Abstract

An isolated polynucleotide encoding a modified luciferase polypeptide and substrates. The OgLuc variant polypeptide has at least 60% amino acid sequence identity to SEQ ID NO: 1 and at least one amino acid substitution at a position corresponding to an amino acid in SEQ ID NO: 1. The OgLuc variant polypeptide has at least one of enhanced luminescence, enhanced signal stability, and enhanced protein stability relative to the corresponding polypeptide of the wild-type Oplophorus luciferase.

Claims (85)

1. A polypeptide comprising an amino acid sequence having at least 80% sequence identity with the amino acid sequence of SEQ ID NO: 1 with the exception of amino acid substitutions at positions corresponding to positions 4, 18, 27, 33, 43, 54, 68, 72, 75, 90, and 166 of SEQ ID NO: 1; wherein the amino acid sequence of the polypeptide has less than 100% sequence identity with SEQ ID NO: 1 and wherein said polypeptide has enhanced luminescence activity relative to a polypeptide consisting of the amino acid sequence of SEQ ID NO: 3.

The polypeptide of claim 1 , wherein the polypeptide further comprises an amino acid substitution relative to SEQ ID NO: 1 at one or more positions corresponding to positions 38, 109, 15, 19, 21, 22, 36, 40, 56, 58, 70, 93, 95, 102, 110, 112, 119, 127, 128, 149, 152, 155, 158, 159, 1, 6, 10, 14, 16, 24, 25, 31, 32, 39, 42, 46, 47, 48, 49, 50, 55, 59, 60, 66, 67, 69, 71, 74, 86, 87, 94, 96, 97, 98, 100, 106, 111, 113, 117, 125, 126, 129, 130, 136, 142, 145, 146, 147, 148, 150, 154, 163, or 168 of SEQ ID NO: 1.

3. The polypeptide of claim 2 , wherein said amino acid sequence comprises one or more of (i)-(lxxvi):

(i) an amino acid at the position corresponding to position 18 of SEQ ID NO: 1 is aspartate, phenylalanine, glycine, histidine, isoleucine, lysine, leucine, methionine, asparagine, proline, arginine, serine, valine, tryptophan, or tyrosine,

(ii) an amino acid at the position corresponding to position 27 of SEQ ID NO: 1 is valine, alanine, aspartate, glycine or isoleucine,

(iii) an amino acid at the position corresponding to position 43 of SEQ ID NO: 1 is asparagine,

(iv) an amino acid at the position corresponding to position 38 of SEQ ID NO: 1 is isoleucine or phenylalanine,

(v) an amino acid at the position corresponding to position 109 of SEQ ID NO: 1 is phenylalanine,

(vi) an amino acid at the position corresponding to position 18 of SEQ ID NO: 1 is arginine,

(vii) an amino acid at the position corresponding to position 19 of SEQ ID NO: 1 is asparagine,

(viii) an amino acid at the position corresponding to position 21 of SEQ ID NO: 1 is leucine or methionine,

(ix) an amino acid at the position corresponding to position 22 of SEQ ID NO: 1 is isoleucine or phenylalanine,

(x) an amino acid at the position corresponding to position 36 of SEQ ID NO: 1 is methionine or glutamate,

(xi) an amino acid at the position corresponding to position 40 of SEQ ID NO: 1 is threonine, isoleucine, leucine or glutamine,

(xii) an amino acid at the position corresponding to position 58 of SEQ ID NO: 1 is isoleucine or leucine,

(xiii) an amino acid at the position corresponding to position 70 of SEQ ID NO: 1 is valine,

(xiv) an amino acid at the position corresponding to position 93 of SEQ ID NO: 1 is arginine,

(xv) an amino acid at the position corresponding to position 18 of SEQ ID NO: 1 is serine or aspartate,

(xvi) an amino acid at the position corresponding to position 102 of SEQ ID NO: 1 is methionine, glutamate, or threonine,

(xvii) an amino acid at the position corresponding to position 18 of SEQ ID NO: 1 is isoleucine,

(xviii) an amino acid at the position corresponding to position 112 of SEQ ID NO: 1 is cysteine,

(xix) an amino acid at the position corresponding to position 119 of SEQ ID NO: 1 is methionine,

(xx) an amino acid at the position corresponding to position 127 of SEQ ID NO: 1 is alanine,

(xxi) an amino acid at the position corresponding to position 128 of SEQ ID NO: 1 is alanine or serine,

(xxii) an amino acid at the position corresponding to position 149 of SEQ ID NO: 1 is isoleucine or methionine,

(xxiii) an amino acid at the position corresponding to position 152 of SEQ ID NO: 1 is histidine, serine, glutamate or threonine,

(xxiv) an amino acid at the position corresponding to position 155 of SEQ ID NO: 1 is threonine,

(xxv) an amino acid at the position corresponding to position 158 of SEQ ID NO: 1 is isoleucine,

(xxvi) an amino acid at the position corresponding to position 159 of SEQ ID NO: 1 is serine or isoleucine,

(xxvii) an amino acid at the position corresponding to position 1 of SEQ ID NO: 1 is isoleucine,

