Abstract

A method for magnetic cellular manipulation may include contacting a composition with a biological sample to form a mixture. The composition may include a plurality of particles. Each particle in the plurality of particles may include a magnetic substrate. The magnetic substrate may be characterized by a magnetic susceptibility greater than zero. The composition may also include a chargeable silicon-containing compound. The chargeable silicon-containing compound may coat at least a portion of the magnetic substrate. The biological sample may include cells and/or cellular structures. The method may also include applying a magnetic field to the mixture to manipulate the composition.

Claims (11)

1. A method of separating viable sperm from dead or damaged sperm comprising:

diluting a semen sample comprising viable sperm, and dead or damaged sperm, with a buffered media;

adding magnetic particles to the diluted semen sample to form an admixture, wherein each magnetic particle is no greater than 1000 nm and coated with a chargeable silicon-containing compound and a chargeable silicon-containing compound;

binding a portion of the dead or damaged sperm to the magnetic particles in the diluted admixture through an electrical charge interaction; and

magnetically separating the bound dead or damaged sperm from unbound viable sperm.

2. The method of claim 1 , wherein the magnetic particles comprise Fe 3 O 4 .

3. The method of claim 1 , wherein the magnetic particles comprise 2-(carbomethoxy)ethyltrimethoxysilane.

4. The method of claim 1 , wherein each magnetic particle is between 30 nm to 1000 nm in size.

5. The method of claim 1 , further comprising the step of centrifuging the semen sample prior to the step of adding the magnetic particles.

6. The method of claim 5 , further comprising the step of decanting the centrifuged semen sample.

7. The method of claim 6 , further comprising the step of resuspending the centrifuged semen sample in buffered media.
