Abstract

Isoquinoline compounds for treating various diseases and pathologies are disclosed. More particularly, the present disclosure concerns the use of an isoquinoline compound or analogs thereof, in the treatment of disorders characterized by the activation of Wnt pathway signaling (e.g., cancer, abnormal cellular proliferation, angiogenesis, Alzheimer's disease, lung disease, inflammation, auto-immune diseases and osteoarthritis), the modulation of cellular events mediated by Wnt pathway signaling, as well as neurological conditions/disorders/diseases linked to overexpression of DYRK1A.

Claims (69)

1. A compound, or a pharmaceutically acceptable salt thereof, of Formula I:

wherein:

R 1 , R 2 , R 4 , and R 5 are independently selected from the group consisting of H, F, and Cl;

R 3 is selected from the group consisting of phenyl optionally substituted with 1-3 R 7 and a 6-10-membered heteroaryl optionally substituted with 1-3 R 8 ;

R 6 is selected from the group consisting of —(C 1-4 alkylene) p (5-10-membered)heteroaryl optionally substituted with 1-3 R 9 , —(C 1-4 alkylene) p phenyl optionally substituted with 1-3 R 25 , —(C 1-4 alkylene) p (3-10-membered)heterocyclyl optionally substituted with 1-3 R 10 , —(C 1-4 alkylene) p (3-7-membered)carbocyclyl optionally substituted with 1-3 R 11 , —(C 1-4 alkylene)N(R 12 )(R 13 ), —N(R 14 )(R 15 ), —CF(C 1-9 alkyl) 2 , —(C 1-4 alkylene) p O(C 1-9 alkyl), and —(C 2-9 alkynyl) optionally substituted with one or more F and/or Cl; wherein each alkyl of —CF(C 1-9 alkyl) 2 is, independently, optionally substituted with one or more F and/or Cl; wherein each —(C 1-4 alkylene) is, independently, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

R 7 is selected from the group consisting of F, Cl, and —N(R 16 ) 2 ;

R 8 is selected from the group consisting of F, Cl, unsubstituted —(C 1-9 alkyl), unsubstituted —(C 2-9 alkenyl), unsubstituted —(C 2-9 alkynyl), unsubstituted —(C 1-9 chloroalkyl), unsubstituted —(C 1-9 fluoroalkyl), —CN, —N(R 17 )(R 18 ), —(C 1-4 alkylene) p X 1 R 19 , —C(═O)N(R 20 ) 2 , and —(C 1-4 alkylene) p (3-10-membered)heterocyclyl optionally substituted with 1-3 R 21 ; wherein each —(C 1-4 alkylene) is, independently, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

each R 9 is independently selected from the group consisting of F, Cl, unsubstituted —(C 1-9 alkyl), unsubstituted —(C 2-9 alkenyl), unsubstituted —(C 2-9 alkynyl), unsubstituted —(C 1-9 chloroalkyl), unsubstituted —(C 1-9 fluoroalkyl), —X 2 R 22 , —C(═O)N(R 23 ) 2 , —(C 1-4 alkylene) p N(R 24 ) 2 , —(C 1-4 alkylene) p (3-10-membered)heterocyclyl optionally substituted with 1-3 R 25 , and —(C 1-4 alkylene) p (3-7-membered)carbocyclyl optionally substituted with 1-3 R 26 ; wherein each —(C 1-4 alkylene) is, independently, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

each R 10 is independently selected from the group consisting of F, Cl, unsubstituted —(C 1-9 alkyl), unsubstituted —(C 2-9 alkenyl), unsubstituted —(C 2-9 alkynyl), unsubstituted —(C 1-9 chloroalkyl), unsubstituted —(C 1-9 fluoroalkyl), —(C 1-4 alkylene) p OR 27 , —(C 1-4 alkylene) p (3-10-membered)heterocyclyl optionally substituted with 1-3 R 28 , —(C 1-4 alkylene) p (3-7-membered)carbocyclyl optionally substituted with 1-3 R 29 , —C(═O)(R 30 ), —(C 1-4 alkylene) p C(═O)OR 30 , —N(R 20 ) 2 , —(C 1-4 alkylene)phenyl optionally substituted with one or more F and/or Cl, —(C 1-4 alkylene) p (5-10-membered)heteroaryl optionally substituted with one or more F and/or Cl, and —SO 2 (R 31 ); wherein each —(C 1-4 alkylene) is, independently, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

