Profiling peptides and methods for sensitivity profiling
View Patent ↗The present disclosure is generally directed to profiling peptides, compositions, and kits, as well as methods of use thereof. The profiling peptides comprise an Mcl-1 binding domain, and optionally a cellular uptake moiety. The methods of using such profiling peptides include predicting sensitivity of a cancer, selecting a treatment, treating a cancer, producing a sensitivity profile, and the like.
1. A profiling peptide comprising the sequence of SEQ ID NO:14.
2. A composition comprising a profiling peptide comprising the sequence of SEQ ID NO:14 and a carrier.
3. The profiling peptide of claim 1 , consisting of the sequence of SEQ ID NO:14.
4. The composition of claim 2 , wherein the profiling peptide consists of the sequence of SEQ ID NO:14.
5. The composition of claim 2 , further comprising a cancer cell.
6. The composition of claim 5 , wherein the cancer cell is a cell of a hematologic cancer.
7. The composition of claim 6 , wherein the hematologic cancer is multiple myeloma, myelodysplastic syndrome (MDS), acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), acute lymphocytic leukemia, chronic lymphogenous leukemia, chronic lymphocytic leukemia (CLL), mantle cell lymphoma, diffuse large B-cell lymphoma, follicular lymphoma, or non-Hodgkin's lymphoma.
8. A kit comprising:
a profiling peptide comprising the sequence of SEQ ID NO:14;
and a detecting agent.
9. The kit of claim 8 , further comprising a positive control and a negative control.
10. The kit of claim 9 , wherein the positive control is carbonyl cyanide m-chlorophenyl hydrazone (CCCP).
11. The kit of claim 8 , further comprising written instructions.
12. The kit of claim 8 , wherein the detecting agent is a dye.
13. The kit of claim 8 , wherein the dye is a 3,3′-Dihexyloxacarbocyanine Iodide (DiOC 6 ).
14. The kit of claim 8 , wherein the dye is a potentiometric dye.
15. The kit of claim 8 , wherein the profiling peptide consists of the sequence of SEQ ID NO:14.
