Abstract

A method has been developed of preventing or limiting formation of adhesions by administering to a site in need thereof, in the absence of or after bleeding or leakage of fluid has been substantially stopped, a self-assembling material which forms a barrier to formation of adhesions. In certain embodiments, the self assembling materials are peptidomimetics, nucleotidomimetics, di- and triblock copolymers, N-alkylacrylamides, or dendrimers. These materials are also useful in a method for regeneration or repair of tissue or cells forming tissue.

Claims (18)

1. A method of limiting formation of adhesions or scars, comprising

administering to a region of a subject's body at risk of forming adhesions, a self-assembling peptidomimetic formulation in an amount effective to form a barrier structure to limit the formation of adhesions or scars in the region,

wherein the self-assembling formulation comprises peptidomimetics selected from the group consisting of α-peptides, β-peptides, γ-peptides, δ-peptides, and co-polymers thereof,

wherein the α-peptide peptidomimetics are selected from the group consisting of N,N′ linked oligoureas, oligopyrrolinones, oxazolidin-2-ones, azatides and azapeptides.

2. The method of claim 1 , wherein the self-assembling peptidomimetic formulation comprises a copolymer of α-peptides and one or more of β-peptides, γ-peptides, or δ-peptides.

3. The method of claim 1 , further comprising providing one or more therapeutic agents, prophylactic agents, diagnostic agents, or cells.

4. The method of claim 1 , wherein the self-assembling peptidomimetic formulation comprises a pharmaceutically acceptable carrier or matrix or support material for administration onto or into the body.

5. The method of claim 4 , wherein the self-assembling peptidomimetic formulation is administered as a dry powder, liquid, or incorporated into hydrogels, or sponges.

6. The method of claim 1 comprising applying the self-assembling peptidomimetic formulation to a burn.

7. The method of claim 1 comprising applying the self-assembling peptidomimetic formulation to a wound or surgery site or a site of inflammation.

8. The method of claim 1 , wherein the self-assembling peptidomimetic formulation is administered as a coating on a medical device or implant.

The method of claim 1 , wherein the self-assembling peptidomimetic formulation is administered to the site of a surgical procedure at a time point selected from group consisting of before the initiation of the surgical procedure, during the surgical procedure, after commencement of the surgical procedure, and combinations thereof.

10. The method of claim 1 , wherein the self-assembling peptidomimetic formulation has a concentration of Li+, Na+, K+, and Cs+ ions less than 50 mM.

11. The method of claim 1 , wherein the self-assembling peptidomimetic formulation is administered in the form of a solution.

12. The method of claim 11 , wherein the concentration of self-assembling peptidomimetics in solution is between 1.0% and 10.0%, inclusive.

13. The method of claim 11 , wherein the concentration of self-assembling peptidomimetics is between 2.0% and 3.0%, inclusive.

14. The method of claim 11 , further comprising the step of forming the solution by dissolving a powder of the self-assembling peptidomimetics with water or oil.

15. The method of claim 1 , wherein the self-assembling peptidomimetic formulation is administered to damaged skin or tissue that is bleeding to form a self-assembled barrier structure which is maintained in contact with the tissue following the cessation of bleeding.