alternatively, if two R 10 are attached to the same carbon atom can together represent ═O to form a carbonyl group;

each R 11 is independently selected from the group consisting of F, Cl, unsubstituted —(C 1-9 alkyl), unsubstituted —(C 2-9 alkenyl), unsubstituted —(C 2-9 alkynyl), unsubstituted —(C 1-9 chloroalkyl), unsubstituted —(C 1-9 fluoroalkyl), —(C 1-4 alkylene) p OR 27 , —N(R 20 ) 2 , —C(═O)(R 30 ), —C(═O)OR 30 , —(C 1-4 alkylene) p (3-10-membered)heterocyclyl optionally substituted with 1-3 R 28 , and —(C 1-4 alkylene) p (3-7-membered)carbocyclyl optionally substituted with 1-3 R 29 ; wherein each —(C 1-4 alkylene) is, independently, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

R 12 is attached to the nitrogen and is selected from the group consisting of H, unsubstituted —(C 1-9 alkyl), unsubstituted —(C 2-9 alkenyl), unsubstituted —(C 2-9 alkynyl), unsubstituted —(C 1-9 chloroalkyl), unsubstituted —(C 1-9 fluoroalkyl), —(C 1-4 alkylene) p (3-10-membered)heterocyclyl optionally substituted with 1-3 R 32 , and 3-7-membered -carbocyclyl optionally substituted with 1-3 R 33 ; wherein —(C 1-4 alkylene) is, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

R 13 is attached to the nitrogen and is selected from the group consisting of unsubstituted —(C 1-9 alkyl), unsubstituted —(C 2-9 alkenyl), unsubstituted —(C 2-9 alkynyl), unsubstituted —(C 1-9 chloroalkyl), unsubstituted —(C 1-9 fluoroalkyl), —(C 1-4 alkylene) p (3-10-membered)heterocyclyl optionally substituted with 1-3 R 32 , and 3-7-membered -carbocyclyl optionally substituted with 1-3 R 33 ; wherein —(C 1-4 alkylene) is, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

R 14 is attached to the nitrogen and selected from the group consisting of H, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), and unsubstituted —(C 1-5 fluoroalkyl);

R 15 is attached to the nitrogen and is selected from the group consisting of —(C 1-4 alkylene) p (3-10-membered)heterocyclyl optionally substituted with 1-3 R 32 , and —(C 1-4 alkylene) p (3-7-membered)carbocyclyl optionally substituted with 1-3 R 33 ; wherein each —(C 1-4 alkylene) is, independently, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

R 16 is independently selected from the group consisting of H, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), —(C 1-4 alkylene)phenyl optionally substituted with one or more F and/or Cl or unsubstituted —(C 1-5 alkyl), —(C 1-4 alkylene) p (5-10-membered)heteroaryl optionally substituted with one or more F and/or Cl or unsubstituted —(C 1-5 alkyl), and —(C 1-4 alkylene) p (3-10-membered)heterocyclyl optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-5 alkyl); wherein each —(C 1-4 alkylene) is, independently, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

alternatively, two adjacent R 16 are taken together to form a 3-10-membered -heterocyclyl ring optionally substituted with 1-3 R 32 ;

R 17 is attached to the nitrogen and is selected from the group consisting of H, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), and unsubstituted —(C 1-5 fluoroalkyl);

