Heterobicyclic N-aminophenyl-amides as inhibitors of histone deacetylase
This invention provides compounds that are inhibitors of HDAC2. The compounds (e.g., compounds according to Formula (I) or any one of Compounds 100-175) accordingly are useful for treating, alleviating, or preventing a condition in a subject such as a neurological disorder, memory or cognitive function disorder or impairment, extinction learning disorder, fungal disease or infection, inflammatory disease, hematological disease, or neoplastic disease, or for improving memory or treating, alleviating, or preventing memory loss or impairment.
1. A compound of Formula I:
or a pharmaceutically acceptable salt thereof, wherein:
R 1 is an optionally substituted, bicyclic fused, spirofused or bridged, non-aromatic heterocyclyl, comprising at least two heteroatoms independently selected from N, O and S, wherein each ring of R 1 comprises at least one of the heteroatoms; and
R 2 is selected from heteroaryl and para-substituted aryl, wherein R 2 is optionally further substituted, and R 2 is other than optionally substituted biphenyl and optionally substituted thiophenyl;

6. The compound of claim 5 , wherein R 2 is selected from para-substituted phenyl, pyridinyl, furanyl, 1H-Pyrazolyl, thiazolyl, isoxazolyl, oxazolyl, isothiazolyl, 5,6-dihydro-4H-Pyrrolol[1,2-b]pyrazolyl, pyridazinyl, and pyrimidinyl, and wherein R 2 is optionally additionally substituted.

8. The compound of claim 7 , wherein R 2 is selected from 4-fluorophenyl, 4-methoxyphenyl, 4-chlorophenyl, 4-difluromethoxyphenyl, 4-cyanophenyl, 2,4-difluorophenyl, 3,4-difluorophenyl, pyridin-3-yl, pyridin-4-yl, furan-2-yl, furan-3-yl, and 1 -methyl- 1H-Pyrazol-4-yl.
9. The compound of claim 8 , wherein R 2 is selected from 4-fluorophenyl, 4-methoxyphenyl, 4-difluromethoxyphenyl, pyridin-4-yl, furan-2-yl, and furan-3-yl.
10. The compound of claim 1 , wherein the compound is selected from:
or a pharmaceutically acceptable salt thereof.
11. A pharmaceutical composition comprising a compound of claim 1 or a pharmaceutically acceptable salt thereof; and a pharmaceutically acceptable carrier.
12. A method of treating a condition in a subject selected from
a. a cognitive function disorder or impairment associated with Alzheimer's disease, Huntington's disease, seizure induced memory loss, schizophrenia, Rubinstein Taybi syndrome, Rett Syndrome, Fragile X, Lewy body dementia, vascular dementia, frontotemporal dementia, ADHD, dyslexia, bipolar disorder and social, cognitive and learning disorders associated with autism, traumatic head injury, attention deficit disorder, anxiety disorder, conditioned fear response, panic disorder, obsessive compulsive disorder, posttraumatic stress disorder (PTSD), phobia, social anxiety disorder, substance dependence recovery, Age Associated Memory Impairment (AAMI), Age Related Cognitive Decline (ARCD), ataxia, or Parkinson's disease;
b. a hematological disease selected from acute myeloid leukemia, acute promyelocytic leukemia, acute lymphoblastic leukemia, chronic myelogenous leukemia, myelodysplastic syndromes, and sickle cell anemia;
c. a neoplastic disease; and
d. an extinction learning disorder selected from fear extinction and post-traumatic stress disorder, comprising administering to the subject in need thereof an effective amount of a compound of claim 1 , or a pharmaceutically acceptable salt thereof.
13. The method of claim 12 , wherein the condition is Alzheimer's disease, Huntington's disease, frontotemporal dementia, Freidreich's ataxia, post-traumatic stress disorder (PTSD), Parkinson's disease, or substance dependence recovery.
