Analyte detection from breath samples
Systems and techniques for collecting and analyzing breath samples to detect one or more target analytes are disclosed.
1. A cartridge for use in obtaining breath samples for analysis to detect an analyte, wherein the cartridge is configured to separately and releasably interface with a handheld sample collection unit and a base station, the cartridge comprising:
one or more sample ports;
a first reaction channel, a second reaction channel, and a third reaction channel, the one or more sample ports fluidically connectable within the cartridge with the first reaction channel, wherein the cartridge is configured to facilitate analysis within the second reaction channel of a first control for the analyte contained within the cartridge and analysis within the third reaction channel of a second control for the analyte; and
one or more pumping assist ports, the one or more pumping assist ports fluidically connected to the first reaction channel within the cartridge by one or more flow paths and positioned so as to interface, when the cartridge is interfaced with the handheld sample collection unit, with a suction port on the handheld sample collection unit that is fluidically connected with a suction pump within the handheld sample collection unit.

The cartridge of claim 1 , further comprising one or more optical measurement sites, the optical measurement sites positioned within the cartridge so as to align with an optical measurement module of the base station when the cartridge is interfaced with the base station, wherein at least a portion of the cartridge that is between each optical measurement site and the optical measurement module when the cartridge is interfaced with the base station is transparent.
3. The cartridge of claim 1 , wherein the first reaction channel having a hydraulic diameter (d) between 0.1 mm and 1 mm.
4. The cartridge of claim 3 , wherein the first reaction channel has a length that is between 15 μL/d 2 and 45 μL/d 2 .
5. The cartridge of claim 1 , wherein the first reaction channel has a volume of between 15 μL and 100 μL.
6. The cartridge of claim 1 , wherein the first reaction channel has a hydraulic diameter less than 1 mm.
7. The cartridge of claim 1 , further comprising:
a first blister containing a predetermined amount of a first substrate;
a second blister containing a predetermined amount of a second substrate;
a third blister containing a predetermined amount of a first buffer;
a fourth blister containing a predetermined amount of a wash fluid; and
a reservoir containing a predetermined quantity of lyophilized antibody that is selected to bind with the analyte.
8. The cartridge of claim 7 , wherein the analyte is tetrahydrocannabinol (THC) and the lyophilized antibody is a horseradish peroxidase (HRP) conjugated monoclonal antibody that is specific to cannabinol.
9. The cartridge of claim 2 , wherein one of the one or more optical measurement sites is part of the first reaction channel.
10. The cartridge of claim 1 , further comprising a memory device, the memory device configured to store data pertaining to a breath sample collected using the cartridge in an encrypted format.
11. The cartridge of claim 7 , wherein the first blister contains a substance selected from the group consisting of TMB (3,3′,5,5′-tetramethylbenzidine), OPD (o-phenylenediamine), and ABTS (2,2′-Azinobis [3-ethylbenzothiazoline-6-sulfonic acid]).
12. The cartridge of claim 1 , further comprising:
a plurality of fluid flow channels within the cartridge and arranged to transport fluid between different locations within the cartridge; and
a plurality of valve membranes, wherein:
each valve membrane is positioned along one of the fluid flow channels, and each of the valve membranes is arranged to allow for selective enablement or disablement of a different portion of the fluid flow channels responsive to an actuation input.
13. The cartridge of claim 7 , further comprising:
a first reservoir configured to receive the first substrate when the first substrate is expelled from the first blister;
a second reservoir configured to receive the second substrate when the second substrate is expelled from the second blister;
a third reservoir configured to receive the first buffer when the first buffer is expelled from the third blister; and
a fourth reservoir configured to receive the wash fluid when the wash fluid is expelled from the fourth blister.
14. The cartridge of claim 1 , wherein the cartridge further comprises an evidence receptacle configured to receive a portion of fluid that is received via the one or more sample ports when fluid is directed through the one or more sample ports.
15. The cartridge of claim 7 , further comprising a substrate mixing reservoir configured to receive the first substrate from the first blister and the second substrate from the second blister, wherein the substrate mixing reservoir is:
a) fluidically interposed between the first reaction channel and the first blister, and
b) fluidically interposed between the first reaction channel and the second blister.

The cartridge of claim 10 , wherein the memory device is configured to store one or more pieces of data selected from the group consisting of: test subject name, test subject age, test subject driver's license number, test subject social security number, test subject date of birth, test subject photograph, test subject fingerprint, test subject signature, test subject voiceprint, ambient temperature, ambient humidity, ambient barometric pressure, identifying information of a person taking a sample, time and date of sample collection, location of sample collection, amount of breath sample passed through the cartridge, a duration of sample collection using the cartridge, measured levels of one or more analytes collected using the cartridge, and combinations of any two or more of the above.

The cartridge of claim 1 , further comprising a blood alcohol concentration (BAC) port that is positioned so as to fluidically connect with a BAC sensor in the handheld sample collection unit when the cartridge is interfaced with the handheld sample collection unit, wherein the BAC port is fluidically connected with the one or more sample ports within the cartridge.
18. The cartridge of claim 1 , wherein the first reaction channel, the second reaction channel, and the third reaction channel are each of the same size and shape.
19. The cartridge of claim 18 , wherein the first reaction channel, the second reaction channel, and the third reaction channel are parallel to each other.
20. The cartridge of claim 1 , wherein the cartridge further includes a breath inlet port configured to interface with a removable mouthpiece, wherein the one or more sample ports are fluidically interposed between the breath inlet port and the first reaction channel.
21. The cartridge of claim 20 , further comprising the removable mouthpiece, wherein the removable mouthpiece is connected with the breath inlet port such that the breath inlet port is fluidically interposed between the removable mouthpiece and the one or more sample ports.