R 18 is independently selected from the group consisting of H, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), unsubstituted —(C 1-5 haloalkyl), —(C═O)R 34 , and —(C 1-4 alkylene) p (3-10-membered)heterocyclyl optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-5 alkyl); wherein —(C 1-4 alkylene) is, independently, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

each R 19 is independently selected from the group consisting of H, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), and —(C 1-4 alkylene) p (3-10-membered)heterocyclyl optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-5 alkyl); wherein —(C 1-4 alkylene) is optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

each R 20 is independently selected from the group consisting of H, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), and unsubstituted —(C 1-5 fluoroalkyl);

each R 21 is independently selected from the group consisting of F, Cl, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), and —N(R 20 ) 2 , —CN, and —OH;

each R 22 is independently selected from the group consisting of H, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), —(C 1-4 alkylene)N(R 34 ) 2 , —(C 1-4 alkylene)phenyl optionally substituted with 1-5 R 35 , —(C 1-4 alkylene) p (3-10-membered)heterocyclyl optionally substituted with 1-3 R 36 , and —(C 1-4 alkylene) p (3-7-membered)carbocyclyl optionally substituted with 1-3 R 29 ; wherein each —(C 1-4 alkylene) is, independently, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

each R 23 is independently selected from the group consisting of H, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), and unsubstituted —(C 1-5 fluoroalkyl);

each R 24 is independently selected from the group consisting of H, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), and —(C 1-4 alkylene)N(R 34 ) 2 ; wherein —(C 1-4 alkylene) is optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

each R 25 is independently selected from the group consisting of F, Cl, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), —CN, —C(═O)R 37 , —N(R 38 ) 2 , —(C 1-4 alkylene) p OR 27 , —(C 1-4 alkylene) p (3-10-membered)heterocyclyl optionally substituted with 1-3 R 32 , and —(C 1-4 alkylene) p (3-7-membered)carbocyclyl optionally substituted with 1-3 R 26 ; wherein each —(C 1-4 alkylene) is, independently, optionally substituted with one or more F and/or Cl;

each R 26 is independently selected from the group consisting of F, Cl, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), and —CN;

each R 27 is independently selected from the group consisting of H, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), and —(C 1-4 alkylene) p (3-7-membered)carbocyclyl optionally substituted with 1-3 R 29 , and —(C 1-4 alkylene) p (3-10-membered)heterocyclyl optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-5 alkyl); wherein each —(C 1-4 alkylene) is, independently, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

each R 28 is independently selected from the group consisting of F, Cl, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), —CN, and —(C 1-4 alkylene) p (3-7-membered)carbocyclyl optionally substituted with 1-3 R 29 ; wherein each —(C 1-4 alkylene) is, independently, optionally substituted with one or more and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

each R 29 is independently selected from the group consisting of F, Cl, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), and —CN;

R 30 is selected from the group consisting of H, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1 -5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), —(C 1-4 alkylene)phenyl optionally substituted with one or more F and/or Cl or unsubstituted —(C 1-5 alkyl), —(C 1-4 alkylene) p (5-10-membered)heteroaryl optionally substituted with one or more F and/or Cl or unsubstituted —(C 1-5 alkyl), and —(C 1-4 alkylene) p (3-10-membered)heterocyclyl optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-5 alkyl); wherein each —(C 1-4 alkylene) is, independently, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

each R 32 is independently selected from the group consisting of H, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), and —(C 1-4 alkylene) p (3-7-membered)carbocyclyl optionally substituted with 1-3 R 29 ; wherein each —(C 1-4 alkylene) is, independently, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

each R 33 is independently selected from the group consisting of H, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), and —(C 1-4 alkylene) p (3-7-membered)carbocyclyl optionally substituted with 1-3 R 29 ; wherein each —(C 1-4 alkylene) is, independently, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

