Abstract

Embodiments of the invention provide shaped masses comprising one or more drugs such as proteins or polypeptides and methods for forming such shaped masses. One embodiment provides a shaped mass comprising a drug such as a protein or polypeptide having a biological activity in the body of a mammal. The shaped mass is formed by compression of a precursor material comprising the drug wherein an amount of biologically active drug in the mass is a preserved above a minimum level. Drugs which may be incorporated into the shaped mass may include one or more glucose regulating proteins such as insulin, incretins; and immunoglobulins such as TNF-inhibiting antibodies or interleukin neutralizing antibodies. Embodiments of the shaped mass may be incorporated into a tissue penetrating member which is inserted into the intestinal wall allowing for the oral delivery of proteins and peptides which would otherwise be degraded in the intestinal tract.

Claims (28)

1. A compressed mass comprising a TNF inhibiting antibody and a pharmaceutical excipient, wherein a percentage by weight of biologically active protein of the TNF inhibiting antibody in the compressed mass is in a range of about 60% to about 95%, the compressed mass having a density in a range of about 0.80 mg/mm 3 to about 1.10 mg/mm 3 .

2. The compressed mass of claim 1 , wherein the biological activity of the TNF inhibiting antibody in the compressed mass is in a range of about 67% to about 99% as compared to the biological activity of the TNF inhibiting antibody prior to formation of the compressed mass.

3. The compressed mass of claim 1 , wherein the TNF inhibiting antibody comprises adalimumab.

4. The compressed mass of claim 3 , wherein the amount of biologically active protein of adalimumab in the compressed mass is in a range of about 0.73 mg to about 1.09 mg.

5. The compressed mass of claim 3 , wherein the compressed mass has a weight in a range of about 1.09 mg to about 1.64 mg.

6. The compressed mass of claim 3 , wherein the length of the compressed mass is in a range of about 3.12 mm to about 4.29 mm.

7. The compressed mass of claim 3 , wherein a percentage by weight of biologically active protein of adalimumab in the compressed mass is in a range of about 59% to about 79%.

8. The compressed mass of claim 3 , wherein the compressed mass comprises between about 1 mg to about 4 mg of adalimumab.

9. The compressed mass of claim 1 , wherein the compressed mass is formed by compression of a powder comprising the TNF inhibiting antibody and pharmaceutical excipient.

10. The compressed mass of claim 1 , wherein the compressed mass has a tissue penetrating shape.

11. The compressed mass of claim 1 , wherein the compressed mass comprises a biodegradable material which breaks down in a wall of a small intestine so as to release the TNF inhibiting antibody.

12. The compressed mass of claim 1 , wherein the pharmaceutical excipient comprises at least one of a lubricant, a binding agent, or a bulking agent.

A compressed mass comprising adalimumab and a pharmaceutical excipient, wherein a percentage by weight of biologically active protein of adalimumab in the compressed mass is in a range of about 59% to about 79%, and wherein the compressed mass has a density in a range of about 0.86 mg/mm 3 to about 1.05 mg/mm 3 .

14. The compressed mass of claim 13 , wherein the amount of biologically active protein of adalimumab in the compressed mass is in the range of about 0.73 mg to about 1.09 mg.

15. The compressed mass of claim 13 , wherein the compressed mass has a weight in a range of about 1.09 mg to about 1.64 mg.

16. The compressed mass of claim 13 , wherein the length of the compressed mass is in a range of about 3.12 mm to about 4.29 mm.

17. The compressed mass of claim 16 , wherein the compressed mass comprises between about 1 mg to about 4 mg of adalimumab.

18. A compressed mass comprising adalimumab and a pharmaceutical excipient, wherein:

i) a percentage by weight of biologically active adalimumab in the compressed mass is in a range of about 59% to about 79%;

ii) the compressed mass has a density in a range of about 0.86 mg/mm 3 to about 1.05 mg/mm 3 ; and

iii) the compressed mass degrades in a tissue to release the adalimumab into a bloodstream.

19. The compressed mass of claim 18 , wherein the tissue is an intestinal tissue or an intestinal wall tissue.

20. The compressed mass of claim 19 , where the intestinal wall is a wall of a small intestine.

21. The compressed mass of claim 20 , wherein the tissue is an intramuscular tissue.

22. The compressed mass of claim 18 , wherein the amount of biologically active protein of adalimumab in the compressed mass is in a range of about 0.73 mg to about 1.09 mg.

23. The compressed mass of claim 18 , wherein the compressed mass has a weight in a range of about 1.09 mg to about 1.64 mg.

24. The compressed mass of claim 18 , wherein the length of the compressed mass is in a range of about 3.12 mm to about 4.29 mm.

25. The compressed mass of claim 18 , wherein the compressed mass comprises between about 1 mg to about 4 mg of adalimumab.
