COMPOSITIONS AND METHODS FOR TARGETING CANCER STEM CELLS
The present invention provides methods of testing anti-STn antibodies for their ability target cancer stem cells by isolating cancer stem cells from cultures, contacting them with anti-STn antibodies being tested, and measuring resulting cell viability.
1 . A method of testing an anti-sialyl Tn antigen (STn) antibody for the ability to target cancer stem cells (CSCs) comprising:
(a) culturing one or more cell lines,
(b) isolating one or more sub-fractions, wherein said one or more sub-fractions express at least one CSC marker,
(c) contacting said one or more sub-fractions with said anti-STn antibody, and
(d) measuring cell viability in said one or more sub-fractions, wherein decreased cell viability in the one or more sub-fractions indicates the ability of the anti-STn antibody to target CSCs.
2 . The method of claim 1 , wherein said one or more cell lines comprise one or more ovarian cancer cell lines.
3 . The method of claim 2 , wherein said one or more ovarian cancer cell lines are selected from the group consisting of A2780, SKOV3, OVCAR3 and OV90.
4 . The method of claim 1 , wherein said one or more cell lines are derived from breast, ovarian, pancreatic, bladder, cervical, colon and/or lung tissue.
5 . The method of claim 1 , wherein said CSCs comprise CD133 and/or CD44 biomarkers.
6 . The method of claim 1 , wherein said anti-STn antibody further comprises at least one conjugate.
7 . The method of claim 6 , wherein said at least one conjugate comprises at least one chemotherapeutic agent.
8 . The method of claim 7 , wherein said at least one chemotherapeutic agent is selected from the group consisting of paclitaxel and carboplatin.
9 . The method of claim 1 , wherein said anti-STn antibody is tested for the ability to induce antibody-dependent cell-mediated cytotoxicity (ADCC) and/or inhibit the cell cycle.
10 . The method of claim 1 , wherein said anti-STn antibody comprises an antibody-drug conjugate (ADC).
11 . The method of claim 10 , wherein said ADC comprises one or more therapeutic compounds or one or more cytotoxic agents.
12 . The method of claim 11 , wherein said one or more therapeutic compounds or one or more cytotoxic agents are conjugated to said anti-STn antibody directly or via a linker.
13 . The method of claim 12 , wherein said antibody-drug conjugate comprises one or more cytotoxic agents and wherein said one or more cytotoxic agents are cytoskeletal inhibitors or DNA damaging agents.
14 . The method of claim 13 , wherein said antibody-drug conjugate comprises one or more cytotoxic agents selected from the group consisting of monomethyl auristatin E (MMAE) and monomethyl auristatin F (MMAF).
15 . The method of claim 1 , wherein said anti-STn antibody comprises the heavy chain amino acid sequence of SEQ ID NO: 5 and the light chain amino acid sequence of SEQ ID NO: 6.
16 . A method of reducing CSC viability comprising:
(a) selecting an anti-STn antibody, wherein the anti-STn antibody is capable of reducing CSC viability, the method of selecting comprising:
(i) culturing one or more cell lines,
(ii) isolating one or more sub-fractions of the one or more cell lines, wherein the one or more sub-fractions express at least one CSC marker,
(iii) contacting the one or more sub-fractions with the anti-STn antibody,
(iv) measuring cell viability in said one or more sub-fractions, and
(v) selecting the anti-STn antibody if cell viability is reduced, and
(b) using the anti-STn antibody to reduce CSC viability.
17 . The method of claim 16 , wherein said anti-STn antibody targets N-acetylneuraminic sialyl Tn antigen (AcSTn) and/or N-glycolylneuraminic sialyl Tn antigen (GcSTn).
18 . The method of claim 17 , wherein said anti-STn antibody comprises the heavy chain amino acid sequence of SEQ ID NO: 5 and the light chain amino acid sequence of SEQ ID NO: 6.
19 . A method of targeting CSCs in a subject comprising:
(a) selecting an anti-STn antibody, wherein the anti-STn antibody is capable of reducing CSC viability, the method of selecting comprising:
(i) culturing one or more cell lines,
(ii) isolating one or more sub-fractions of the one or more cell lines, wherein the one or more sub-fractions express at least one CSC marker,
(iii) contacting the one or more sub-fractions with the anti-STn antibody,
(iv) measuring cell viability in said one or more sub-fractions, and
(v) selecting the anti-STn antibody if cell viability is reduced, and
(b) using the anti-STn antibody to target CSCs in the subject.
20 . The method of claim 19 , wherein the anti-STn antibody targets AcSTn and/or GcSTn.
21 . The method of claim 20 , wherein said anti-STn antibody comprises the heavy chain amino acid sequence of SEQ ID NO: 5 and the light chain amino acid sequence of SEQ ID NO: 6.
