Abstract

The present invention is directed to epinephrine spray formulations. The present invention is further directed to methods of treating anaphylaxis by administering epinephrine spray formulations to subjects in need of such treatments.

Claims (49)

1. An epinephrine spray solution comprising:

from about 1% to about 15% w/w epinephrine or a salt thereof;

from 0.9% to 80% w/w water; and

optionally, one or more excipients selected from the group consisting of acids selected from the group consisting of hydrochloric acid, malic acid, tartaric acid, citric acid, succinic acid and combinations thereof, stabilizers selected from the group consisting of butylated hydroxyanisole, butylated hydroxytoluene, ascorbic acid, methionine, sodium ascorbate, sodium thiosulfate, sodium bisulfite, sodium metabisulfite, ascorbyl palmitate, thioglycerol, alpha tocopherol, cysteine hydrochloride, benzalkonium chloride, citric acid, ethylenediaminetetraacetic acid, sodium citrate, propyl gallate, 8-hydroxyquinoline, boric acid, histidine and combinations thereof, solvents, antioxidants, preservatives, permeation enhancers, viscosity modifiers, sweeteners, sweetness enhancers, pH modifiers, isotonicity agents and flavoring agents,

wherein the solution has a pH from about 2 to about 5.5, wherein the solution does not contain a cyclodextrin and wherein w/w denotes weight by weight of the solution.

2. The solution of claim 1 , wherein the solvent is selected from the group consisting of ethanol, glycerin, propylene glycol, polyethylene glycol 400 and a combination thereof.

3. The solution of claim 1 , wherein the acid is at a concentration of about 0.1% w/w to about 60% w/w.

4. The solution of claim 2 , wherein the solvent comprises from about 2% w/w to about 60% w/w ethanol.

5. The solution of claim 1 , wherein the acid is diluted hydrochloric acid.

6. The solution of claim 1 , wherein the epinephrine salts are selected from the group consisting of citrate, hydrochloride, halide, sulfate, bitartrate, tartrate, phosphate, acetate, malate, maleate, succinate, ascorbate, carbonate, mesylate and lactate.

7. The solution of claim 1 , wherein the stabilizer comprises EDTA at a concentration from about 0.005% w/w to about 0.5% w/w.

8. The solution of claim 1 , wherein the stabilizer comprises sodium bisulfite or sodium metabisulfite or a combination thereof at a concentration from about 0.005% w/w to about 5 w/w.

9. The solution of claim 1 , wherein the stabilizer comprises benzalkonium chloride at a concentration from about 0.005% to about 0.5% w/w.

10. An epinephrine spray solution comprising:

from about 0.1% to about 15% w/w epinephrine, or a salt thereof;

from about 1% to about 65% w/w hydrochloric acid with a normality from about 0.1 to 12N;

from about 2% to about 98% w/w water;

from about 0.005% to about 0.5% w/w sodium bisulfite;

from about 0.005% to about 0.5% w/w edetate disodium dihydrate;

from about 0.005% to about 0.5% w/w benzalkonium chloride;

wherein w/w denotes weight by weight of the solution and wherein the solution does not contain sorbitol.

11. The solution of claim 10 , wherein the water is at a concentration from about 15% to about 80% w/w.

12. The solution of claim 10 , further comprising from about 2% to about 60% w/w ethanol.

13. The solution of claim 10 , further comprising from about 0.05% to about 5% sodium chloride.

14. The solution of claim 10 , further comprising from about 1% to about 20% w/w propylene glycol.

15. An epinephrine spray formulation comprising:

about 2.96% w/w epinephrine base;

about 32.93% w/w hydrochloric acid with a normality of about 0.5N;

about 0.05% w/w edetate disodium dihydrate;

about 0.15% w/w sodium bisulfite;

about 0.01% w/w benzalkonium chloride;

about 0.6% w/w sodium chloride; and

about 63.30% w/w water.

16. An epinephrine spray formulation comprising:

about 3.18% w/w epinephrine base;

about 35.47% w/w hydrochloric acid with a normality of about 0.5N;

about 0.05% w/w edetate disodium dihydrate;

about 0.15% w/w sodium bisulfate;

about 0.01% w/w benzalkonium chloride;

about 40.0% w/w ethanol; and

about 16.14% w/w water.

17. A method of treating anaphylaxis comprising administering the solution of claim 1 to a subject in need thereof.

18. The method of claim 17 , wherein the subject is suffering from nasal congestion.

19. The method of claim 17 , wherein administration occurs via an intranasal route.

20. The method of claim 17 , wherein administration occurs via a multi-dose device that delivers more than one dose of the solution of claim 1 to the subject.

21. The method of claim 20 , wherein the multi-dose device is a bi-dose device that delivers two doses of the solution of claim 1 to the subject.

22. The solution of claim 1 , wherein the epinephrine or salt thereof is at a concentration from about 1% to about 10% w/w.

23. The solution of claim 1 , wherein the epinephrine or salt thereof is at a concentration from about 1% to about 13% w/w.

24. The solution of claim 1 , wherein the epinephrine or salt thereof is at a concentration from about 1% to about 7% w/w.
