Abstract

The present invention provides, among other things, a method of intra-articular delivery of messenger RNA (mRNA), comprising administering into a joint of a subject in need of delivery a composition comprising an mRNA encoding a protein, such that the administering of the composition results in expression of the protein encoded by the mRNA in the joint.

Claims (16)

1. A method of intra-articular delivery of messenger RNA (mRNA), comprising

directly injecting into a joint of a subject in need thereof an effective amount of a composition comprising one or more mRNAs encoding an antibody, wherein the one or more mRNAs are encapsulated within one or more lipid nanoparticles comprising one or more cationic lipids, one or more non-cationic lipids, one or more cholesterol-based lipids and one or more PEG-modified lipids, such that the injecting of the composition results in expression of the antibody encoded by the one or more mRNAs in the joint for at least 6 hours.

2. The method of claim 1 , wherein the composition is injected directly into the synovial cavity.

3. The method of claim 1 , wherein the expression and/or activity of the one or more polypeptides is detected in cartilage, ligament, muscle, synovial intimal cells, chondrocytes and/or fluids of the joint.

4. The method of claim 1 , wherein the expression and/or activity of the one or more polypeptides is detectable at least about 12 hours, 18 hours, 24 hours, 2 days, 3 days, 4 days, 5 days, 6 days, 1 week, 2 weeks, 3 weeks, 4 weeks, or 1 month post-injection.

5. The method of claim 1 , wherein the one or more polypeptides encoded by the one or more mRNAs normally function in the joint.

6. The method of claim 1 , wherein the one or more polypeptides encoded by the one or more mRNAs are anti-inflammatory.

7. A method of treating a joint disease, disorder or condition, comprising directly injecting into a joint one or more mRNAs using a method according to any one of the preceding claims, wherein directly injecting the composition into the joint treats the joint disease, disorder or condition.

8. The method of claim 7 , wherein the joint disease, disorder or condition is an inflammatory disease, disorder or condition.

9. The method of claim 8 , wherein the inflammatory disease, disorder or condition is arthritis.

10. The method of claim 7 , wherein the composition is directly injected into the joint once a week.

11. The method of claim 7 , wherein the composition is directly injected into the joint twice a month.

12. The method of claim 7 , wherein the composition is directly injected into the joint once a month.

13. The method of claim 1 , wherein the one or more cationic lipids comprise cKK-E12:

14. The method of claim 1 , wherein the nanoparticles have a size less than about 40-100 nm.

15. The method of claim 1 , wherein the one or more mRNAs comprise one or more modified nucleotides.
