Abstract

Oral dosage forms of osteoclast inhibitors, such as zoledronic acid, in an acid or a salt form can be used to treat or alleviate pain or related conditions, such as arthritis.

Claims (28)

1. A method of treating arthritis, comprising: 1) fasting a human being for at least one hour, wherein the human being is suffering from arthritis, followed by 2) orally administering a dosage form comprising zoledronic acid to the human being, followed by 3) fasting the human being for at least one hour, wherein the zoledronic acid is administered to the human being in a manner that results in a bioavailability of zoledronic acid that is about 1.5% to about 2%.

2. The method of claim 1 , wherein about 200 mg to about 300 mg of the zoledronic acid is orally administered to the human being within a period of about one month.

3. The method of claim 2 , wherein the zoledronic acid is orally administered for one to two months.

4. The method of claim 1 , wherein a weekly dose of about 50 mg to about 100 mg of the zoledronic acid is orally administered to the human being.

5. The method of claim 4 , wherein the zoledronic acid is orally administered for one to two months.

6. The method of claim 1 , wherein the zoledronic acid is administered in a disodium salt form.

7. The method of claim 1 , wherein the dosage form is orally administered in a manner that is intended to maximize the bioavailability of zoledronic acid.

8. A method of treating arthritis, comprising: 1) fasting a human being for at least one hour, wherein the human being is suffering from arthritis, followed by 2) orally administering a dosage form comprising zoledronic acid to the human being, followed by 3) fasting the human being for at least one hour, wherein the zoledronic acid is administered in a manner that results in a bioavailability of zoledronic acid that is about 2% to 3%.

9. The method of claim 8 , wherein about 200 mg to about 300 mg of the zoledronic acid is orally administered to the human being within a period of about one month.

10. The method of claim 9 , wherein the zoledronic acid is orally administered for one to two months.

11. The method of claim 8 , wherein a weekly dose of about 50 mg to about 100 mg of the zoledronic acid is orally administered to the human being.

12. The method of claim 11 , wherein the zoledronic acid is orally administered for one to two months.

13. The method of claim 8 , wherein a weekly dose of about 50 mg to about 100 mg of the zoledronic acid in a disodium salt form is orally administered to the human being.

14. The method of claim 8 , wherein the dosage form is orally administered in a manner that is intended to maximize the bioavailability of zoledronic acid.

15. A method of treating arthritis, comprising: 1) fasting a human being for at least one hour, wherein the human being is suffering from arthritis, followed by 2) orally administering a dosage form comprising zoledronic acid, in a salt form, to the human being, followed by 3) fasting the human being for at least one hour;

wherein the dosage form has a bioavailability of zoledronic acid in a beagle dog that is improved by about 30% to about 200% as compared to zoledronic acid in the diacid form as determined by the following test:

comparing the AUC 0-∞ obtained by orally administering the dosage form to a first beagle dog to the AUC 0-∞ obtained by orally administering a reference dosage form containing zoledronic acid in a diacid form to a second beagle dog;

wherein the dosage form and the reference dosage form contain identical excipients with amounts of the excipients adjusted to the same molar ratio to account for the difference in molecular weights between zoledronic acid in the diacid form and zoledronic acid in the salt form;

wherein the first beagle dog is fasted for two hours before the dosage form is administered and is fasted for two hours after the dosage form is administered; and

wherein the second beagle dog is fasted for two hours before the reference dosage form is administered and is fasted for two hours after the reference dosage form is administered.

16. The method of claim 15 , wherein about 200 mg to about 300 mg of the zoledronic acid in the salt form is orally administered to the human being within a period of about one month.

17. The method of claim 16 , wherein the zoledronic acid in the salt form is orally administered for one to two months.

18. The method of claim 15 , wherein a weekly dose of about 50 mg to about 100 mg of the zoledronic acid in the salt form is orally administered to the human being.

19. The method of claim 18 , wherein the zoledronic acid in the salt form is orally administered for one to two months.

20. The method of claim 15 , wherein a weekly dose of about 50 mg to about 100 mg of the zoledronic acid in a disodium salt form is orally administered to the human being.

21. The method of claim 15 , wherein the dosage form is orally administered in a manner that is intended to maximize the bioavailability of zoledronic acid.

22. The method of claim 15 , wherein the zoledronic acid in the salt form is at least 10% of the weight of the dosage form.

23. The method of claim 15 , wherein the dosage form is substantially free of agents that enhance the bioavailability of zoledronic acid beyond that achieved from zoledronic acid in the salt form.
