Abstract

The present invention relates to stabilization of cells at ambient temperatures. More particularly, the present invention relates to formulations, compositions, kits and methods that allow dehydration and rehydration of cells and dramatically increased recovery of functional cells after dry storage at room temperature.

Claims (37)

1 .- 72 . (canceled)

73 . A method for substantially dry storage of one or more cell at ambient temperatures in the absence of refrigeration, comprising:

(i) incubating the one or more cell with a dehydration formulation comprising a dry storage stabilizer and at least one apoptosis inhibitor; and

(ii) dehydrating the one or more cell in the presence of a dehydration formulation to generate one or more substantially dry stored cell.

74 . A method for substantially dry storage of one or more cell at ambient temperatures in the absence of refrigeration, comprising:

(i) incubating the one or more cell with a predehydration formulation comprising at least one apoptosis inhibitor;

(ii) removing the predehydration formulation from the one or more cell; and

(iii) dehydrating the one or more cell in the presence of a dehydration formulation comprising at least one dry storage stabilizer that is not a disaccharide to generate one or more substantially dry stored cell.

75 . The method of claim 73 , wherein the dehydration formulation is one of the dehydration formulations of Table 1.

76 . The method of claim 74 , wherein the dehydration formulation is one of the dehydration formulations of Table 1.

77 . The method of claim 74 , wherein the apoptosis inhibitor is a reversible apoptosis inhibitor.

78 . The method of claim 74 , further comprising immobilizing one or more cell to a solid support prior to incubating the one or more cell with the predehydration formulation.

79 . The method of claim 74 , further comprising rehydrating the one or more substantially dry stored cell to generate a rehydrated cell using a rehydration formulation comprising at least one apoptosis inhibitor.

80 . The method of claim 76 , further comprising rehydrating the one or more substantially dry stored cell to generate a rehydrated cell using a rehydration formulation comprising at least one apoptosis inhibitor.

81 . The method of claim 76 , wherein the apoptosis inhibitor is a reversible apoptosis inhibitor.

82 . The method of claim 79 , wherein the at least one apoptosis inhibitor in the predehydration formulation is the same as the at least one apoptosis inhibitor in the rehydration formulation.

83 . The method of claim 79 , wherein the at least one apoptosis inhibitor in the predehydration formulation is different from the at least one apoptosis inhibitor in the rehydration formulation.

84 . The method of claim 73 , wherein the dehydration formulation is selected from the group consisting of:

(a) L-Glutamine, EDTA, Tris-HCl, and PVA;

(b) Alanine-Glutamine, EDTA, Tris-HCl, Proline, and PVA;

(c) Alanine-Glutamine, EDTA, Tris-HCl, trans-4-hydroxy L-proline, and PVA;

(d) hydroxyectoine, EDTA, Tris-HCl, L-Glutamine, and PVA;

(e) Alanine-Glutamine, Betaine, Bis-Tris, and PVA; and

(f) Alanine-Glutamine, Betaine, MES, and PVA.

85 . The method of claim 74 , wherein the dehydration formulation is selected from the group consisting of:

(a) L-Glutamine, EDTA, Tris-HCl, and PVA;

(b) Alanine-Glutamine, EDTA, Tris-HCl, Proline, and PVA;

(c) Alanine-Glutamine, EDTA, Tris-HCl, trans-4-hydroxy L-proline, and PVA;

(d) hydroxyectoine, EDTA, Tris-HCl, L-Glutamine, and PVA;

(e) Alanine-Glutamine, Betaine, Bis-Tris, and PVA; and

(f) Alanine-Glutamine, Betaine, MES, and PVA.

86 . The method of claim 74 , wherein the dehydration formulation comprises L-Glutamine, EDTA, Tris-HCl, and PVA.

87 . The method of claim 74 , wherein the dehydration formulation comprises Alanine-Glutamine, EDTA, Tris-HCl, Proline, and PVA.

88 . The method of claim 74 , wherein the dehydration formulation comprises Alanine-Glutamine, EDTA, Tris-HCl, trans-4-hydroxy L-proline, and PVA.

89 . The method of claim 74 , wherein the dehydration formulation comprises hydroxyectoine, EDTA, Tris-HCl, L-Glutamine, and PVA.

90 . The method of claim 74 , wherein the dehydration formulation comprises Alanine-Glutamine, Betaine, Bis-Tris, and PVA.

91 . The method of claim 74 , wherein the dehydration formulation comprises Alanine-Glutamine, Betaine, MES, and PVA.
