Abstract

According to an illustrative embodiment a method to promote healing of a wound is provided comprising contacting the wound with a biologically active composition comprising a lipoic acid derivative and gelatin. In another embodiment a wound dressing is provided comprising a scaffold coated with a biologically active composition comprising a lipoic acid derivative. In a further embodiment, a system is provided for treating a tissue site of a patient, the system comprising a reduced-pressure source to supply reduced pressure, a manifold to distribute reduced pressure to a tissue site and a scaffold coated with a biologically active composition comprising a lipoic acid derivative. Methods for producing such a system and scaffold are also disclosed.

Claims (19)

1. A reduced-pressure therapy system for treating a tissue site, comprising:

a manifold at least partially coated with a composition comprising a gelatin substrate, a lipoic acid derivative, and a chelating agent, wherein the composition reduces excessive matrix metalloprotease activity, and wherein the lipoic acid derivative has the structure:

wherein n 1 and n 2 are, independently, C 1 -C 10 alkyl; and R 1 is H, C 1 -C 10 alkyl, C 6 -C 14 aryl, an alkyl ammonium or a protonated amino acid;

a drape adapted to provide a sealed space around the manifold and the tissue site; and

a reduced-pressure source configured to deliver reduced pressure to the sealed space.

2. The system of claim 1 , further comprising a scaffold adapted to be positioned between the manifold and the tissue site, wherein the scaffold has a plurality of passages.

3. The system of claim 2 , wherein the scaffold comprises polyurethane or collagen.

4. The system of claim 2 , further comprising a release layer adapted to be positioned between the scaffold and the manifold.

5. The system of claim 1 , further comprising an interface for fluidly coupling the reduced-pressure source to the sealed space.

6. The system of claim 1 , wherein the lipoic acid derivative comprises alpha-lipoic acid or sodium lipoate.

7. The system of claim 1 , wherein the chelating agent comprises ethylenediaminetetraacetic acid.

8. The system of claim 1 , wherein the composition further comprises an antimicrobial agent, a growth factor, or a preservative.

9. The system of claim 8 , wherein the antimicrobial agent is non-ionic silver, polyhexamethylene biguanide, chlorhexidine, benzalconium chloride, or triclosan.

10. The system of claim 1 , wherein the composition further preserves granulation tissue.

11. The system of claim 1 , wherein the composition is formulated for time-controlled release of the lipoic acid derivative.

12. The system of claim 1 , wherein the composition comprises 0.1% to 25% w/w gelatin substrate.

13. The system of claim 1 , wherein the composition comprises a bloom value of between about 150-300 g.

14. The system of claim 1 , wherein the composition comprises a bloom value of between about 200-250 g.

15. The system of claim 1 , wherein the composition comprises a bloom value of about 225 g.
