Abstract

The present invention relates to selective inhibitors of PI3K delta protein kinases, methods of preparing them, pharmaceutical compositions containing them and methods of treatment and/or prevention of kinase mediated diseases or disorders with them.

Claims (29)

1. (S)-2-(1-(4-amino-3-(3-fluoro-4-isopropoxyphenyl)-1H-pyrazolo[3,4-d]-pyrimidin-1-yl)ethyl)-6-fluoro-3-(3-fluorophenyl)-4H-chromen-4-one 4-methylbenzenesulfonate having an enantiomeric excess of at least 95%.

2. A pharmaceutical composition comprising a compound of claim 1 and at least one pharmaceutically acceptable carrier.

3. The pharmaceutical composition of claim 2 , wherein the compound has an enantiomeric excess of at least 98%.

4. A method for the treatment of leukemia comprising administering to a subject in need thereof an effective amount of (S)-2-(1-(4-amino-3-(3-fluoro-4-isopropoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)ethyl)-6-fluoro-3-(3-fluorophenyl)-4H-chromen-4-one 4-methylbenzenesulfonate having an enantiomeric excess of at least 95%.

5. The method of claim 4 , wherein (S)-2-(1-(4-amino-3-(3-fluoro-4-isopropoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)ethyl)-6-fluoro-3-(3-fluorophenyl)-4H-chromen-4-one 4-methylbenzenesulfonate has an enantiomeric excess of at least 98%.

6. The method of claim 4 , further comprising the step of administering simultaneously or sequentially to a subject in need thereof at least one other anti-cancer agent.

The method of claim 4 , wherein the leukemia is selected from acute leukemia, acute lymphocytic leukemia, acute lymphoblastic leukemia, acute myelocytic leukemia, acute myelogenous leukemia, chronic myelogenous leukemia, promyelocytic leukemia, chronic lymphocytic leukemia, acute myeloid leukemia, chronic granulocytic leukemia, hairy-cell leukemia and erythroleukemia.

A method of treating chronic lymphocytic leukemia (CLL) in a human subject in need thereof comprising administering to the human subject an effective amount of (S)-2-(1-(4-amino-3-(3-fluoro-4-isopropoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)ethyl)-6-fluoro-3-(3-fluorophenyl)-4H-chromen-4-one 4-methylbenzenesulfonate having an enantiomeric excess of at least 95%.

9. The method of claim 8 , wherein (S)-2-(1-(4-amino-3-(3-fluoro-4-isopropoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)ethyl)-6-fluoro-3-(3-fluorophenyl)-4H-chromen-4-one 4-methylbenzenesulfonate has an enantiomeric excess of at least 98%.

10. The method of claim 8 , further comprising the step of administering simultaneously or sequentially to the human subject in need thereof at least one other anti-cancer agent.

A method of treating diffuse large B-cell lymphoma (DLBCL) in a human subject in need thereof comprising administering to the human subject an effective amount of (S)-2-(1-(4-amino-3-(3-fluoro-4-isopropoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)ethyl)-6-fluoro-3-(3-fluorophenyl)-4H-chromen-4-one 4-methylbenzenesulfonate having an enantiomeric excess of at least 95%.

12. The method of claim 11 , wherein (S)-2-(1-(4-amino-3-(3-fluoro-4-isopropoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)ethyl)-6-fluoro-3-(3-fluorophenyl)-4H-chromen-4-one 4-methylbenzenesulfonate has an enantiomeric excess of at least 98%.

13. The method of claim 11 , further comprising the step of administering simultaneously or sequentially to the human subject in need thereof at least one other anti-cancer agent.

A method of treating non-Hodgkin lymphoma (NHL) in a human subject in need thereof comprising administering to the human subject an effective amount of ((S)-2-(1-(4-amino-3-(3-fluoro-4-isopropoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)ethyl)-6-fluoro-3-(3-fluorophenyl)-4H-chromen-4-one 4-methylbenzenesulfonate having an enantiomeric excess of at least 95%.

