Abstract

The present invention relates to methods of selecting, screening, engineering, making and modifying antibodies that have improved bioavailability upon subcutaneous administration to a human. Antibodies and variant antibodies with improved bioavailability upon subcutaneous administration to a human are also described.

Claims (23)

1. A method of making an antibody, comprising a) selecting and combining two or more human germline framework sequences, regions, or amino acids to generate an antibody sequence; and b) calculating a net charge of the antibody sequence with GPMAW (General Protein/Mass Analysis for Windows) in a computer, to make the antibody which has: a net charge of between ±4 and ±12 at pH 6.6 to 7.4, wherein the antibody has a higher bioavailability than the human germline framework sequences, regions, or amino acids.

2. The method of claim 1 , wherein the antibody is an IgG1 antibody.

3. The method of claim 1 , wherein the net charge or the pI is calculated using the entire sequence of the antibody, including a C-terminal lysine.

4. The method of claim 1 , wherein the net charge is between +4 to +10.

5. The method of claim 1 , further comprising making one or more amino acid substitutions in the antibody.

6. The method of claim 1 , wherein the net charge is calculated at a pH value from 6.7 to 7.3.

7. The method of claim 1 , wherein the combination:

(a) changes a positive amino acid to a neutral or negative amino acid;

(b) changes a negative amino acid to a neutral or positive amino acid; or

(c) changes a neutral amino acid to a positive or negative amino acid.

8. The method of claim 1 , wherein there is a positive or negative or neutral amino acid at any one or a combination of Kabat residues H1, H6, H12, H13, H16, H19, H23, H43, H75, H81, H83, H85, L17, L18, L42, L74, L77, or L79 of the antibody.

9. The method of claim 1 , wherein the antibody has a bioavailability of at least 60% upon subcutaneous administration to a human.

10. The method of claim 9 , wherein the bioavailability is a predicted bioavailability.

11. The method of claim 1 , wherein the net charge is calculated at a pH value from 6.8 to 7.2.

12. The method of claim 1 , wherein the net charge is calculated at a pH value from 6.9 to 7.1.

13. The method of claim 1 , wherein the antibody has a bioavailability of at least 65% upon subcutaneous administration to a human.

14. The method of claim 1 , wherein the antibody has a bioavailability of at least 70% upon subcutaneous administration to a human.

15. The method of claim 1 , wherein the antibody has a bioavailability of at least 75% upon subcutaneous administration to a human.

16. The method of claim 1 , wherein the net charge is between +4 to +8.

17. The method of claim 1 , wherein the net charge is between +4 to +6.

18. The method of claim 1 , wherein the antibody has a pI of between 8.4 and 9.3.

19. The method of claim 1 , wherein the antibody has a pI of between 8.5 to 9.3.

20. The method of claim 1 , wherein the antibody has a pI of between 8.2 to 5.2.
