomer, wherein the monomer comprises at least one of an acrylate, acrylamide, acrylic, or monomer; and

polymerizing the monomer with a chelating agent comprising a polymerizable group, wherein the chelating agent comprises a compound of Formula I:

wherein n is between 1 and 18, inclusive;

Xa and Xb may independently be O, S or N; and

R may be alkyl or H,

wherein the polymerizable group of the chelating agent is polymerized with the monomer, wherein the chelating agent is configured to chelate a radioisotope, wherein the microsphere comprises between about 1% and about 20% by weight of chelating agent, wherein the microsphere comprises a diameter ranging from about 1 micrometer to about 2000 micrometers, and wherein the microsphere is suitable for embolization.

2. The method of claim 1 , wherein at least one of Xa and Xb is O.

3. The method of claim 1 , wherein the monomer comprises an acrylamide.

4. The method of claim 1 , wherein the monomer is selected from at least one of the following: N-[tris(hydroxymethyl)methyl]-acrylamide or sodium acrylate.

5. The method of claim 1 , wherein the monomer comprises N-[tris(hydroxymethyl)methyl]-acrylamide.

6. The method of claim 5 , wherein the chelating agent is incorporated into a polymer of the polymeric microsphere.

7. The method of claim 1 , wherein the method further comprises:

associating the microsphere with a radioisotope.

8. The method of claim 7 , wherein the radioisotope is both a β- and γemitter.

9. The method of claim 7 , wherein the radioisotope is selected from at least one of the following: 90 Y, 32 P, 99m Tc, 111 In, 67 Ga, 166 Dy, 201 Th, 131 I, 140 La, 153 Sm, 165 Dy, 166 Ho, 169 Er, 175 Yb, 177 Lu, 186 Re, or 188 Re.

10. The method of claim 7 , wherein the radioisotope is not leached from the microsphere to an extent greater than about 3% of its original level by weight over a period of three months.

11. A method of preparing an embolic microsphere, comprising:

fabricating a polymeric microsphere comprising a polymerized monomer and a chelating agent, wherein fabricating comprises:

obtaining a monomer, wherein the monomer comprises an acrylamide, and

polymerizing the monomer with a chelating agent comprising a polymerizable group, wherein the chelating agent comprises a compound of Formula I:

wherein n is between 1 and 18, inclusive;

Xa and Xb may independently be O, S or N; and

R may be alkyl or H,

wherein the polymerizable group of the chelating agent is polymerized with the monomer and incorporated into a polymer of the polymeric microsphere, wherein the chelating agent is configured to chelate a radioisotope, wherein the microsphere comprises between about 1% and about 20% by weight of chelating agent, and wherein the microsphere comprises a diameter ranging from about 1 micrometer to about 2000 micrometers.

12. The method of claim 11 , wherein at least one of Xa and Xb is O.

13. The method of claim 11 , wherein the method further comprises:

associating the microsphere with a radioisotope.

14. The method of claim 11 , wherein the radioisotope is selected from at least one of the following: 90 Y, 32 P, 99m Tc, 111 In, 67 Ga, 166 Dy, 201 Th, 131 I, 140 La, 153 Sm, 165 Dy, 166 Ho, 169 Er, 175 Yb, 177 Lu, 186 Re or 188 Re.

15. The method of claim 11 , wherein the microsphere is non-biodegradable.

16. The method of claim 1 , wherein the microsphere is non-biodegradable.
