Abstract

The present disclosure relates to fluidic systems and devices for processing, extracting, or purifying one or more analytes. These systems and devices can be used for processing samples and extracting nucleic acids, for example by isotachophoresis. In particular, the systems and related methods can allow for extraction of nucleic acids, including non-crosslinked nucleic acids, from samples such as tissue or cells. The systems and devices can also be used for multiplex parallel sample processing.

Claims (29)

1. A fluidic device comprising:

a) a first fluidic channel having length and width;

b) a second fluidic channel having length and width; and

c) an open elution reservoir positioned above said first and second fluidic channels and fluidically connected to said first and second fluidic channels through a first through hole and a second through hole, respectively, wherein said first through hole has a larger volume than said second through hole.

2. The device of claim 1 , wherein said first through hole has a circular, an elliptical, an oval, a square, a rectangular, or an elongated cross-sectional shape.

3. The device of claim 1 , wherein said first through hole has a D-shaped cross-section.

4. The device of claim 1 , wherein said first through hole is defined by one or more walls extending between said first fluidic channel and a floor of said open elution reservoir.

5. The device of claim 1 , wherein said first through hole has a maximum dimension across which is smaller than a maximum dimension across said open elution reservoir.

6. The device of claim 1 , wherein said first through hole comprises a guide wall configured to guide placement of a pipette tip toward said first fluidic channel and constrain an orientation and a position of said pipette tip to align said pipette tip with said first fluidic channel.

7. The device of claim 6 , wherein said first through hole comprises a first region located at said first fluidic channel which is narrower than a second region located at an entry way for ambient air of said open elution reservoir.

8. The device of claim 7 , wherein said first region and said second region comprise D-shaped cross-sections.

9. The device of claim 7 , wherein said guide wall tapers at an angle between said first region and said second region.

10. The device of claim 1 , wherein said first through hole and said second through hole have different shapes.

11. The device of claim 10 , wherein said first through hole has a D-shaped cross-section and said second through hole has a circular cross-section.

12. The device of claim 1 , further comprising:

d) a buffering reservoir in fluidic communication with said second fluidic channel.

13. The device of claim 12 , further comprising a high-voltage electrode disposed in said buffering reservoir and an electrode in electrical communication with said first fluidic channel, and configured to generate an electric current between said first through hole and said second through hole.

14. The device of claim 1 , wherein said first through hole and said second through hole open onto a same plane within said open elution reservoir.

15. The device of claim 1 , wherein said first through hole opens onto a first plane within said open elution reservoir and said second through hole opens onto a second plane within said open elution reservoir, wherein said first plane and said second plane within said open elution reservoir are different.

16. The device of claim 15 , wherein said first plane is positioned lower than said second plane within said open elution reservoir.

17. The device of claim 15 , wherein said first through hole is in a floor of said open elution reservoir and said second through hole is in a step, ramp, plateau, or platform within said open elution reservoir and above said floor of said open elution reservoir.

18. The device of claim 1 , wherein a volume of said open elution reservoir between said first and second through holes is no more than about 2.5 ml.

19. The device of claim 1 , wherein said open elution reservoir comprises a filleted edge between a floor of said open elution reservoir and a wall of said open elution reservoir.

20. The device of claim 1 , wherein said open elution reservoir comprises a circular, elongate, or oval cross-section.

21. The device of claim 1 , wherein said first through hole has a volume of less than about 15 ul.

22. The device of claim 1 , wherein said first through hole has a volume of less than about 7 ul.

23. The device of claim 1 , wherein said first through hole comprises a rise in fluid height from about 100 um to about 400 um at the first fluidic channel to about 2 mm to about 3 mm within said open elution reservoir.

24. The device of claim 1 , wherein said first through hole has an area within a range of about 1 mm 2 to about 2.75 mm 2 and said second through hole has an area within a range of about 0.8 mm 2 to about 1.5 mm 2 .

25. The device of claim 1 , wherein said first and second fluidic channels are on a same horizontal plane.
