Abstract

The present invention relates to RNAi agents, e.g., double stranded RNAi agents, targeting the programmed cell death 1 ligand 1 (PD-L1) gene, and methods of using such RNAi agents to inhibit expression of a PD-L1 gene and methods of treating subjects having a PD-L1-associated disorder.

Claims (31)

1. A double stranded ribonucleic acid (dsRNAi) agent for inhibiting expression of programmed cell death 1 ligand 1 (PD-L1), wherein said dsRNAi agent comprises a sense strand and an antisense strand, wherein said sense strand comprises at least 15 contiguous nucleotides differing by no more than 3 nucleotides from any one of nucleotides 3222-3243 of the nucleotide sequence of SEQ ID NO:1 and said antisense strand comprises at least 15 contiguous nucleotides differing by no more than 3 nucleotides from the complementary portion of the nucleotide sequence of SEQ ID NO:2,

wherein substantially all of the nucleotides of said sense strand and substantially all of the nucleotides of said antisense strand comprise nucleotide modifications, and

wherein said sense strand is conjugated to an N-acetylgalactosamine (GalNAc) derivative attached at the 3′-terminus.

2. The dsRNAi agent of claim 1 , wherein

(a) the sense strand comprises 5′-ACCUGCAUUAAUUUAAUAAAA-3′ (SEQ ID NO:116) and the antisense strand comprises 5′—UUUUAUUAAAUUAAUGCAGGUAC-3′ (SEQ ID NO:117); or

(b) the sense strand comprises 5′—UACCUGCAUUAAUUUAAUAAA-3′ (SEQ IDNO:110) and said antisense strand comprises 5′—UUUAUUAAAUUAAUGCAGGUACA-3′ (SEQ ID NO:111).

3. The dsRNAi agent of claim 1 , wherein:

(b) the sense strand comprises 5′-ascscugcAfuUfAfAfuuuaauaaaaL96-3′ (SEQ IDNO:220) and said antisense strand comprises 5′—UfsUfsuuaUfuAfAfauuaAfuGfcaggusasc-3′ (SEQ ID NO:221);

(c) the sense strand comprises 5′-usasccugCfaUfUfAfauuuaauaaaL96-3′ (SEQ IDNO:212) and said antisense strand comprises 5′-usUfsuauUfaAfAfuuaaUfgCfagguascsa-3′ (SEQ ID NO:213); or

(d) the sense strand comprises 5′-usasccugCfaUfUfAfauuuaauaaaL96-3′ (SEQ IDNO:214) and said antisense strand comprises 5′—UfsUfsuauUfaAfAfuuaaUfgCfagguascsa-3′ (SEQ ID NO:215);

wherein a, c, g, and u are 2′-O-methyl (2′-OMe) A, 2′-OMe C, 2′-OMe G, and 2′-OMe U, respectively; Af, Cf, Gf, and Uf are 2′-fluoro A, 2′-fluoro C, 2′-fluoro G, and 2′-fluoro U, respectively; s is a phosphorothioate linkage; and L96 is N-[tris(GalNAc-alkyl)-(amidodecanoyl)]-4-hydroxyprolinol.

The dsRNAi agent of claim 1 , wherein at least one of said nucleotide modifications is selected from the group consisting of a deoxy-nucleotide, a 3′-terminal deoxy-thymine (dT) nucleotide, a 2′-O-methyl modified nucleotide, a 2′-fluoro modified nucleotide, a 2′-deoxy-modified nucleotide, a locked nucleotide, an unlocked nucleotide, a conformationally restricted nucleotide, a constrained ethyl nucleotide, an abasic nucleotide, a 2′-amino-modified nucleotide, a 2′-O-allyl-modified nucleotide, 2′-C-alkyl-modified nucleotide, 2′-hydroxly-modified nucleotide, a 2′-methoxyethyl modified nucleotide, a 2′-O-alkyl-modified nucleotide, a morpholino nucleotide, a phosphoramidate, a non-natural base comprising nucleotide, a tetrahydropyran modified nucleotide, a 1,5-anhydrohexitol modified nucleotide, a cyclohexenyl modified nucleotide, a nucleotide comprising a phosphorothioate group, a nucleotide comprising a methylphosphonate group, a nucleotide comprising a 5′-phosphate, and a nucleotide comprising a 5′-phosphate mimic.

5. The dsRNAi agent of claim 1 , wherein each strand is no more than 30 nucleotides in length.

6. The dsRNAi agent of claim 1 , wherein at least one strand comprises a 3′ overhang of at least 1 nucleotide.

7. The dsRNAi agent of claim 1 , wherein the GalNAc derivative is

8. The dsRNAi agent of claim 7 , wherein the dsRNAi agent is conjugated to the GalNAc derivative as shown in the following schematic

and, wherein X is 0 or S.

9. The dsRNAi agent of claim 8 , wherein the X is 0.

10. The dsRNAi agent of claim 1 , wherein the nucleotide modifications are selected from the group consisting of LNA, HNA, CeNA, 2′-methoxyethyl, 2′-O-alkyl, 2′-O-allyl, 2′-C— allyl, 2′-fluoro, 2′-deoxy, 2′-hydroxyl, and combinations thereof.

11. The dsRNAi agent of claim 10 , wherein the nucleotide modifications are 2′-O-methyl or 2′-fluoro modifications.

12. The dsRNAi agent of claim 1 , wherein the ligand is one or more GalNAc derivatives attached through a bivalent or trivalent branched linker.

13. The dsRNAi agent of claim 1 , wherein said agent further comprises at least one phosphorothioate or methylphosphonate internucleotide linkage.

14. The dsRNAi agent of claim 1 ,

wherein substantially all of the nucleotides of said sense strand comprise a nucleotide modification selected from the group consisting of a 2′-O-methyl modification and a 2′-fluoro modification,

wherein said sense strand comprises two phosphorothioate internucleotide linkages at the 5′-terminus,

wherein substantially all of the nucleotides of said antisense strand comprise a nucleotide modification selected from the group consisting of a 2′-O-methyl modification and a 2′-fluoro modification, and

wherein said antisense strand comprises two phosphorothioate internucleotide linkages at the 5′-terminus and two phosphorothioate internucleotide linkages at the 3′-terminus.

15. A pharmaceutical composition for inhibiting expression of a PD-Ll gene comprising the dsRNAi agent of claim 1 .

16. The pharmaceutical composition of claim 15 , further comprising a lipid formulation.

17. A method of inhibiting PD-Ll expression in a cell, the method comprising contacting the cell with the double stranded RNAi agent of claim 1 , thereby inhibiting expression of the PD-Ll gene in the cell.

18. The method of claim 17 , wherein said cell is within a human subject.
