Abstract

The present disclosure provides methods, systems, compositions, and kits to address the need for microbiome-related treatment of health conditions and disease. The present disclosure provides for treatment of metabolic conditions using microbial compositions.

Claims (24)

1. A method of producing a microbial composition suitable for human consumption, the method comprising:

(a) culturing Akkermansia muciniphila in a culture medium comprising substantially a plant-based culture medium, thereby generating cultured Akkermansia muciniphila ; and

(b) formulating the cultured Akkermansia muciniphila into a unit dosage form, wherein the unit dosage form comprises:

(i) the cultured Akkermansia muciniphila ; and

(ii) a second microbe comprising a 16S rRNA sequence that comprises at least about 97% sequence identity to a 16S rRNA sequence of Eubacterium hallii,

thereby producing the microbial composition suitable for human consumption.

2. The method of claim 1 , wherein the unit dosage form comprises the cultured Akkermansia muciniphila and the second microbe in a powdered form.

3. The method of claim 1 , wherein the unit dosage form comprises the cultured Akkermansia muciniphila and the second microbe in a lyophilized form.

4. The method of claim 3 , wherein the culturing of the Akkermansia muciniphila is carried out anaerobically.

5. The method of claim 1 , wherein the unit dosage form is a capsule.

6. The method of claim 1 , wherein the unit dosage form is a food composition in a form of a bar.

7. The method of claim 1 , wherein the unit dosage form further comprises the plant-based culture medium.

8. The method of claim 7 , wherein the unit dosage form comprises the plant-based culture medium in a powdered form.

9. The method of claim 1 , wherein the unit dosage form comprises at least 1×10 5 CFUs of the cultured Akkermansia muciniphila.

10. The method of claim 1 , wherein the unit dosage form further comprises one or more of: Clostridium beijerinckii, Clostridium butyricum, Clostridium indolis, Bifidobacterium infantis , or Faecalibacterium prausnitzii.

11. The method of claim 1 , wherein the second microbe is Eubacterium hallii.

12. The method of claim 1 , wherein the unit dosage form comprises at least 1×10 5 CFUs of the second microbe.

13. The method of claim 1 , wherein the unit dosage form does not contain dairy.

14. The method of claim 1 , wherein the unit dosage form further comprises Bifidobacterium infantis.

15. The method of claim 1 , wherein the plant-based culture medium comprises an amino acid source that is a plant-protein hydrolysate.

16. The method of claim 1 , wherein the unit dosage form further comprises a prebiotic.

17. The method of claim 1 , wherein the culture medium further comprises one or more vitamins, one or more salts, a buffer, or a combination thereof.

18. The method of claim 1 , wherein the culture medium consists essentially of the plant-based culture medium.

19. The method of claim 1 , wherein the culture medium consists of the plant-based culture medium, one or more salts, one or more vitamins, and a buffer.