R 34 is attached to the nitrogen and is selected from the group consisting of H, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), and unsubstituted —(C 1-5 fluoroalkyl);

each R 35 is independently selected from the group consisting of F, Cl, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), and —CN;

each R 36 is independently selected from the group consisting of F, Cl, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 chloroalkyl), unsubstituted —(C 1-5 fluoroalkyl), —CN, —OH, —C(═O)R 37 , —N(R 38 ) 2 , and —(C 1-4 alkylene) p (3-7-membered)carbocyclyl optionally substituted with 1-3 R 29 ; wherein each —(C 1-4 alkylene) is, independently, optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

each R 37 is a 5-10-membered heteroaryl optionally substituted with 1-6 R 39 ;

each R 38 is independently selected from the group consisting of H, unsubstituted —(C 1-5 alkyl), unsubstituted —(C 2-5 alkenyl), unsubstituted —(C 2-5 alkynyl), unsubstituted —(C 1-5 haloalkyl), and —(C 1-4 alkylene)N(R 34 ) 2 ; wherein —(C 1-4 alkylene) is optionally substituted with one or more F and/or Cl or one or more unsubstituted —(C 1-3 alkyl);

each R 39 is a 3-10-membered -heterocyclyl optionally substituted with 1-3 R 29 ;

each X 1 is selected from the group consisting of O and S;

each X 2 is selected from the group consisting of O, S, and NR 34 ;

each p is independently 0 or 1; and

with the proviso that Formula I is not a structure selected from the group consisting of:

2. The compound of claim 1 , wherein R 1 , R 2 , R 4 , and R 5 are H.

3. The compound of claim 2 , wherein R 3 is phenyl optionally substituted with 1-2 R 7 .

4. The compound of claim 2 , wherein R 3 is pyridinyl optionally substituted with 1 R 8 .

5. The compound of claim 2 , wherein R 3 is pyrimidinyl optionally substituted with 1 R 8 .

6. The compound of claim 2 , wherein R 3 is pyridazinyl optionally substituted with 1 R 8 .

7. The compound of claim 2 , wherein R 6 is -heterocyclyl optionally substituted with 1-2 R 10 .

8. The compound of claim 2 , wherein R 6 is a —CH 2 heterocyclyl optionally substituted with 1-2 R 10 .

9. The compound of claim 7 , wherein R 6 is either a piperidinyl or a pyrrolidinyl both optionally substituted with 1-2 R 10 .

10. The compound of claim 9 , wherein R 6 is a piperidinyl substituted with one —N(R 20 ) 2 .

11. The compound of claim 10 , wherein each R 20 is independently selected from the group consisting of H, Me, —(C 1 -4 chloroalkyl), and —(C 1-4 fluoroalkyl).

12. The compound of claim 7 , wherein R 6 is selected from the group consisting of

and q is 1-2.

13. The compound of claim 2 , wherein R 6 is -carbocyclyl substituted with 1-2 R 11 .

14. The compound of claim 13 , wherein R 6 is cyclohexyl substituted with 1 R 11 .

15. The compound of claim 1 , wherein the compound of Formula I is selected from the group consisting of:

or a pharmaceutically acceptable salt thereof.

16. A pharmaceutical composition comprising a therapeutically effective amount of a compound of claim 1 , or a pharmaceutically acceptable salt thereof, and a pharmaceutically acceptable excipient.

A method of treating a disorder or disease in a patient, wherein the disorder or disease is selected from the group consisting of: chronic inflammation, systemic inflammation, diabetes, cancer, pulmonary fibrosis, idiopathic pulmonary fibrosis (IPF), degenerative disc disease, bone/osteoporotic fractures, a bone or cartilage disease, a neurological condition/disorder/disease, osteoarthritis, lung disease, a fibrotic disorder, the method comprising administering to the patient a therapeutically effective amount of a compound of claim 1 , or a pharmaceutically acceptable salt, or a pharmaceutical composition, wherein the patient is a human.

