Abstract

The present invention is directed to compositions that include programmed cell death 1 (PD-1) binding domains and antibodies that include such PD-1 binding domains. Also provided are nucleic acid compositions that encode the binding domains and antibodies, expression vector compositions that include the nucleic acids, and host cells that include the expression vector compositions.

Claims (48)

1. A composition comprising a Programmed Cell Death 1 (PD-1) binding domain, the PD-1 binding domain comprising:

a) a light chain variable domain comprising a vhCDR1 having SEQ ID NO: 37778; a vhCDR2 having SEQ ID NO: 37779, and a vhCDR3 having SEQ ID NO: 37780; and

b) a heavy chain variable domain comprising a vlCDR1 having SEQ ID NO: 37782, a vlCDR2 having SEQ ID NO: 37783, and a vlCDR3 having SEQ ID NO: 37784.

2. The composition of claim 1 , wherein the light chain variable domain has the sequence of SEQ ID NO: 37777 and the heavy chain variable domain has the sequence of SEQ ID NO: 37781.

3. The composition of claim 1 , wherein the PD-1 binding domain is a single chain variable fragment (scFv).

4. The composition of claim 3 , wherein the scFv has the amino acid sequence of SEQ ID NO: 37785.

5. An antibody comprising a PD-1 binding domain, the PD-1 binding domain comprising:

a) a light chain variable domain comprising a vhCDR1 having SEQ ID NO: 37778; a vhCDR2 having SEQ ID NO: 37779, and a vhCDR3 having SEQ ID NO: 37780; and

b) a heavy chain variable domain comprising a vlCDR1 having SEQ ID NO: 37782, a vlCDR2 having SEQ ID NO: 37783, and a vlCDR3 having SEQ ID NO: 37784.

6. The antibody of claim 5 , wherein the PD-1 binding domain is an scFv.

7. A bispecific antibody comprising:

a) a first monomer comprising an anti-PD-1 scFv-linker-CH2-CH3, wherein CH2-CH3 is a first IgG Fc domain and the anti-PD1 scFv comprises a first heavy chain variable domain having the amino acid sequence of SEQ ID NO: 37781, a first light chain variable domain having the amino acid sequence of SEQ ID NO: 37777, and a scFv linker that attaches the first variable heavy chain with the first variable light chain;

b) a second monomer comprising a VH-CH1-hinge-CH2-CH3, wherein VH is a second heavy chain variable domain and CH2-CH3 is a second IgG Fc domain; and

c) a light chain comprising a second light chain variable domain, wherein the second heavy chain variable domain and second light chain variable domain form an antigen binding domain.

8. The antibody of claim 7 , wherein the anti-PD-1 scFv has the amino acid sequence of SEQ ID NO: 37785.

9. An antibody comprising:

a) a first monomer comprising a VH1-CH1-linker 1-VL2-scFv linker-VH2-linker 2-CH2-CH3,

wherein VL2-scFv linker-VH2 is an anti-PD1 scFV, linker 1 and linker 2 are a first linker and second linker, respectively, and CH2-CH3 is a first IgG Fc domain;

b) a second monomer comprising a VH1-CH1-hinge-CH2-CH3, wherein VH1 is a first variable heavy domain and CH2-CH3 is a second IgG Fc domain;

c) a common light chain comprising a first variable light domain (VL1); and

wherein the first variable heavy domain on the first monomer and the first variable light domain form a first binding domain, and the first variable heavy domain on the second monomer and the first variable light domain form a second binding domain, and

wherein the VH2 is a second variable heavy domain having the amino acid sequence of SEQ ID NO: 37781 and the VL2 is a second variable light domain having the amino acid sequence of SEQ ID NO: 37777.

10. The heterodimeric antibody of claim 9 , wherein the anti-PD-1 scFv has the amino acid sequence of SEQ ID NO: 37785.

11. A nucleic acid composition comprising:

a) a first nucleic acid encoding the heavy chain variable domain of claim 1 ; and

b) a second nucleic acid encoding the light chain variable domain of claim 1 .

12. An expression vector composition comprising:

a) a first expression vector comprising the first nucleic acid of claim 11 ; and

b) a second expression vector comprising the second nucleic acid of claim 11 .

13. A host cell comprising the expression vector composition of claim 12 .

14. A nucleic acid composition comprising:

a) a first nucleic acid encoding the first monomer of claim 7 ;

b) a second nucleic acid encoding the second monomer of claim 7 ; and

c) a third nucleic acid encoding the light chain of claim 7 .

15. An expression vector composition comprising:

a) a first expression vector comprising the first nucleic acid of claim 14 ;

b) a second expression vector comprising the second nucleic acid of claim 14 ; and

c) a third expression vector comprising the third nucleic acid of claim 14 .

16. A host cell comprising the expression vector composition of claim 15 .

17. A nucleic acid composition comprising:

a) a first nucleic acid encoding the first monomer of claim 9 ;

b) a second nucleic acid encoding the second monomer of claim 9 ; and

c) a third nucleic acid encoding the common light chain of claim 9 .

18. An expression vector composition comprising:

a) a first expression vector comprising the first nucleic acid of claim 17 ;

b) a second expression vector comprising the second nucleic acid of claim 17 ; and

c) a third expression vector comprising the third nucleic acid of claim 17 .

19. A host cell comprising the expression vector composition of claim 18 .
