Abstract

A hand-held molecular diagnostic test device includes a housing, an amplification (or PCR) module, and a detection module. The amplification module is configured to receive an input sample, and defines a reaction volume. The amplification module includes a heater such that the amplification module can perform a polymerase chain reaction (PCR) on the input sample. The detection module is configured to receive an output from the amplification module and a reagent formulated to produce a signal that indicates a presence of a target amplicon within the input sample. The amplification module and the detection module are integrated within the housing.

Claims (53)

1. A method of detecting a target amplicon using a disposable molecular diagnostic test device, comprising:

loading a sample into a sample input device, the sample input device including a proximal end portion and a distal end portion, the distal end portion configured to be matingly received within an input portion of the disposable molecular diagnostic test device, the disposable molecular diagnostic test device including a housing, a control module, and an amplification module, the control module and the amplification module being within the housing;

moving the proximal end portion of the sample input device relative to the disposable molecular diagnostic test device from a first position to a second position after the distal end portion of the sample input device is within the input portion to actuate the disposable molecular diagnostic test device to:

cause the control module to provide power from a power source to a heater of the amplification module in response to actuation of a switch of the control module caused by the moving;

amplify the sample conveyed from the sample input device by transferring heat from the heater into a reaction volume defined by the amplification module to produce the target amplicon; and

react, within the disposable molecular diagnostic test device each of A) the target amplicon and B) a reagent formulated to produce a signal indicating a presence of the target amplicon within the sample;

reading a result associated with the signal; and

discarding, after the reading, the disposable molecular diagnostic test device.

2. The method of claim 1 , wherein the signal is a non-fluorescent signal.

3. The method of claim 1 , wherein:

the reaction volume is a serpentine flow path; and

a first heating portion of the heater produces a first temperature zone within a first portion of the serpentine flow path and a second heating portion of the heater produces a second temperature zone within a second portion of the serpentine flow path, the first temperature zone and the second temperature zone maintained such that a flow of the sample within the serpentine flow path is thermally cycled to amplify a target nucleic acid within the sample to produce the target amplicon.

4. The method of claim 2 , wherein the signal is a colorimetric signal.

5. The method of claim 1 , wherein the moving the proximal end portion of the sample input device causes the sample to be conveyed from the sample input device towards the reaction volume.

6. The method of claim 5 , wherein:

the distal end portion of the sample input device includes a locking mechanism; and

the locking mechanism engages with the disposable molecular diagnostic test device to lock the sample input device within the input portion.

7. The method of claim 6 , wherein the locking mechanism is a recess that engages with a mating protrusion to prevent removal of the sample input device from the disposable molecular diagnostic test device.

8. The method of claim 5 , wherein the distal end portion of the sample input device is a tubular portion that is matingly received within the input portion of the disposable molecular diagnostic test device.

9. The method of claim 1 , wherein:

the input portion of the disposable molecular diagnostic test device defines a sample input volume; and

the moving the proximal end portion of the sample input device to the second position pressurizes the sample input volume.

10. The method of claim 1 , wherein:

the control module is implemented in at least one of a processor or a memory; and

the control module produces a thermal control signal to adjust an output of the heater to amplify the sample within the reaction volume of the amplification module.

11. The method of claim 10 , further comprising:

coupling the disposable molecular diagnostic test device to the power source.

12. The method of claim 11 , wherein the power source is a battery.

13. The method of claim 11 , wherein the power source includes a plurality of 1.5 VDC batteries.

14. The method of claim 1 , wherein the switch is a mechanical switch.

15. A method of detecting a target nucleic acid using a molecular diagnostic test device, comprising:

providing the molecular diagnostic test device, the molecular diagnostic test device being a stand-alone molecular diagnostic test device comprising an input portion, a flow path adapted to receive a biological sample, a reaction volume within the flow path, a control module, a power source, and a heater;

loading the biological sample into a sample reservoir defined within a sample transfer device, the sample transfer device including a proximal end portion and a distal end portion;

moving, after the distal end portion of the sample transfer device is received within the input portion, the proximal end portion of the sample transfer device relative to the molecular diagnostic test device to cause the biological sample to flow from the sample reservoir into the flow path and to cause actuation of a switch of the control module;

the molecular diagnostic test device performing process operations in response to the moving, the process operations comprising:

providing power from the power source to the heater after the switch is actuated to amplify, within the reaction volume, the target nucleic acid within the biological sample to produce a target amplicon; and

reacting, within the flow path, each of the target amplicon and a reagent formulated to produce a signal indicating a presence of the target amplicon within the biological sample; and

reading a result associated with the signal.

16. The method of claim 15 , further comprising:

discarding, after the reading, the molecular diagnostic test device.

17. The method of claim 15 , wherein:

the control module is implemented in at least one of a processor or a memory; and

the providing power from the power source includes regulating, via the control module, the power from the power source to the heater to produce a temperature set point within the reaction volume.

18. The method of claim 17 , wherein:

the molecular diagnostic test device includes a temperature sensor; and

the regulating is based on feedback from the temperature sensor.

19. The method of claim 15 , wherein:

the distal end portion of the sample transfer device includes a locking mechanism; and

the locking mechanism engages with the molecular diagnostic test device to lock the sample transfer device within the input portion.

20. The method of claim 19 , wherein the locking mechanism is a recess that engages with a mating protrusion to prevent removal of the sample transfer device from the molecular diagnostic test device.

21. The method of claim 15 , wherein the moving the proximal end portion of the sample transfer device pressurizes the sample reservoir.

22. The method of claim 15 , wherein the power source is a battery.

23. The method of claim 17 , wherein the stand-alone molecular diagnostic test device includes a temperature sensor.