The method of claim 17 , wherein the cancer is selected from the group consisting of: osteoma, hemangioma, granuloma, xanthoma, osteitis deformans, meningioma, meningiosarcoma, and gliomatosis, astrocytoma, medulloblastoma, glioma, ependymoma, germinoma (pinealoma), glioblastoma multiform, oligodendroglioma, oligodendrocytoma, schwannoma, retinoblastoma, and congenital tumors, neurofibroma, meningioma, glioma, and sarcoma.

The method of claim 17 , wherein the disorder or disease is a neurological condition/disorder/disease, wherein the neurological condition/disorder/disease is selected from the group consisting of: Alzheimer's disease, amyotrophic lateral sclerosis (ALS), down syndrome, frontotemporal dementia (FTD) including FTD with Parkinsonism-17 (FTDP-17), behavioural variant frontotemporal dementia (bvFTD), FTD in patients with motor neuron disease (MND) (typically amyotrophic lateral sclerosis, also called FTD-ALS), corticobasal degeneration (CBD) (also called corticobasal ganglionic degeneration), progressive supranuclear palsy, primary progressive aphasia (PPA), Prion Diseases, globular glial tauopathy (GGT), myotonic dystrophy type 1 (DM1) (also called Steinert disease), myotonic dystrophy type 2 (DM2) (also called proximal myotonic myopathy), Guam complex, argyrophilic grain disease, dementia pugilistica, post-encephalitic parkinsonism, lewy body dementia, Parkinson's disease, Pick's disease, and additional diseases with pronounced neurodegeneration such as autism, dementia, epilepsy, Huntington's disease, multiple sclerosis; diseases and disorders associated with acquired brain injury such as chronic traumatic encephalopathy, traumatic brain injury, tumor, stroke, frontotemporal dementias, dementia with lewy bodies, prion diseases, multiple system atrophy, inclusion body myositis, degenerative myopathies, diabetic neuropathy, other metabolic neuropathies, endocrine neuropathies, orthostatic hypotension, and Charcot-Marie-Tooth disease.

20. The method of claim 17 , wherein the disorder or disease is a neurological condition/disorder/disease associated with tau protein, amyloid, alpha-synuclein, Tar DNA-binding Protein of 43KDa (TDP-43), Prion protein PrP or fused in sarcoma (FUS) pathology.

21. The method of claim 17 , wherein the disorder or disease is a fibrotic disorder, wherein the fibrotic disorder is selected from the group consisting of: skin fibrosis; scleroderma; progressive systemic fibrosis; lung fibrosis; muscle fibrosis; kidney fibrosis; glomerulosclerosis; glomerulonephritis; hypertrophic scar formation; uterine fibrosis; renal fibrosis; cirrhosis of the liver, liver fibrosis; adhesions; chronic obstructive pulmonary disease; fibrosis following myocardial infarction; pulmonary fibrosis; fibrosis and scarring associated with diffuse/interstitial lung disease; central nervous system fibrosis; fibrosis associated with proliferative vitreoretinopathy (PVR); restenosis; endometriosis; ischemic disease, and radiation fibrosis.

22. The method of claim 17 , wherein the disorder or disease is systemic inflammation.

23. The method of claim 17 , wherein the disorder or disease is chronic inflammation associated eye disorders, joint pain, arthritis (rheumatoid, osteo, psoriatic gout), cancers (colon, breast, lung, pancreas, and others), gastrointestinal disorders (ulcerative colitis and inflammatory bowel diseases), pulmonary disorders (chronic obstructive pulmonary disorder and asthma), allergies, skin disorders (atopic dermatitis and psoriasis), diabetes, pancreatitis, tendonitis, hepatitis, heart disease, myocarditis, stroke, lupus, and neurological disorders such as multiple sclerosis, Parkinson's and dementia including Alzheimer's disease.

24. A method of preventing or reducing angiogenesis in a patient, the method comprising administering to the patient a therapeutically effective amount of a compound of claim 1 , or a pharmaceutically acceptable salt, or a pharmaceutical composition.