15. The method of claim 14 , wherein (S)-2-(1-(4-amino-3-(3-fluoro-4-isopropoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)ethyl)-6-fluoro-3-(3-fluorophenyl)-4H-chromen-4-one 4-methylbenzenesulfonate has an enantiomeric excess of at least 98%.

16. The method of claim 14 , further comprising the step of administering simultaneously or sequentially to the human subject in need thereof at least one other anti-cancer agent.

A method for the treatment of lymphoid lineage, B-cell lymphoma, T-cell lymphoma, Hodgkin's lymphoma, hairy cell lymphoma or Burkett's lymphoma in a subject in need thereof, the method comprising administering to the subject an effective amount of (S)-2-(1-(4-amino-3-(3-fluoro-4-isopropoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)ethyl)-6-fluoro-3-(3-fluorophenyl)-4H-chromen-4-one 4-methylbenzenesulfonate having an enantiomeric excess of at least 95%.

18. The method of claim 17 , wherein (S)-2-(1-(4-amino-3-(3-fluoro-4-isopropoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)ethyl)-6-fluoro-3-(3-fluorophenyl)-4H-chromen-4-one 4-methylbenzenesulfonate has an enantiomeric excess of at least 98%.

19. The method of claim 17 , further comprising the step of administering simultaneously or sequentially to a subject in need thereof at least one other anti-cancer agent.

20. The method of claim 17 , wherein the subject suffers from B-cell lymphoma.

21. The method of claim 17 , wherein the subject suffers from T-cell lymphoma.

22. The method of claim 17 , wherein the subject suffers from Hodgkin's lymphoma.

23. A method for the treatment of lymphoid lineage, B-cell lymphoma, T-cell lymphoma, Hodgkin's lymphoma, hairy cell lymphoma, Burkett's lymphoma, hematopoietic tumors of myeloid lineage, multiple myelomas, smoldering multiple myeloma, nonsecretory myeloma, osteosclerotic myeloma, plasma cell leukemia, solitary plasmacytoma, or extramedullary plasmacytoma in a subject in need thereof, the method comprising administering to the subject an effective amount of (S)-2-(1-(4-amino-3-(3-fluoro-4-isopropoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)ethyl)-6-fluoro-3-(3-fluorophenyl)-4H-chromen-4-one 4-methylbenzenesulfonate having an enantiomeric excess of at least 95%.

24. The method of claim 23 , wherein (S)-2-(1-(4-amino-3-(3-fluoro-4-isopropoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)ethyl)-6-fluoro-3-(3-fluorophenyl)-4H-chromen-4-one 4-methylbenzenesulfonate has an enantiomeric excess of at least 98%.

25. The method of claim 23 , further comprising the step of administering simultaneously or sequentially to a subject in need thereof at least one other anti-cancer agent.

26. A method for the treatment of multiple myeloma (MM), small lymphocytic lymphoma (SLL), indolent non-Hodgkin's lymphoma (I-NHL), mantle cell lymphoma (MCL), follicular lymphoma, Waldestrom's macroglobulinemia (WM), T-cell lymphoma or B-cell lymphoma in a subject in need thereof, the method comprising administering to the subject an effective amount of (S)-2-(1-(4-amino-3-(3-fluoro-4-isopropoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)ethyl)-6-fluoro-3-(3-fluorophenyl)-4H-chromen-4-one 4-methylbenzenesulfonate having an enantiomeric excess of at least 95%.

27. The method of claim 26 , wherein (S)-2-(1-(4-amino-3-(3-fluoro-4-isopropoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)ethyl)-6-fluoro-3-(3-fluorophenyl)-4H-chromen-4-one 4-methylbenzenesulfonate has an enantiomeric excess of at least 98%.

28. The method of claim 26 , further comprising the step of administering simultaneously or sequentially to a subject in need thereof at least one other anti-cancer agent.

29. The method of claim 26 , wherein the subject suffers from multiple myeloma.
