CONJUGATED ANTIBODIES AGAINST LY75 FOR THE TREATMENT OF CANCER
The invention provides antibodies which bind to LY75. Nucleic acid molecules encoding the antibodies, expression vectors, host cells and methods for expressing the antibodies are also provided. The antibodies may be used for the treatment of cancer, including pancreatic cancer, ovarian cancer, breast cancer, colorectal cancer, esophageal cancer, skin cancer, thyroid cancer, lung cancer, bladder cancer, multiple myeloma and lymphoma.

38 . A method of treating cancer expressing LY75 in a patient in need of such treatment comprising:
administering to said mammal an effective amount of a composition comprising at least one member of a group consisting of an isolated antibody, an antigen-binding portion thereof, and combinations thereof (“said antibody/antigen-binding portion group”), wherein said member of said antibody/antigen-binding portion group binds to LY75, and comprises:
a) a heavy chain variable region comprising:
i) a first vhCDR comprising SEQ ID NO: 5;
ii) a second vhCDR comprising SEQ ID NO: 6; and
iii) a third vhCDR comprising SEQ ID NO: 7; and
b) a light chain variable region comprising:
i) a first vlCDR comprising SEQ ID NO: 8;
ii) a second vlCDR comprising SEQ ID NO: 9; and
iii) a third vlCDR comprising SEQ ID NO: 10.
39 . The method of treating cancer in a mammal in need of such treatment of claim 38 wherein said member of said antibody/antigen-binding portion group induces antibody-dependent cell-mediated cytotoxicity (ADCC) and/or complement dependent cytotoxicity (CDC).
40 . The method of treating cancer in a mammal in need of such treatment of claim 38 wherein said cancer comprises at least one of the group consisting of:
pancreatic cancer, ovarian cancer, breast cancer, colorectal cancer, esophageal cancer, skin cancer, thyroid cancer, lung cancer, kidney cancer, liver cancer, head and neck cancer, bladder cancer, gastric cancer, leukaemia, myeloma, and lymphoma.
41 . The method of treating cancer in a mammal in need of such treatment of claim 40 wherein
said leukaemia comprises at least one of the group consisting of: acute myeloid leukaemia and chronic lymphocytic leukaemia;
said myeloma is a multiple myeloma; and
said lymphoma comprises at least one of the group consisting of: diffuse large B-cell lymphoma (DLBCL), B-Cell Lymphoma, Follicular Lymphoma, Mantle Cell Lymphoma, Lymphoma of Mucosa-Associated Lymphoid Tissue (MALT), T-Cell/Histiocyte-Rich B-Cell Lymphoma, Burkitt's Lymphoma, Lymphoplasmacytic Lymphoma, Small Lymphocytic Lymphoma, Marginal Zone Lymphoma, T Cell Lymphoma, Peripheral T-Cell Lymphoma, Anaplastic Large Cell Lymphoma and AngioImmunoblastic T-Cell Lymphoma.
42 . The method of treating cancer in a mammal in need of such treatment of claim 38 wherein said cancer comprises at least one of the group consisting of:
bladder cancer, pancreatic cancer, triple-negative breast cancer and DLBCL.
43 . The method of treating cancer in a mammal in need of such treatment of claim 38 further comprising
administering to said mammal an effective amount of a composition comprising a drug conjugate covalently bound to at least one member of said antibody/antigen-binding portion group.
44 . The method of treating cancer in a mammal in need of such treatment of claim 43 further comprising a maytansinoid covalently bound to at least one member of said antibody/antigen-binding portion group
45 . The method of treating cancer in a mammal in need of such treatment of claim 44 further comprising a maytansioid covalently bound to at least one member of said antibody/antigen-binding portion group and said mytansinoid is selected from the group consisting of DM1 and DM4.
46 . A composition for administering to a patient in need of a treatment for cancer expressing LY75, said composition comprising:
at least one member of a group consisting of an isolated antibody, an antigen-binding portion thereof, and combinations thereof (“said antibody/antigen-binding portion group”), wherein said member of said antibody/antigen-binding portion group binds to LY75, and comprises:
a) a heavy chain variable region comprising:
i) a first vhCDR comprising SEQ ID NO: 5;
ii) a second vhCDR comprising SEQ ID NO: 6; and
iii) a third vhCDR comprising SEQ ID NO: 7; and
b) a light chain variable region comprising:
i) a first vlCDR comprising SEQ ID NO: 8;
ii) a second vlCDR comprising SEQ ID NO: 9; and
iii) a third vlCDR comprising SEQ ID NO: 10.
47 . The composition for administering to a patient in need of a treatment for cancer of claim 46 further comprising a maytansinoid covalently bound to at least one member of said antibody/antigen-binding portion group.
48 . The composition for administering to a patient in need of a treatment for cancer of claim 47 wherein said maytansinoid is selected from the group consisting of DM1 and DM4.
