Multi-targeted single entity conjugates
The present invention relates, in general to, compounds, compositions and methods useful for modulating gene expression of multiple target nucleic acids by a single chemical entity.
1. A multi-targeted molecule comprising a first double-stranded siRNA molecule and a second double-stranded siRNA molecule, wherein at least one ligand is conjugated with the multi-targeted molecule, and wherein the first and second siRNAs do not overlap with each other, and wherein:
(i) the first and second siRNAs are connected together by a linker comprising the nucleotide sequence UUU;
(ii) the sense strand of the first siRNA comprises a single-strand overhang at its 3′-end and the antisense strand of the second siRNA comprises a single-strand overhang at its 3′-end, wherein nucleic acid sequence of the single-strand overhang of the sense strand is substantially complementary to the nucleotide sequence of the single-strand overhang of the antisense strand, and wherein the two single-strand overhangs form a duplex; or
(iii) the antisense strand of the first siRNA comprises a single-strand overhang at its 3′-end and the antisense strand of the second siRNA comprises a single-strand overhang at its 3′-end, wherein nucleic acid sequence of the single-strand overhang of the antisense strand is substantially complementary to the nucleotide sequence of the single-strand overhang of the antisense strand, and wherein the two single-strand overhangs form a duplex.
2. The multi-targeted molecule of claim 1 , wherein the first and second siRNA are each conjugated with at least one ligand.
3. The multi-targeted molecule of claim 1 , wherein said multi-targeted molecule inhibits gene expression of at least two target nucleic acids by at least 75% each relative to when the first and second siRNA are not connected together.
4. The multi-targeted molecule of claim 1 , wherein the sense strand of the siRNAs comprises one block of two phosphorothioate internucleotide linkages and the antisense strand of the siRNAs comprises two blocks of two phosphorothioate internucleotide linkages.
5. The multi-targeted molecule of claim 1 , wherein at least one of the first and second siRNA comprises a 3′-F modification at 5′-terminal of at least one strand.
6. The multi-targeted molecule of claim 1 , wherein 3′-end of a sense strand of one siRNA is covalently linked to the 3′-end of an antisense strand of the other siRNA.
7. The multi-targeted molecule of claim 1 , wherein 5′-end of a sense strand of one siRNA is covalently linked to the 5′-end of an antisense strand of the other siRNA.
8. The multi-targeted molecule of claim 1 , wherein 3′-end of a sense strand of one siRNA is covalently linked to the 5′-end of an antisense strand of the other siRNA.
9. The multi-targeted molecule of claim 1 , wherein 5′-end of a sense strand of one siRNA is covalently linked to the 3′-end of an antisense strand of the other siRNA.
10. The multi-targeted molecule of claim 1 , wherein the length of the substantially complementary sequence is from 5 to 25 nucleotides.
11. The multi-targeted molecule of claim 1 , wherein the first and second siRNA are each conjugated with at least one ligand.
12. The multi-targeted molecule of claim 1 , wherein less than 5% of the duplex formed by the two single-strand overhangs is cleaved by a nuclease.
13. The multi-targeted molecule of claim 1 , wherein at least 50% of the duplex formed by the two single-strand overhangs is cleaved by a nuclease.
