Abstract

Implantable devices and methods for delivery of lidocaine or other drugs to a patient are provided. In one embodiment, the device includes a first drug portion which has a first drug housing which contains a first drug formulation in a solid form which includes a pharmaceutically acceptable salt of lidocaine; and a second drug portion which includes a second drug housing which contains a second drug formulation which includes lidocaine base. In another embodiment, the device includes a drug reservoir component which has an elastic tube having at least one lumen bounded by a porous sidewall having an open-cell structure, a closed-cell structure, or a combination thereof; and a drug formulation contained within the at least one lumen, wherein the device is deformable between a low-profile deployment shape and a relatively expanded retention shape.

Claims (29)

1. An intravesical drug delivery device for controlled drug delivery comprising:

a drug reservoir component which comprises an elastic tube having at least one lumen bounded by a porous sidewall having an open-cell structure, a closed-cell structure, or a combination thereof;

a retention frame coupled to the drug reservoir component; and

a first drug formulation contained within the at least one lumen, wherein the first drug formulation is in the form of a plurality of aligned tablets, wherein the tablets are cylindrical with flat end faces,

wherein the device reservoir component is deformable between a low-profile deployment shape and a coiled retention shape, and

wherein the retention frame comprises one or more loops or curls when the device reservoir component is in the coiled retention shape and biases the device reservoir component to return from the low-profile deployment shape to the coiled retention shape.

2. The device of claim 1 , wherein the first drug formulation is configured to become solubilized in vivo for release through the porous sidewall.

3. The device of claim 1 , further comprising a second drug formulation.

4. The device of claim 3 , wherein the first drug formulation comprises a high solubility drug and the second drug formulation comprises a low solubility drug.

5. The device of claim 1 , wherein the porous sidewall has an open-cell structure.

6. The device of claim 5 , wherein the porous sidewall comprises at least one passageway, wherein the passageway is an aperture formed completely through the porous sidewall by drilling, punching, or molding.

7. The device of claim 5 , wherein the porous sidewall is coated to form a closed-cell structure.

8. The device of claim 5 , wherein the porous sidewall comprises pores randomly distributed throughout the porous sidewall.

9. The device of claim 1 , further comprising a retention frame lumen in which the retention frame is disposed.

10. An intravesical drug delivery device for controlled drug delivery comprising:

a device body comprising a drug reservoir component which comprises an elastic tube having a drug reservoir lumen bounded by a porous sidewall having an open-cell structure;

a retention frame coupled to the drug reservoir component; and

a first drug formulation contained within the drug reservoir lumen, wherein the first drug formulation is in the form of a plurality of aligned tablets, wherein the tablets are cylindrical with flat end faces,

wherein the device body is deformable between a low-profile deployment shape and a coiled retention shape, and

wherein the retention frame comprises one or more loops or curls when the device body is in the coiled retention shape and biases the device body to return from the low-profile deployment shape to the coiled retention shape.

The device of claim 10 , wherein the device body further comprises a retention frame lumen in which the retention frame is disposed, wherein the drug reservoir lumen and the retention frame lumen are parallel to one another over the full length of the device body, in both the deployment shape and the coiled retention shape.

12. An intravesical drug delivery device for controlled drug delivery comprising:

a device body comprising a drug reservoir component which comprises an elastic tube having a drug reservoir lumen bounded by a porous sidewall having an open-cell structure;

a retention frame coupled to the drug reservoir component; and

a drug formulation contained within the drug reservoir lumen, wherein the drug formulation is in the form of a plurality of aligned tablets, wherein the tablets are cylindrical with flat end faces,

wherein:

the intravesical drug delivery device is deformable between a low-profile deployment shape and a coiled retention shape, and

the retention frame comprises one or more windings, coils, loops or spirals so that the retention frame functions as a spring to bias the intravesical drug delivery device to return from the low-profile deployment shape to the coiled retention shape.

The device of claim 12 , wherein the device body further comprising a retention frame lumen in which the retention frame is disposed, wherein the drug reservoir lumen and the retention frame lumen are parallel to one another over the full length of the device body, in both the deployment shape and the coiled retention shape.
