Abstract

There is disclosed compositions and methods relating to or derived from anti-PD-L1 antibodies. More specifically, there is disclosed fully human antibodies that bind PD-L1, PD-L1-binding fragments and derivatives of such antibodies, and PD-L1-binding polypeptides comprising such fragments. Further still, there is disclosed nucleic acids encoding such antibodies, antibody fragments and derivatives and polypeptides, cells comprising such polynucleotides, methods of making such antibodies, antibody fragments and derivatives and polypeptides, and methods of using such antibodies, antibody fragments and derivatives and polypeptides, including methods of treating or diagnosing subjects having PD-L1 related disorders or conditions, including various inflammatory disorders and various cancers.

Claims (14)

1. A recombinant fully human anti-PD-L1 antibody comprising a heavy chain variable domain comprising an amino acid sequence of SEQ ID NO. 147 and a light chain variable domain comprising an amino acid sequence of SEQ ID NO. 148, wherein the antibody is an IgG1.

2. A fully human anti-PD-L1 antibody Fab fragment comprising a heavy chain variable domain comprising an amino acid sequence of SEQ ID NO. 147, and a light chain variable domain comprising an amino acid sequence of SEQ ID NO. 148.

3. A single chain human anti-PD-L1 antibody comprising a heavy chain variable domain, a light chain variable domain, and a peptide linker connecting the heavy chain variable domain and light chain variable domain, wherein the heavy chain variable domain comprises the amino acid sequence of SEQ ID NO. 147, and the light chain variable region comprises the amino acid sequence of SEQ ID NO. 148.

4. A method for treating a human subject having cancer, said method comprising the step of administering an effective amount of the recombinant fully human anti-PD-L1 antibody of claim 1 to the human subject.

The method of claim 4 , wherein the cancer is selected from the group consisting of ovarian cancer, colon cancer, breast cancer, lung cancer, myeloma, a neuroblastic-derived central nervous system tumor, a monocytic leukemia, a B-cell derived leukemia, a T-cell derived leukemia, a B-cell derived lymphoma, a T-cell derived lymphoma, skin cancer, small cell lung cancer, non-small cell lung cancer , head and neck cancer, pancreatic cancer, bladder cancer, bone cancer, cartilage cancer, liver cancer, lymph node cancer, nervous tissue cancer, skeletal muscle cancer, spinal cord cancer, spleen cancer, brain cancer, colorectal cancer, thyroid cancer, prostate cancer, vaginal cancer, kidney cancer, thymus cancer, thyroid cancer, stomach cancer, cancer of the urogenital tract, cancer of the ureter, cancer of the urethra, cancer of the uterus, cancer of the testes, and a mast cell derived tumor.

A recombinant fully human anti-PD-L1 antibody, or an antigen-binding fragment thereof, comprising a heavy chain variable domain comprising complementarity determining regions (CDRs) as set forth in an amino acid sequence of SEQ ID NO. 147; and comprising a light chain variable domain comprising CDRs as set forth in an amino acid sequence of SEQ ID NO. 148.

7. The anti-PD-L1 antibody, or antigen binding fragment thereof, of claim 6 , wherein the antibody is classified as an isotype selected from the group consisting of an IgG, an IgM, IgD, IgA, and IgE.

8. The anti-PD-L1 antigen binding fragment of claim 6 , wherein the fragment is selected from the group consisting of a Fab fragment, a Fab′, a F(ab′)2, an Fv, a domain antibody, and a single-chain antibody.

9. A pharmaceutical composition comprising the anti-PD-L1 antibody, or antigen-binding fragment thereof, of claim 6 , and a pharmaceutically acceptable excipient.

10. A method of treating a human subject having cancer comprising the step of administering an effective amount of the anti-PD-L1antibody, or antigen binding fragment thereof, of claim 6 .

The method of claim 10 , wherein the cancer is selected from the group consisting of ovarian cancer, colon cancer, breast cancer, lung cancer, myeloma, a neuroblastic-derived central nervous system tumor, a monocytic leukemia, a B-cell derived leukemia, a T-cell derived leukemia, a B-cell derived lymphoma, a T-cell derived lymphoma, skin cancer, small cell lung cancer, non-small cell lung cancer, head and neck cancer, pancreatic cancer, bladder cancer, bone cancer, cartilage cancer, liver cancer, lymph node cancer, nervous tissue cancer, skeletal muscle cancer, spinal cord cancer, spleen cancer, brain cancer, colorectal cancer, thyroid cancer, prostate cancer, vaginal cancer, kidney cancer, thymus cancer, thyroid cancer, stomach cancer, cancer of the urogenital tract, cancer of the ureter, cancer of the urethra, cancer of the uterus, cancer of the testes, and a mast cell derived tumor.

12. A method for treating a human subject having cancer, said method comprising the step of administering an effective amount of the fully human anti-PD-L1 antibody Fab fragment of claim 2 to the human subject.

13. A method for treating a human subject having cancer, said method comprising the step of administering an effective amount of the single chain human antibody of claim 3 to the human subject.

14. The anti-PD-L1 antibody of claim 7 , wherein the antibody is an IgG4.
