DETECTION OF ACUTE MYELOID LEUKAEMIA
The present invention relates to diagnostic screens, antibodies, methods and kits for detection/prognosis of acute myeloid leukaemia. The diagnostic screen detects the presence (+) or absence (−) of the cell surface polypeptide markers i) CD34+; ii) CD45RA+; and iii) CD90− and/or CD123+. Antibodies specific for one or more of said cell surface polypeptide markers may be used in the diagnostic screen of the invention. Diagnostic and prognostic methods for detecting and monitoring minimal residual disease based on said screen also form part of the invention.

21 . A method of detecting the presence or absence of polypeptide markers CD34, CD45RA, CD123 and CD38 on a blood cell of a patient comprising:
1) obtaining a blood cell sample from the patient;
2) contacting the sample with:
i) a first antibody that specifically binds to CD34,
ii) a second antibody that specifically binds to CD45RA,
iii) a third antibody that specifically binds to CD123, and
iv) a fourth antibody that specifically binds to CD38; and
3) detecting binding of the first, second, and third antibodies to the polypeptide markers CD34, CD45RA and CD123 on said blood cell, respectively; and detecting the absence of binding of the fourth antibody to the polypeptide marker CD38 on said blood cell.
22 . The method of claim 21 , wherein at least one of the antibodies is a labeled antibody.
23 . The method of claim 22 , wherein the labeled antibody is labeled with a fluorescent dye directly bound thereto.
24 . The method of claim 21 , wherein the binding or absence of binding of the antibodies is detected sequentially.
25 . The method of claim 23 , wherein the binding or absence of binding of the antibodies is detected by fluorescence-activated cell sorting (FACS).
26 . The method of claim 21 , wherein step 2) further comprises contacting the sample with one or more selected from the group consisting of:
v) an antibody that specifically binds to CD47;
vi) an antibody that specifically binds to CD19;
vii) an antibody that specifically binds to CCR8;
viii) an antibody that specifically binds to RHAMM;
ix) an antibody that specifically binds to CD86; and
x) an antibody that specifically binds to CD90.
27 . The method of claim 21 further comprising:
4) administering a therapy for acute myeloid leukaemia.
28 . A method comprising:
a) obtaining the results of the method of claim 21 ; and
b) administering a therapy for acute myeloid leukaemia.