(xxviii) an amino acid at the position corresponding to position 6 of SEQ ID NO: 1 is tyrosine,

(xxix) an amino acid at the position corresponding to position 10 of SEQ ID NO: 1 is tyrosine,

(xxx) an amino acid at the position corresponding to position 14 of SEQ ID NO: 1 is valine or serine,

(xxxi) an amino acid at the position corresponding to position 16 of SEQ ID NO: 1 is glutamate,

(xxxii) an amino acid at the position corresponding to position 24 of SEQ ID NO: 1 is alanine,

(xxxiii) an amino acid at the position corresponding to position 25 of SEQ ID NO: 1 is leucine,

(xxxiv) an amino acid at the position corresponding to position 31 of SEQ ID NO: 1 is isoleucine,

(xxxv) an amino acid at the position corresponding to position 32 of SEQ ID NO: 1 is proline or histidine,

(xxxvi) an amino acid at the position corresponding to position 39 of SEQ ID NO: 1 is isoleucine,

(xxxvii) an amino acid at the position corresponding to position 42 of SEQ ID NO: 1 is histidine,

(xxxviii) an amino acid at the position corresponding to position 46 of SEQ ID NO: 1 is glutamine,

(xxxix) an amino acid at the position corresponding to position 47 of SEQ ID NO: 1 is proline at position 17,

(xl) an amino acid at the position corresponding to position 48 of SEQ ID NO: 1 is arginine,

(xli) an amino acid at the position corresponding to position 49 of SEQ ID NO: 1 is aspartate, glycine, or lysine,

(xlii) an amino acid at the position corresponding to position 50 of SEQ ID NO: 1 is lysine or serine,

(lix) an amino acid at the position corresponding to position 111 of SEQ ID NO: 1 is asparagine,

(lx) an amino acid at the position corresponding to position 113 of SEQ ID NO: 1 is histidine, threonine, or lysine,

(lxi) an amino acid at the position corresponding to position 117 of SEQ ID NO: 1 is phenylalanine,

(lxii) an amino acid at the position corresponding to position 125 of SEQ ID NO: 1 is leucine,

(lxiii) an amino acid at the position corresponding to position 126 of SEQ ID NO: 1 is arginine,

(lxiv) an amino acid at the position corresponding to position 129 of SEQ ID NO: 1 is glutamine,

(lxv) an amino acid at the position corresponding to position 130 of SEQ ID NO: 1 is lysine,

(lxvi) an amino acid at the position corresponding to position 136 of SEQ ID NO: 1 is methionine,

(lxvii) an amino acid at the position corresponding to position 142 of SEQ ID NO: 1 is valine, glutamate, lysine or tryptophan,

(lxviii) an amino acid at the position corresponding to position 145 of SEQ ID NO: 1 is leucine or glutamine,

(lxix) an amino acid at the position corresponding to position 146 of SEQ ID NO: 1 is glycine,

(lxx) an amino acid at the position corresponding to position 147 of SEQ ID NO: 1 is asparagine,

(lxxi) an amino acid at the position corresponding to position 148 of SEQ ID NO: 1 is threonine,

(lxxii) an amino acid at the position corresponding to position 150 of SEQ ID NO: 1 is valine,

(lxxiii) an amino acid at the position corresponding to position 154 of SEQ ID NO: 1 is serine,

(lxxiv) an amino acid at the position corresponding to position 163 of SEQ ID NO: 1 is isoleucine,

(lxxv) an amino acid at the position corresponding to position 168 of SEQ ID NO: 1 is phenylalanine, or

(lxxvi) an amino acid at the position corresponding to position 56 of SEQ ID NO: 1 is valine.

4. The polypeptide of claim 1 , wherein the polypeptide further comprises an amino acid substitution relative to SEQ ID NO: 1 at one or more positions corresponding to positions 1, 10, 14, 16, 24, 25, 38, 40, 70, 87, 97, 100, 102, 111, 113, 125, 129, 130, 142, 146, 147, 149, 150, and 152 of SEQ ID NO: 1.

5. The polypeptide of claim 1 , wherein said amino acid sequence comprises isoleucine or alanine at the position corresponding to position 54 and valine at the position corresponding to position 90 of the amino acid sequence of SEQ ID NO: 1.

The polypeptide of claim 4 , wherein said amino acid sequence comprises leucine at the position corresponding to position 18 of SEQ ID NO: 1, tyrosine at the position corresponding to position 68 of SEQ ID NO: 1, glutamine at the position corresponding to position 72 of SEQ ID NO: 1, and lysine at the position corresponding to position 75 of SEQ ID NO: 1.

The polypeptide of claim 4 , wherein said amino acid sequence comprises asparagine at the position corresponding to position 33 of SEQ ID NO: 1, valine at the position corresponding to position 27 of SEQ ID NO: 1, arginine at the position corresponding to position 43 of SEQ ID NO: 1, and aspartate at the position corresponding to position 68 of SEQ ID NO: 1.

8. The polypeptide of claim 1 , wherein said polypeptide is a soluble monomer.

9. A fusion polypeptide comprising the polypeptide of claim 1 and a polypeptide of interest.
