Abstract

A system for monitoring impedance of excitable cells in vitro, which includes a device for monitoring cell-substrate impedance at 20 millisecond resolution, which includes a nonconductive substrate with one or more electrode arrays fabricated in one or more wells, wherein cell attachment on the substrate can result in a detectable change in impedance between electrodes within each electrode array; an impedance analyzer capable of impedance measurement at 20 millisecond time resolution; electronic circuitry that can engage said device and selectively connect said two or more electrode arrays of said device to said impedance analyzer; and a software program that controls said electronic circuitry and records and analyzes data obtained from said impedance analyzer.

Claims (20)

1. A system for monitoring impedance of excitable cells in vitro, comprising:

a) a device for monitoring cell-substrate impedance at 20 millisecond resolution, the device comprising

i) a nonconductive substrate;

ii) one or more electrode arrays fabricated on said substrate;

iii) one or more wells on said substrate, wherein each of said one or more arrays is associated with one of said one or more wells; wherein each electrode array is individually addressable, further wherein said substrate has a surface suitable for cell attachment, wherein said cell attachment on said substrate can result in a detectable change in impedance between electrodes within each electrode array;

b) an impedance analyzer capable of impedance measurement at 20 millisecond time resolution;

c) electronic circuitry that can engage said device and selectively connect said two or more electrode arrays of said device to said impedance analyzer; and

d) a software program that controls said electronic circuitry and records and analyzes data obtained from said impedance analyzer.

2. The system according to claim 1 , wherein each of said electrode arrays comprises two electrode structures having substantially a same surface area.

3. The system according to claim 2 , wherein said electrode structures are each constructed from a material selected from the group consisting of metal, gold, platinum, titanium and chromium.

4. The system according to claim 2 , wherein said electrode structures have a width between 20 microns and 500 microns.

5. The system according to claim 2 , wherein a gap between electrode structures within each electrode array is between 5 microns and 100 microns.

6. The system according to claim 1 , wherein each electrode array is organized in a configuration selected from the group consisting of interdigitated, circle-on-line, diamond-on-line, concentric, sinusoidal, and castellated.

7. The system according to claim 1 , wherein said 20 millisecond time resolution allows for impedance measurements at more than two time points within each excitation cycle of excitable cells introduced to wells.

8. The system according to claim 7 , wherein said excitation cycle is a cell beating cycle.

9. The system according to claim 8 , wherein said excitable cells are cardiomyocytes.

10. The system according to claim 7 , wherein said excitable cells undergo excitation-contraction coupling.

11. The system according to claim 1 , wherein said impedance measurement at 20 millisecond time resolution comprises at least two consecutive impedance measurements 20 milliseconds or less apart.

12. The system according to claim 11 , wherein said at least two consecutive impedance measurements are less than 10 milliseconds apart.

13. The system according to claim 1 , wherein said one or more wells is selected from the group consisting of 6 wells, 8 wells, 16 wells, 32 wells, 96 wells and 384 wells.
