Abstract

The present disclosure provides engineered Vibrio sp. organisms. The organisms can be engineered to have an altered Chromosome II so that the altered Chromosome II can be used in Vibrio sp. and other organisms for the cloning or amplification of nucleic acid molecules and for the expression and production of proteins and peptides in a Vibrio sp. organism. One or more genetic elements have been deleted from Chromosome II and/or relocated from Chromosome II to Chromosome I. The engineered Vibrio sp. organisms of the invention can also have signal sequences fused to proteins or peptides to be secreted from the cell. In some embodiments the engineered Vibrio sp. organisms can have sequences that enable them to retain viability after incubation at low temperatures.

Claims (14)

1. A cold tolerant Vibrio natriegens organism comprising an exogenous sequence encoding one or more enzymes from a reactive oxygen species detoxification system and able to generate new colonies after being stored at a temperature of 4° C. or less for at least 14 days.

2. The organism of claim 1 wherein the organism comprises a heterologous nucleotide sequence encoding any one or more of:

a) one or more alkyl hydroperoxide reductase gene(s) under the control of an exogenous promoter and operable in Vibrio natriegens , or

b) a catalase gene under the control of an exogenous promoter and operable in Vibrio natriegens , or

c) a glutathione S-transferase gene under the control of an exogenous promoter and operable in Vibrio natriegens.

3. The organism of claim 2 wherein the heterologous nucleotide sequence encodes an alkyl hydroperoxide reductase operon under the control of an exogenous promoter and operable in Vibrio natriegens.

4. The organism of claim 3 wherein the alkyl hydroperoxide reductase operon is the alkyl hydroperoxide reductase operon from E. coli (ahpCF).

5. The organism of claim 2 wherein the heterologous nucleotide sequence encodes a catalase gene under the control of an exogenous promoter and operable in Vibrio natriegens.

6. The organism of claim 3 wherein the catalase gene is katG or katE under the control of a heterologous promoter and operable in Vibrio natriegens.

7. The organism of claim 2 wherein the heterologous nucleotide sequence encodes a glutathione S-transferase gene under the control of an exogenous promoter and operable in Vibrio natriegens.

8. The organism of claim 2 wherein the any one or more of alkyl hydroperoxide reductase operon, the catalase gene, and the glutathione S-transferase gene are comprised on ChI or ChII.

9. The organism of claim 2 further comprising an exogenous vector, and wherein the alkyl hydroperoxide reductase operon, or the catalase gene, or the glutathione S-transferase gene are comprised on the vector.

10. The organism of claim 2 comprised on a solid media.

11. The organism of claim 2 wherein the heterologous nucleotide sequence encoding any one of a), b), or c) enables the organism to generate new colonies after storage at about 4° C. for at least 19 days.
