Abstract

A method of making a nanocomposite includes forming at least one gold nanorod; coating a silver layer on an outer surface of the gold nanorod; assembling a Raman reporter molecule layer on the coated silver layer; coating a pegylated layer on the assembled Raman reporter molecule layer; and conjugating the coated pegylated layer with an active layer, the active layer comprising at least one of a targeting molecule configured to bind to the target of interest and a functional molecule configured to interact with the target of interest.

Claims (39)

1. A nanocomposite, comprising:

a nanostructure formed by at least one nanomaterial; and

an active layer conjugated to the nanostructure, and comprising at least one of:

a targeting molecule configured to bind to a target of interest; and

a functional molecule configured to interact with the target of interest,

wherein the active layer comprises a biomarker of traumatic brain injury (TBI) or an antibody that specifically binds to the biomarker, wherein the biomarker of TBI comprises at least one of S100 calcium-binding protein B (S100B), neuron-specific enolase (NSE), myelin basic protein (MBP) caspase-3, interleukins, tau protein, neurofilament light polypeptide (NEFL), neurofilament heavy polypeptide (NEFH), and glial fibrillary acidic protein.

2. The nanocomposite of claim 1 , wherein the nanostructure has a spherical shape, a tubular shape, a cylindrical shape, or a rod-like triangular shape.

3. The nanocomposite of claim 1 , wherein the nanomaterial comprises at least one of silver coated gold rods, quantum dots, nanowires, nanotubes, nanofibers, and fullerenes.

4. The nanocomposite of claim 1 , wherein the nanomaterial comprises gold, silver, gold/silver, copper, iron, iron oxide, TiO 2 , SiO 2 , and carbon.

The nanocomposite of claim 1 , wherein the target of interest is at least one of a cancer cell and a pathogen, and the functional molecule is a virus, a phage, a drug, a growth factor, antibiotics, a gene, a plasmid, a vaccine, or an antibody that specifically binds to S100 calcium-binding protein B (S100B).

6. The nanocomposite of claim 5 , wherein the target of interest is the cancer cell, and the functional molecule is the virus capable of reproducing in the cancer cell.

7. The nanocomposite of claim 1 , wherein the nanostructure comprises:

at least one gold nanorod; and

a silver layer surrounding the at least one gold nanorod, the silver layer comprising silver nanoparticles.

8. The nanocomposite of claim 7 ,

wherein the gold nanorod has an aspect ratio (AR) in a range of about 1-9, a length in a range of about 10-100 nm, and a diameter in a range of about 1-40 nm; and

wherein the silver layer has a thickness in a range of about 0.5-5 nm.

The nanocomposite of claim 1 , further comprising a reporter layer disposed between the nanomaterial and the active layer, wherein the reporter layer is detectable by at least one of surface enhanced Raman spectroscopy (SERS), magnetic resonance imaging (MRI), x-ray radiography, computed tomography (CT), and infrared spectroscopy (IR).

10. The nanocomposite of claim 9 , wherein the reporter layer comprises 4-mercaptobenzoic acid (4MBA), p-aminothiophenol (PATP), p-nitrothiophenol (PNTP), 4-(methylsulfanyl) thiophenol (4MSTP), or other molecules with unique Raman spectra and intense Raman peak intensities.

11. The nanocomposite of claim 9 , further comprising a pegylated layer disposed between the reporter layer and the active layer, wherein the pegylated layer comprises at least one of thiolated polyethylene glycol (HS-PEG), thiolated polyethylene glycol acid (HS-PEG-COOH) and HS-PEG-NHx.

The nanocomposite of claim 1 , wherein the targeting molecule is molecule of anti-epithelial cell adhesion molecule antibody (anti-EpCAM), anti-CD44 antibody, anti-insulin-like growth factor 1 receptor antibody (anti-IGF-1), anti-Keratin 18 antibody, or one or more antibodies specific to the target of interest.

13. The nanocomposite of claim 12 , wherein the functional molecule is conjugated to at least one of the pegylated layer and the targeting molecule.

14. A nanocomposite, comprising:

a nanostructure formed by at least one nanomaterial, wherein the at least one nanomaterial comprises at least one of quantum dots, carbon nanotubes, and fullerenes; and

an active layer conjugated to the nanostructure, and comprising:

a targeting molecule configured to bind to a target of interest; and

a functional molecule configured to interact with the target of interest,

wherein the target of interest is a cancer cell, and the functional molecule is a virus capable of reproducing in the cancer cell.

15. The nanocomposite of claim 14 , wherein the nanostructure has a spherical shape, a tubular shape, a cylindrical shape, or a rod-like triangular shape.

The nanocomposite of claim 14 , further comprising a reporter layer disposed between the nanomaterial and the active layer, wherein the reporter layer is detectable by at least one of surface enhanced Raman spectroscopy (SERS), magnetic resonance imaging (MRI), x-ray radiography, computed tomography (CT), and infrared spectroscopy (IR).

17. A nanocomposite, comprising:

a nanostructure formed by at least one nanomaterial; and

an active layer conjugated to the nanostructure, and comprising at least one of:

a targeting molecule configured to bind to a target of interest; and

a functional molecule configured to interact with the target of interest,

wherein the target of interest is a plant, and the functional molecule is a growth factor capable of promoting growth of the plant.

18. The nanocomposite of claim 17 , wherein the nanostructure has a spherical shape, a tubular shape, a cylindrical shape, or a rod-like triangular shape.

19. The nanocomposite of claim 17 , wherein the nanomaterial comprises at least one of silver coated gold rods, quantum dots, nanowires, nanotubes, nanofibers, and fullerenes.

20. The nanocomposite of claim 17 , further comprising a reporter layer disposed between the nanomaterial and the active layer, wherein the reporter layer is detectable by at least one of surface enhanced Raman spectroscopy (SERS), magnetic resonance imaging (MRI), x-ray radiography, computed tomography (CT), and infrared spectroscopy (IR).
